Method for isolating and concentrating bacteriophages using a three-phase fractionation technique

Abstract

The subject of the application is a method for isolating and concentrating bacteriophages using a three-phase fractionation technique, characterized in that it comprises the following steps: a) saturating a sample with ammonium sulfate at a concentration of 10% to 50% (w / v); b) adding tert-butanol to a sample saturated with ammonium sulfate in proportions of 1:0.5 to 1:2 (v / v); c) separating the phases of the mixture produced in step b) to form an upper phase containing tert-butanol, a middle phase containing precipitated bacteriophage virions, and a lower phase containing ammonium sulfate; d) separating the middle phase from the remaining two phases and purifying the bacteriophage virions; wherein the sample is a bacteriophage lysate or a wastewater sample.
