[0249] The toxicology and toxicokinetic (TK) profiles of the API were characterized in cynomolgus monkeys in a 28-day repeat SC dose study. The 150 mg / mL API solution for SC injection was formulated in 20 mM glutamic / histidine, 2.8% (w / v) proline, 0.02% (w / v) polysorbate 80, at pH 5.3, and supplied in 2R Type I glass vials with a 13 mm rubber stopper integrated into a plastic cap for clinical trial distribution. The formulation was a sterile, colorless to slightly yellow, slightly opalescent solution, essentially free of particles. The labelled volume was 1 mL per vial and there was an overfill of 0.22 mL per vial.

[0250] A GLP-compliant 28-day repeat dose toxicology and TK study was performed in cynomolgus monkeys to determine the toxicity and TK profile of the API by SC injection. Three doses (0, 38, and 240 mg / kg) were tested over a period of 28 days (atotal of two doses on Days 1 and 22) in cynomolgus monkeys (3 / sex / group) . The local tolerance was also determined.
