Abstract
The present utility model relates to a method for determining monolaurin using highperformance liquid chromatography with photodiode array detection (HPLC-PDA). The method comprises the following steps: (1) preparing a mobile phase consisting of methanol, acetonitrile, and an acetate buffer; (2) preparing monolaurin standard solutions by serial dilution; (3) filtering the standards and sample solutions; (4) subjecting the filtered solutions to HPLC equipped with a PDA detector; (5) detecting monolaurin; and (6) quantifying monolaurin using a calibration curve generated from the peak areas of the standard solutions. The use of an acetate buffer at pH 4.0 minimizes hydroxyl ionization and improves chromatographic peak symmetry. The method achieves high linearity (r² = 0.9999), precision (%RSD < 1.00%), accuracy (102.04% recovery), and sensitivity (LOD = 0.429 mg / mL; LOQ = 1.29 mg / mL).
