Abstract
The utility model relates to a process for propagating yellow bamboo (Bambusa vulgaris var. striata) using Cardava (Musa acuminata × Musa balbisiana) leaf extract and indole-3-butyric acid (IBA). The process comprises preparing an aqueous Cardava banana leaf extract at 8–12% (v / v) by macerating the leaves in distilled water at a ratio of 1:9 (w / v) for 20–30 minutes, sequentially filtering the extract through 0.45 μm and 0.22 μm filters, and adjusting the pH to 5.8; supplementing Murashige and Skoog (MS) medium with the leaf extract and 2 mg / L IBA; inoculating surface-sterilized yellow bamboo nodal explants onto the enriched medium under aseptic conditions and incubating at 25 ± 2 °C under a 16-hour light and 8-hour dark photoperiod until roots and shoots develop; and transferring the resulting plantlets to nursery beds under 50 60% shade while maintaining soil pH at 6.0–6.5 and soil nitrogen at ≥0.20%. The process provides a defined procedure for the in vitro propagation and subsequent acclimatization of yellow bamboo.
