[0003] The present application relates to the field of biopharmaceutical technology, and in particular to an antibody drug conjugate.BACKGROUND

[0004] Malignant tumors, also known as cancer, have become the "number one killer" threatening the lives and health of people worldwide. The fundamental causes of high cancer mortality rates are the spread and metastasis of cancer cells, as well as the tendency for most patients to relapse and develop drug resistance after treatment. Existing clinical treatments, such as surgery, radiotherapy, and chemotherapy, have limited or only short-term effects on cancer cell metastasis, recurrence, and drug resistance, and cannot improve the long-term survival of patients. At present, surgical resection is effective for about 10-20%of early-stage patients, but is almost ineffective for patients who have spread and metastasized. Radiotherapy can only treat local lesions and is often used as preoperative and postoperative adjuvant treatment and as a radical treatment for a few types of cancer. Chemotherapy can be used for patients who have spread and metastasized, but due to its severe side effects and the tendency to develop short-term or long-term drug resistance, it can only have a significant short-term effect on about 20-30%of patients. Even when comprehensive treatment measures combining surgery, radiotherapy, and chemotherapy are employed, the long-term efficacy in terms of 5-year survival has remained stagnant at around 20-30%for many years. Approximately 70-80%of patients experience metastasis, recurrence, and drug resistance within 5 years after treatment and die within that period. Even among patients with early-stage cancer who show no signs of metastasis at the time of diagnosis, some still develop metastasis and relapse after treatment and eventually die.SUMMARY OF THE INVENTION

[0005] Currently, the identification of tumor-associated antigens and the subsequent delivery of drugs to tumor cells in the form of antibody drug conjugates is an effective method for treating tumors. This drug form combines the targeting ability of monoclonal antibodies to tumor cells with the potent tumor-killing advantages of small molecule cytotoxins. It can significantly reduce the toxic side effects of small molecule toxins and enhance drug efficacy, thereby offering significant therapeutic advantages.

[0006] The applicant found that ITGA2 is a tumor cell-specific, overexpressed tumor-associated antigen, which is up-regulated in most solid tumors, especially in colorectal cancer, cholangiocarcinoma, pancreatic cancer, skin cancer, and other cancers. However, current antibodies that can target ITGA2 alone have poor inhibitory effects on the growth of tumor cells. Therefore, the applicant developed an antibody with a high specific binding ability to tumor cells expressing the ITGA2 protein, which can target ITGA2-positive tumor cells in the form of an antibody drug conjugate to achieve an excellent killing effect on the cells.

[0214] The results, as shown in Figure 8, indicated that the tumor volume treated with A36-LD-38 gradually decreased or even disappeared, while the tumor volume treated with A36-Dxd gradually increased, albeit slightly smaller than that in the PBS group. On days 50, 57, and 64, the mice in the A36-Dxd treatment group were switched to A36-LD-38 treatment (5 mpk) for three times, after which the tumor volume showed a significant decrease. This result demonstrates that A36-LD-38 has good efficacy in the human colon cancer cell line COLO205 CDX model, and is much better than the efficacy of A36-Dxd. A further observation was that when the tumors in the A36-Dxd treatment group grew to 1000 mm3 and became drug-resistant, three more doses of A36-LD-38 could reverse the growth trend of the tumors and shrink them, indicating that the A36-LD-38 molecule could overcome the resistance of Dxd in the COLO205 colorectal cancer model. Similarly, different doses of A36mut-LD-38 also inhibited the tumor growth of COLO205 (Figure 9) , indicating that both the A36 antibody with wild-type IgG1 (A36) and the A36 antibody with mutant IgG1 (A36mut) could be conjugated with LD-38 to form ADCs, and they have great efficacies in the human colon cancer cell line COLO205 CDX model.
