Description

[0001] This application is a divisional application of application filed on March 3, 2022, with application number 202280017434.5 and entitled "Combination Pharmaceutical Composition of CDK4 / 6 Inhibitor and Its Use Thereof".

[0002] Cross-reference to related applications

[0003] This disclosure claims the benefit and priority of Chinese Patent Application No. 202110236331.5, filed with the State Intellectual Property Office of the People's Republic of China on March 3, 2021, the entire contents of which are hereby incorporated herein by reference. Technical Field

[0004] This disclosure pertains to the field of pharmaceutical technology and relates to a combination pharmaceutical composition of CDK4 / 6 inhibitors and its use in the treatment of breast cancer. Background Technology

[0005] Cyclin-dependent kinases (CDKs) 4 / 6 are key regulators of the cell cycle, triggering the transition from the growth phase (G1 phase) to the DNA replication phase (S1 phase). During cell proliferation, the complex formed by CDKs 4 / 6 and cyclin D phosphorylates retinoblastoma protein (Rb). Once phosphorylated, the tumor suppressor protein Rb releases its tightly bound transcription factor E2F in its unphosphorylated state. E2F activates further transcription, propelling the cell cycle through the restriction point (R point) and progressing from G1 to S phase. Therefore, inhibiting CDKs 4 / 6 to prevent the formation of the Cyclin D-CDKs 4 / 6 complex can halt the cell cycle progression from G1 to S phase, thereby inhibiting tumor proliferation. WO2016141881 discloses a 2-hydropyrazole derivative as a selective CDKs 4 / 6 inhibitor, specifically disclosing a compound of formula (I) with the following structure.

[0006]

[0007] Fulvestrant, chemically known as 7-α-[9-(4,4,5,5,5-pentafluoropentanesulfinyl)nonyl]estrogens-1,3,5-(10)-trien-3,17-β-diol, has a relative molecular mass of 606.77. It is a white or off-white powder, insoluble in water. Fulvestrant is a novel estrogen receptor antagonist—an estrogen receptor downregulator—that acts as an anti-breast cancer drug. It binds to estrogen receptors at the cellular level, blocking and degrading estrogen receptors, thereby inhibiting the growth of tumor cells under the influence of estrogen.

[0008] According to the World Cancer Report, in 2012, there were approximately 1.7 million new cases of breast cancer globally, and about 500,000 deaths, accounting for 25% of all new cancer cases in women and 15% of all cancer deaths, ranking first in both categories. Of the new breast cancer cases each year, 3%-10% of women already have distant metastases at the time of diagnosis. Among early-stage patients, 30%-40% may progress to advanced breast cancer, with a 5-year survival rate of approximately 20%.

[0009] Advanced breast cancer (ABC) presents unique challenges in terms of treatment options and efficacy, and currently lacks a universally accepted standard treatment regimen. The overall median survival for advanced breast cancer is 2-3 years, varying depending on the molecular subtype. For HER2-positive ABC patients, anti-HER2 therapy can alter the natural course of the disease and significantly prolong survival; however, for triple-negative ABC patients, the overall prognosis has not yet shown significant improvement. Hormone receptor-positive (HR+) breast cancer accounts for approximately 65%-75% of breast cancers. Endocrine therapy, with its comparable efficacy to chemotherapy and fewer toxic side effects, has become the preferred treatment for hormone receptor-positive (HR+) metastatic breast cancer.

[0010] Although endocrine therapy is the main treatment option for hormone receptor-positive breast cancer, about 30% of hormone receptor-positive breast cancers are resistant to endocrine therapy, and almost all patients develop secondary resistance during subsequent treatment. Therefore, overcoming endocrine therapy resistance has become an urgent problem to be solved in the field of breast cancer treatment. Summary of the Invention
