IMMUNOCONJUGATES AND PHARMACEUTICAL USES THEREOFFIELD OF THE INVENTION

[0001] The present invention relates to an immunoconjugate of with improved ability to target cancers, which contain a bispecific antibody targeting EGFR and c-MET, and a therapeutic motiety belong to the camptothecin group of drugs. The present invention further relates to use of the immunoconjugates in the manufacture of a drugs for treating cancers.BACKGROUND

[0002] Epidermal growth factor receptor (EGFR) and hepatocyte growth factor receptor (HGFR or c-MET) have been found to be overexpressed in many tumors. For example, EGFR is overexpressed in non-small cell lung cancers (NSCLC) , head and neck squamous cell cancers. c-MET receptor is overexpressed in NSCLC and several other solid tumor types. Recently, EGFR and c-MET have been found to be co-expressed in many solid tumors, including head and neck squamous cell carcinoma, NSCLC, colorectal carcinoma, gastric cancers, renal cell carcinoma, and pancreatic ductal adenocarcinoma, etc.

[0003] For patients with aberrant activation of EGFR, tyrosine kinase inhibitors (TKIs) targeting EGFR are the standard of care. However, many patients will acquire resistance to TKIs. Among the mechanisms, increased c-MET pathway activation is frequently found in patients with EGFR TKI resistance. Moreover, treatment strategies targeting both receptors using a combination of single-agent EGFR and MET inhibitors do not cover the wide range of resistance mechanisms. Therefore, novel approaches that simultaneously engage both EGFR and c-MET through a bispecific antibody, is a proved to be an efficacious approach to overcome resistance and achieve greater efficacy in a sub-group of NSCLC patients.

[0004] One way of improving the therapeutic effect of antibody is antibody-drug conjugates (ADCs) . ADC belongs to a type of new anticancer drug, that antibody is coupled with cytotoxin through chemical coupling. The antibody-drug coupling technology connects the small molecule drug and biological protein together, which may have the advantages of both remarkably increasing the potency of drugs and reducing the side effects.

[0005] Given the prevalent challenges of general off-target toxicity and clinical resistance faced by traditional ADCs, the following criteria should guide target selection: (1) The conventional guiding principle involves identifying antigens characterized by good internalization, relatively low expression on normal tissues, and high expression on tumors. (2) Acknowledging the heterogeneity of solid tumors, it becomes imperative to ascertain the expression levels of targets in various tumor subtypes and sites, facilitating the optimal implementation of tailored drug delivery rather than relying on a singular “magic bullet” approach. (3) Owing to the unique dual targeting characteristics of BsADCs, a comprehensive consideration of the deep-level effects of antigen combinations is essential. This encompasses factors include internalization, recycling, rates of turnover, lysosomal degradation, and obligate mechanisms. The thoughtful integration of these considerations is indispensable for effective BsADC design, ensuring a nuanced understanding of the dynamic interplay between antigens and their cellular processes (Acta Pharm Sin B. 2024 May; 14 (5) : 1965-1986) .

[0006] The BsADC is still in the early stages of development, and early clinical trials encounter substantial challenges. The meticulous choice of BsAb, linker and cytotoxic payload is crucial for attaining the desired therapeutic outcomes in ADCs. Take BsAb for instance, whether it has good internalization or not and whether it can be developed into BsADC or not are unexpected and face many uncertainties, and the linker and cytotoxic payload also make significant difference on the therapeutic outcomes in ADCs.

[0007] Since EGFR and c-Met receptors are highly co-expressed and overexpressed in many types of tumor cells, an ADC target both EGFR and c-MET simultaneously (an EGFRxc-MET bispecific ADC) may possess the advantages in improved therapeutic efficacy benefiting a broader range of patient population, regardless of whether the aberrant EGFR and c-MET activation are the driven factor of tumorigenesis. There is an urgent need to develop bispecific antibody-drug conjugates (bispecific ADCs) targeting EGFR×c-MET that possess good internalization and strong antitumor effects.SUMMARY OF THE INVENTION

[0008] The technical problem to be solved by the present invention is to develop an immunoconjugate, which contain a bispecific antibody targeting EGFR and c-Met, and a therapeutic drug, showing good internalization and very strong antitumor effects in cancers. The antitumor effects may be contributed by improved binding affinities of the antibodies, increased internalizations for robust endocytosis, higher DARs.

[0181] The binding of EGFRxc-MET bispecific antibody to the cell surface EGFR was determined by FACS analysis using EGFR expressing and c-Met non-expressing cancer cells SNU-1. The binding of EGFRxc-MET bispecific antibody to the cell surface c-Met was determined by FACS analysis using c-Met expressing and EGFR non-expressing cancer cells SW620. SNU-1 and SW620 cells were maintained in RPMI-1640 medium supplemented with 10%FBS, 1%penicillin and streptomycin. All cell lines were cultured at 37℃ with 5%CO2 in humidified atmosphere.

[0206] Inhibitory effect of the ADCs on tumor cells growth were examined using in vitro cell killing assay. Different tumor cells were collected in log phase growth and distributed into 96-well plates at 2000 cells / well. After overnight incubation, the ADCs were added to each well. The final concentrations of the ADCs in the wells were ranging from 0.003 nM to 200 nM. After 4 to 7 days of incubation, the cell viability in each well was determined by Cell Titer Glo 2.0 Assay (Promega) . IC50 values were generated in GraphPad Prism using a sigmoidal dose-response non-linear regression fit. The data from these experiments are summarized in Table 5. Figure 3 showed representative killing curves for all ADCs tested against the indicated cell lines. The tested BsAb-1 ADCs showed good in vitro toxicity on tumor cells.
