CELL TRANSDUCTION AND PROCESSING METHODS

Abstract

The invention relates to methods, systems, and kits for cell processing, for example, for therapeutic use, such as adoptive cell therapy. The methods provided include transduction processes in which cells and viruses are incubated under conditions that result in the transduction of cells with a viral vector. In some embodiments, incubation is carried out in an internal cavity of a generally rigid centrifugal chamber, such as a cylindrical chamber made of hard plastic, the cavity of which may have a variable volume. The methods include further processing steps, including those performed in such a chamber, such as washing, selection, isolation, culture, and formulation. In particular, the description relates to a method providing advantages over existing processing methods, such as those available for large-scale processing.Such advantages include, for example, reduced cost, streamlining, increased efficiency, increased safety, and increased reproducibility between different subjects and conditions.
