AI Technical Summary
Current EGFR inhibitors face challenges in maintaining stability, hygroscopicity, and oral absorbability, making it difficult to predict the crystal form and physical properties of compounds used as pharmaceuticals, particularly for treating cancers like non-small cell lung cancer.
Development of specific crystal forms and co-crystals of the 7H-pyrrolo[2,3-d]pyrimidin-4-amine derivative, such as free type II and type III crystals, and their salts with acids like succinic and sorbic acid, which exhibit improved stability, low hygroscopicity, and enhanced oral absorption, as characterized by powder X-ray diffraction and thermogravimetric analysis.
The resulting crystals demonstrate superior reproducibility, solid stability, and oral absorption, making them suitable for pharmaceutical applications and effective as antitumor agents, particularly against EGFR mutations.
Abstract
Description
Crystal of 7H-pyrrolo[2,3-d]pyrimidin-4-amine derivative
[0001] The present invention relates to a compound having an epidermal growth factor receptor (EGFR) inhibitory activity, a crystal of a 7H-pyrrolo[2,3-d]pyrimidin-4-amine derivative, a method for crystallizing the same, and the like.
[0002] EGFR is a receptor tyrosine kinase that exerts physiological functions in normal tissues by binding to its ligand, epidermal growth factor (EGF), and contributes to proliferation and apoptosis inhibition in epithelial tissues (Non-Patent Document 1).
[0003] EGFR is also an oncogene, and amplification of the EGFR gene and high protein expression and mutation are known to occur in various types of cancer, such as head and neck cancer, breast cancer, colon cancer, esophageal cancer, pancreatic cancer, lung cancer, ovarian cancer, kidney cancer, bladder cancer, skin cancer, and brain tumors (Non-Patent Document 2). In particular, the number of deaths from lung cancer reaches approximately 1.4 million per year worldwide, and non-small cell lung cancer accounts for more than 80% of lung cancer cases, so the development of an effective treatment is desired (Non-Patent Document 3).
[0004] Currently, several EGFR inhibitors have been reported as antitumor agents. Patent Document 1 describes a compound represented by the following formula (I) as a compound having excellent EGFR inhibitory activity.
[0005]
[0006] The document discloses a pyrimidine compound represented by the formula (chemical name: N-(4-(4-amino-6-ethynyl-5-(quinolin-3-yl)-7H-pyrrolo[2,3-d]pyrimidin-7-yl)bicyclo[2.2.1]heptan-1-yl)-5-methylpyrazine-2-carboxamide (hereinafter also referred to as "compound (1)")).
[0007] Patent Document 1 describes a method for producing compound (1) by adding DMF, DIPEA, 5-methylpyrazine-2-carboxylic acid, and HATU to a THF solution of 7-(4-aminobicyclo[2.2.1]heptan-1-yl)-6-ethynyl-5-(quinolin-3-yl)-7H-pyrrolo[2.3-d]pyrimidin-4-amine, stirring the mixture at room temperature, and then concentrating and purifying the mixture to obtain the compound. However, Patent Document 1 does not specifically disclose what kind of crystalline form compound (1) or a salt thereof may form.

[0010] When a compound is used as an active ingredient of a pharmaceutical, it is desirable for the compound to have a stable crystalline form in order to maintain stable quality and / or facilitate storage and management. Furthermore, it is preferable for the crystalline form of the compound to have low hygroscopicity or good oral absorbability, etc. However, it is difficult to predict whether a specific compound or its salt will form crystals and which crystalline form will have excellent physical properties such as stability.
[0011] In view of the above circumstances, there is a demand for crystals of a compound having EGFR inhibitory activity or a salt thereof, which are excellent in one or more of the properties of stability, hygroscopicity, and oral absorbability.
[0012] As a result of extensive research, the present inventors have obtained several crystalline forms of the free form of Compound (1). Furthermore, they have obtained crystalline forms of salts or co-crystals of Compound (1) with succinic acid, sorbic acid, citric acid, or phosphoric acid.

[0016] The present invention relates to a crystal of N-(4-(4-amino-6-ethynyl-5-(quinolin-3-yl)-7H-pyrrolo[2,3-d]pyrimidin-7-yl)bicyclo[2.2.1]heptan-1-yl)-5-methylpyrazine-2-carboxamide represented by the following formula (I), and a crystal of the same with an acid (a crystal of a salt thereof or a co-crystal thereof). Specifically, the present invention relates to a free-form type II crystal, a free-form type III crystal, a type III crystal with succinic acid, and a type I crystal with sorbic acid of compound (1). Note that, in this specification, the terms type I, type II, type III, etc. are convenient names for distinguishing between crystalline forms, and the crystals according to the present invention are not limited to these names.
[0017] In this specification, a crystal of N-(4-(4-amino-6-ethynyl-5-(quinolin-3-yl)-7H-pyrrolo[2,3-d]pyrimidin-7-yl)bicyclo[2.2.1]heptan-1-yl)-5-methylpyrazine-2-carboxamide (compound (1)) with an acid refers to a crystal of a salt or a co-crystal with an acid. A salt crystal is a crystal in which compound (1) and an acid molecule are bound by an ionic bond, and a co-crystal is a crystal in which compound (1) and an acid molecule are bound by a non-ionic interaction. In the present invention, a crystal of compound (1) with an acid may be a crystal of a salt or a co-crystal, and includes both meanings. For example, in the case of a type III crystal of N-(4-(4-amino-6-ethynyl-5-(quinolin-3-yl)-7H-pyrrolo[2,3-d]pyrimidin-7-yl)bicyclo[2.2.1]heptan-1-yl)-5-methylpyrazine-2-carboxamide with succinic acid, N-(4-(4-amino-6-ethynyl-5-(quinolin-3-yl)-7H-pyrrolo[2,3-d]pyrimidin-7-yl)bicyclo[2.2.1]heptan-1-yl)-5-methylpyrazine-2-carboxamide is This refers to a Type III crystal of N-(4-(4-amino-6-ethynyl-5-(quinolin-3-yl)-7H-pyrrolo[2,3-d]pyrimidin-7-yl)bicyclo[2.2.1]heptan-1-yl)-5-methylpyrazine-2-carboxamide succinate or a Type III co-crystal of N-(4-(4-amino-6-ethynyl-5-(quinolin-3-yl)-7H-pyrrolo[2,3-d]pyrimidin-7-yl)bicyclo[2.2.1]heptan-1-yl)-5-methylpyrazine-2-carboxamide with succinic acid. In one embodiment of the present invention, there is provided a Type III co-crystal of N-(4-(4-amino-6-ethynyl-5-(quinolin-3-yl)-7H-pyrrolo[2,3-d]pyrimidin-7-yl)bicyclo[2.2.1]heptan-1-yl)-5-methylpyrazine-2-carboxamide with succinic acid and a method for producing the same, or a Type I co-crystal of N-(4-(4-amino-6-ethynyl-5-(quinolin-3-yl)-7H-pyrrolo[2,3-d]pyrimidin-7-yl)bicyclo[2.2.1]heptan-1-yl)-5-methylpyrazine-2-carboxamide with sorbic acid and a method for producing the same.
[0018] A crystal is a solid in which atoms and molecules are arranged in a regular repeating structure, and is different from an amorphous (non-crystalline) solid that does not have a repeating structure. Crystal or amorphous solids can be investigated by methods such as powder X-ray diffraction measurement (XRD measurement), differential scanning calorimetry (DSC measurement), simultaneous thermogravimetry-differential thermal analysis (TG-DTA), and single crystal analysis. Crystal polymorphs are the same molecule but have different atomic or molecular arrangements in the crystal, and it is known that the peaks obtained in XRD measurement differ between crystal polymorphs. It is also known that solubility, oral absorbability, and / or stability differ between crystal polymorphs.
[0019] In this specification, the terms "crystal" and "amorphous" are used in their usual sense, and the fact that a material is crystalline can be confirmed by X-ray diffraction spectroscopy.
[0020] Due to the nature of the data, when determining the identity of a crystal, the diffraction angle and overall pattern of a powder X-ray diffraction pattern are important. The relative intensities of a powder X-ray diffraction pattern may vary somewhat depending on the direction of crystal growth, particle size, measurement conditions, maintenance status of the measurement equipment, and the method of preparing the measurement sample, and therefore should not be interpreted strictly. In this specification, the phrase "a powder X-ray diffraction spectrum substantially identical to the powder X-ray diffraction spectrum shown in the figure" refers to a powder X-ray diffraction spectrum that a person skilled in the art would recognize as identical, taking into account slight variations in peak position and intensity relative to the powder X-ray diffraction spectrum shown in the figure. For example, the numerical values of the diffraction angle (2θ) may have a measurement error of approximately ±0.2°.
[0021] In this specification, the term "compound (1)" simply refers to N-(4-(4-amino-6-ethynyl-5-(quinolin-3-yl)-7H-pyrrolo[2,3-d]pyrimidin-7-yl)bicyclo[2.2.1]heptan-1-yl)-5-methylpyrazine-2-carboxamide, and is used to include both "crystal" and "amorphous." In this specification, the term "crystal of compound (1)" is used to include both a crystal of the free form of compound (1) and a crystal of compound (1) with an acid (a crystal of a salt of compound (1) and a co-crystal of compound (1)). In the present specification, crystals in which the molecules other than compound (1) constituting the crystal (other molecules constituting the salt or co-crystal) are not specified refer to crystals of the free form of N-(4-(4-amino-6-ethynyl-5-(quinolin-3-yl)-7H-pyrrolo[2,3-d]pyrimidin-7-yl)bicyclo[2.2.1]heptan-1-yl)-5-methylpyrazine-2-carboxamide, and are not limited to a single crystalline form such as free form Form I, but may include multiple free form crystals. In the present specification, the term "equivalent" refers to molar equivalents. In a crystal of compound (1) and an acid (a crystal or co-crystal of its salt), the equivalent of the acid relative to compound (1) can be analyzed by, for example, NMR or ion chromatography. In the present specification, the crystal may be either a hydrate or an anhydrous form.
[0022] The present invention also encompasses labeled forms of compound (1) or salts thereof, i.e., compound (1) or salts of compound (1), and compounds in which one or more atoms are substituted with a radioactive isotope or a non-radioactive isotope.
[0023] The crystal may be any crystal of compound (1), and may be a single crystal or a polymorphic mixture containing other crystals of compound (1). Specifically, the purity of the crystal may be 40% by weight or more (i.e., 40% by weight or more of single crystals). The purity of the crystal is preferably 50% by weight or more (i.e., 50% by weight or more of single crystals), more preferably 75% by weight or more (i.e., 75% by weight or more of single crystals), more preferably 90% by weight or more (i.e., 90% by weight or more of single crystals), even more preferably 95% by weight or more (i.e., 95% by weight or more of single crystals), even more preferably 98% by weight or more (i.e., 98% by weight or more of single crystals), and particularly preferably 99% by weight or more (i.e., 99% by weight or more of single crystals). This purity can be measured by analysis such as differential scanning calorimetry (DSC measurement).
[0024] In this specification, chemical purity refers to the purity measured by high performance liquid chromatography (HPLC), and when referring to the chemical purity of compound (1), it refers to the purity measured by HPLC of compound (1). In this case, the wavelength of the detector used for purity measurement can be appropriately set. Specifically, the chemical purity of the crystals of compound (1) is preferably 90% or more, more preferably 95% or more, even more preferably 98% or more, and particularly preferably 99% or more.
[0025] The numerical values obtained from various patterns may have some error depending on the direction of crystal growth, particle size, measurement conditions, etc. Therefore, in this specification, the numerical value of the diffraction angle (2θ) in the powder X-ray diffraction pattern may have a measurement error of about ±0.2°. In other words, when the term "diffraction angle (2θ±0.2°)" is used in this specification, it means that a measurement error of ±0.2° is allowed for the numerical value of the diffraction angle (2θ). For example, when the diffraction angle (2θ±0.2°) is described as "6.6°," this means that a diffraction angle of up to 6.6°±0.2° is allowed, and includes diffraction angles of "6.4° to 6.8°." Furthermore, this value can be calculated using the Bragg formula (2d sinθ=nλ) and varies depending on the wavelength measured. In other words, it is also possible to convert it to a diffraction angle at a different measurement wavelength by substituting the measurement wavelength into the above formula. For example, the diffraction angles (2θ) at a wavelength λ of 1.54 Å, which is the characteristic X-ray of CuKα, are 6.6° and 8.8°, respectively, and if the values of d and n are unchanged, the diffraction angles (2θ) at 0.75 Å are 3.2° and 4.3°, respectively.
[0026] The compound (1) used in the crystallization method of the present invention may be, for example, a compound produced by the method described herein. For crystallization, the compound (1) may be used either directly after synthesis without being extracted as crystals, or after being extracted as crystals (crude crystals).
[0027] In this specification, "room temperature" generally refers to about 18°C to about 25°C.
[0028] In this specification, the term "lower alcohol" refers to an alcohol having 1 to 5 carbon atoms, which may be linear or branched. Examples include methanol, ethanol, propanol, isopropyl alcohol, butanol, and pentanol.
[0029] The endothermic peak in a thermogravimetric differential thermal analysis (TG-DTA) curve usually means ±5.0°C, although the measurement temperature may vary depending on the rate of temperature rise per minute, the chemical purity of the sample, etc. Therefore, when the crystal according to the present invention is measured by TG-DTA, an error in the peak (onset value) of ±5.0°C is taken into account. The term "around" used in this case means ±5.0°C.
[0030] One embodiment of the present invention relates to free-form crystals of Compound (1) (type II and type III crystals), crystals of Compound (1) with succinic acid (type III crystals), and crystals of Compound (1) with sorbic acid (type I crystals). The inventors have found that, among the free-form crystals of Compound (1), free-form crystals II and III crystals are stable and can be obtained with good reproducibility, and that free-form crystals II have advantageous properties in pharmaceutical production, such as low hygroscopicity and good oral absorbability. Furthermore, among crystalline forms or co-crystals with a specific acid, crystals with succinic acid and crystals with sorbic acid have been found to be stable, low hygroscopic, can be obtained with good reproducibility, and have good oral absorbability.
[0031] In the present specification, the stability of a crystal can be evaluated, for example, by a solid state stability test shown in the later-described Test Example 1. In one embodiment of the present invention, the change in chemical purity of a crystal of compound (1) after storage for 4 weeks under the conditions described in Test Example 1 is preferably ±0.15% or less, more preferably ±0.10% or less, and even more preferably ±0.05% or less.
[0032] In this specification, the hygroscopicity of a crystal can be evaluated by a dynamic moisture sorption / desorption (DVS) test shown in the following Test Example 2. When evaluated under the conditions described in Test Example 2, the crystal of compound (1) in one embodiment of the invention preferably exhibits a weight increase rate of less than 5%, more preferably less than 3%, and even more preferably less than 1%.
[0033] In this specification, the oral absorbability of the crystals can be evaluated by a plasma concentration measurement test shown in Test Example 5 below.

Another embodiment of the present invention provides a method for treating a tumor, comprising administering to a subject in need thereof an effective amount of the above-mentioned crystal of compound (1) (i.e., a free form crystal of compound (1) or a crystal of compound (1) with an acid (a salt crystal or a co-crystal)). [0121] Test Example 1: Solid-state stability test The free-form type II crystals, free-form type III crystals, free-form type VII crystals, and free-form type VIII crystals of compound (1) obtained in the Examples, Reference Examples, and Comparative Examples were stored for 4 weeks and their solid-state stability was evaluated by the following procedure. Storage conditions: 80°C (closed system) (HIFLEX (high-temperature storage device manufactured by ETAC)) Storage period: 4 weeks Storage amount: about 30 mg Storage container: glass bottle The results are shown in Table 3 below.
[0122] The change in the amount of related substances (the amount of substances detected other than compound (1)) was analyzed by HPLC using the following method. For the analytical sample, approximately 8 mg of each sample was weighed and dissolved in 1 mL of a chloroform-methanol mixture (1:1 v / v). 50 μL of this solution was measured and diluted with 950 μL of an acetonitrile-water mixture (7:3). 5 μL of this solution was accurately measured and used. HPLC Measurement Method (Stability Test) The amount of related substances in the sample solution was measured by HPLC analysis. The handling of the equipment, including data processing, followed the methods and procedures specified for each equipment. Column: InertSustainSwift C18 (2.1 x 50 mm, 1.9 µm) manufactured by GL Sciences UV detection: 220 nm Column temperature: 40°C Flow rate: 0.3 mL / min Sample cooler: 10°C Sample concentration: 0.1 mg / mL Mobile phase A: 10 mmol / L phosphate buffer (pH 6.8) / acetonitrile (95:5) mixture Mobile phase B: acetonitrile The gradient is shown in Table 4.
[0123] As a result, it was found that the free form II crystal, free form III crystal and free form VII crystal of Compound (1) showed almost no increase in related substances and were extremely stable crystals.
[0124] Test Example 2 Dynamic moisture adsorption / desorption (DVS) test A moisture adsorption / desorption test was carried out using the free form II crystal, free form III crystal, free form VII crystal, and free form VIII crystal of compound (1) obtained in the Examples, Reference Examples, and Comparative Examples. The moisture adsorption / desorption test was carried out under the following conditions. Approximately 10 mg of sample was filled into a dedicated quartz holder, and the weight of the sample at each humidity was continuously measured and recorded under the following conditions. The operation of the device, including data processing, was carried out in accordance with the method and procedure specified for each device. Apparatus: VTI SA+ (manufactured by TA Instruments) Drying temperature: 60°C Heating rate: 1°C / min Drying equilibrium: Confirm that there is no decrease of 0.01 wt% in 5 minutes, within a range not exceeding 300 minutes Measurement temperature: 25°C Humidification equilibrium: Confirm that there is no increase of 0.01 wt% in 5 minutes, within a range not exceeding 120 minutes Relative humidity program: Increase in 5% RH increments from 5 to 95% RH, and decrease in 5% RH increments from 95% RH to 5% RH The weight changes within the measurement condition ranges obtained in these tests are shown in Figures 9 to 12.
[0125] As shown in the figure, the free-form type II crystal of compound (1) showed a weight increase of 0.16% at 95% relative humidity in a moisture adsorption / desorption test, which is less than 1%, and was therefore determined to be almost non-hygroscopic. The free-form type III crystal of compound (1) exhibited rapid hygroscopicity when the RH exceeded 65%, but moisture desorption occurred when the humidity was reduced, indicating that this adsorption was reversible. On the other hand, as shown in the figure, the free-form type VII crystal and the free-form type VIII crystal showed a mass increase of 1% or more at 95% relative humidity in a moisture adsorption / desorption test, demonstrating their high moisture absorption. Furthermore, these crystals did not desorb moisture even when the humidity was reduced, indicating that the reaction was irreversible.
[0126] From the above results, it can be said that the free form II crystal of compound (1) has lower hygroscopicity than the free form III crystal, free form VII crystal, and free form VIII crystal of compound (1), and is superior as a candidate compound for pharmaceutical development from the viewpoint of industrial production of pharmaceuticals with stable quality. Since the free form III crystal has low hygroscopicity when the RH is less than 65%, it was considered to be useful as a candidate compound for pharmaceuticals, similar to the free form II crystal, in a humidity-controlled environment. It was confirmed that the free form II crystal and the free form III crystal of compound (1) have excellent properties as pharmaceuticals or pharmaceutical ingredients.
[0127] Example 3 Preparation of Type III Crystal of N-(4-(4-amino-6-ethynyl-5-(quinolin-3-yl)-7H-pyrrolo[2,3-d]pyrimidin-7-yl)bicyclo[2.2.1]heptan-1-yl)-5-methylpyrazine-2-carboxamide (Compound (1)) with Succinic Acid Succinic acid (6.88 g) and acetonitrile (90 mL) were added to Type III crystal (10 g) of free form of Compound (1), and the suspension was stirred at 50°C for about 1 hour, and then filtered to recover the solid. This was suspended and washed twice in ethanol, and dried under reduced pressure at 40°C for about 19.5 hours to obtain 11.71 g of the title crystal.
[0128] The powder X-ray diffraction spectrum (Method A) of the resulting crystals was obtained by the above procedure and is shown in Figure 13. Characteristic diffraction angles observed in the powder X-ray diffraction spectrum include the following peaks: Characteristic diffraction angles (2θ±0.2°): 10.4°, 13.3°, 14.6°, 15.7°, 17.2°, 18.3°, 19.7°, 20.8°, 22.3°, and 27.5°. Other peaks are as shown in Table 5 below. The differential thermal-thermogravimetric simultaneous measurement curve of the obtained crystals was obtained by the above procedure and is shown in Figure 14. [0137] Test Example 3: Solid State Stability Test The solid state stability of the Type III crystals with succinic acid, Type I crystals with citric acid, Type IV crystals with citric acid, Type I crystals with sorbic acid, and Type I crystals with phosphoric acid obtained in the Examples, Reference Examples, and Comparative Examples was evaluated after storage for 4 weeks. The evaluation method, storage, and measurement conditions were the same as in Test Example 1. The results are shown in Table 7 below.
[0138] As a result, it was found that the type III crystal of compound (1) with succinic acid and the type I crystal of compound (1) with sorbic acid showed almost no increase in related substances and were extremely stable crystals. On the other hand, the type I crystal of compound (1) with citric acid, the type IV crystal of compound (1) with citric acid, and the type I crystal of compound (1) with phosphoric acid showed a decrease in purity and were found to be unstable crystals.
[0139] Test Example 4 Dynamic Moisture Sorption / Desorption (DVS) Test A moisture sorption / desorption test was carried out using type III crystals with succinic acid, type I crystals with citric acid, type IV crystals with citric acid, type I crystals with sorbic acid, and type I crystals with phosphoric acid of Compound (1) obtained in the Examples, Reference Examples, and Comparative Examples. The moisture sorption / desorption test was carried out under the same conditions as in Test Example 2. The weight changes obtained in this test within the range of measurement conditions are shown in Figures 23 to 27.
[0140] As shown in the figure, the Type III crystal of compound (1) with succinic acid and the Type I crystal of compound (1) with sorbic acid showed weight increases of 0.28% and 0.26%, respectively, at 95% relative humidity in a moisture adsorption / desorption test, which is less than 1%, and therefore they were determined to have almost no hygroscopicity. On the other hand, the Type I crystal of compound (1) with citric acid, the Type IV crystal of compound (1) with citric acid, and the Type I crystal of compound (1) with phosphoric acid showed mass increases of 1% or more at 95% relative humidity in a moisture adsorption / desorption test, as shown in the figure, demonstrating that they have high hygroscopicity.
[0141] From the above results, it can be said that the Type III crystals of compound (1) with succinic acid and the Type I crystals of compound (1) with sorbic acid have lower hygroscopicity than the Type I crystals of compound (1) with citric acid, the Type IV crystals of compound (1) with citric acid, and the Type I crystals of compound (1) with phosphoric acid, and are superior as candidate compounds for pharmaceutical development from the viewpoint of industrial production of pharmaceuticals with stable quality. It was confirmed that the Type III crystals of compound (1) with succinic acid and the Type I crystals of compound (1) with sorbic acid have excellent properties as pharmaceuticals or pharmaceutical active ingredients.
[0142] Test Example 5 Plasma Concentration Measurement Test For the free form (type II crystal) of compound (1) obtained in the Examples, type III crystal of compound (1) with 1 equivalent of succinic acid, and type I crystal of compound (1) with 1 equivalent of sorbic acid, a plasma concentration measurement test was carried out after oral administration to male beagle dogs set under low gastric conditions.
[0143] (Selection of Beagles) To select three male beagles for use in the study, plasma concentrations were measured in five male beagles maintained under low gastric conditions after solution administration and capsule administration. Three dogs with large differences in AUClast (area under the plasma concentration-time curve from 0 hours after administration to the time point at which the final concentration could be calculated, calculated using the linear trapezoidal method before Tmax and the logarithmic trapezoidal method after Tmax) and Cmax (maximum plasma concentration) after solution administration and capsule administration were selected, and plasma concentrations were measured after oral administration of the free form (type II crystal) of compound (1), the type III crystal of compound (1) with one equivalent of succinic acid, and the type I crystal of compound (1) with one equivalent of sorbic acid, all packed into capsules. Preparation of the administration solution, capsule filling, and animal experiments were performed according to the following procedures.
[0144] (Preparation of Dosage Solution, Capsule Filling, and Animal Experiment Method) The free form of Compound (1) was dissolved in 20% HP-β-CD solution containing 0.1 N hydrochloric acid to prepare a 20 mg / 5 mL solution. The free form of Compound (1) (type II crystal), type III crystal with 1 equivalent of succinic acid, and type I crystal with 1 equivalent of sorbic acid were each filled into gelatin capsules so that the molecular weight equivalent of Compound (1) (free form) was 20 mg / kg. The male beagle dogs used in the test were fasted from 11:00 AM to 12:00 PM on the day before administration of Compound (1), and were fed after blood sampling 8 hours after compound administration. Water was withheld from 30 minutes before compound administration until 2 hours after administration, with free access at other times. Thirty minutes before compound administration, atropine (atropine sulfate injection 0.5 mg "Fuso") was administered intravenously at 0.02 mg / kg, followed by intramuscular administration of pentagastrin at 0.01 mg / kg. Thirty minutes after atropine administration, a capsule filled with the compound was administered orally by force, and the animal was allowed to self-swallow the capsule with 5 mL of distilled water. 50 mL of distilled water was then administered orally by force using a catheter. Alternatively, a solution containing the compound was orally administered at a dose of 20 mg / 5 mL / kg using a catheter, and the catheter was immediately washed with 10 mL of distilled water. After blood samples were taken 15 minutes and 1 hour after compound administration, pentagastrin was again administered intramuscularly at 0.01 mg / kg. Before and after administration (0.25, 0.5, 1, 2, 4, 6, 8, and 24 hours later), the blood collector grasped the dog's forelimbs and engorged the blood vessels with a clothespin or similar tool. The blood collection site was then disinfected with alcohol swabs, and approximately 0.2 mL of blood was collected using a heparinized syringe and needle. After blood collection, the puncture site was held down with absorbent cotton to ensure adequate hemostasis. The collected blood was cooled on ice, and plasma was separated by centrifugation as quickly as possible. The separated plasma was promptly frozen on dry ice and then frozen and stored in a freezer set at -80°C. Using LC / MS / MS, the concentration of compound (1) in each plasma sample was measured by MRM and quantified from the calibration curve. From this, AUClast, Cmax, and Tmax (time to reach maximum blood concentration) were calculated using Phoenix WinNonlin (v7.0.0, Certara USA, Inc.).
[0145] The results are shown in Table 8. It was assumed that the free-form type II crystals of compound (1), the type III crystals with succinic acid, and the type I crystals with sorbic acid are crystalline forms that exert pharmacological effects in vivo. From the above, it was confirmed that all crystalline forms are orally administrable.
