Abstract

The present invention relates to a process for the detection of A2142G and A2143G mutations of 23S rRNA of Helicobacter pylori DNA comprising steps of addition of 16.4 µL of water, 2.5 µL of buffer 1(1.5 mM MgC12), 0.2 µL of Taq polymerase (5 U / µL), 0.5 µL of dNTP mixture (10 mM), 0.4 IAL of primer 1 (10 mM), 0.5 µL of primer 2 (10 mM), 0.75 µL of primer 3 (10 mM), 0.75 µL of primer 4 (10 mM), and 3.0 µL of genomic DNA (50 ng / µL) by setting PCR conditions of denaturation at 95°C for 5 min, followed by 35 cycles of denaturation at 95°C for 15 s, annealing at 60.5°C for 20 s, and extension at 68°C for 30 s, with final extension at 68°C for 2 min and using (a) Primer sequence land 3 for the detection of A2142G genotype (b) Primer sequence 2 and 4 for the detection of A21430 genotype and (c) Primer sequence 1 and 4 for the detection of wild type 23S rRNA genotype.
