Abstract
DC60 (02 / 09 / 59) This invention involves modifying the structure of the dengue virus's PrM-E gene to enable its production. Virus-like particles (VLPs) can be increased in quantity by 1) changing signaling peptides. 1) Signal peptide of the prM gene; 2) Changes in the adhesion region of the E gene; 3) Modification of the genetic code. 4) Modification of the fusion loop subgroup of protein E, which, when delivered, is suitable for insect cells. This gene is introduced into insect cells, causing the production of a larger quantity of virus-like particles without damaging the cells. Fused together, and 5) modifications within the prM gene to cause higher or more complete particle aging. The use of virus-like particles to stimulate antibodies to create immunity against the dengue virus, using a single dose. Combined with adjuvant and combined with adjuvant following live attenuated vaccine virus. Revised the invention summary on 14 / 9 / 59. Suitable for insect cells 4) Modification of the fusion loop subgroup of protein E, which, when the gene is delivered... Entering insect cells leads to the production of a greater amount of VLP and prevents cell fusion; and 5) Modifications within the prM gene lead to higher or complete particle maturation and the use of viral-like particles. Antibody stimulation to create immunity against dengue virus using single or combination therapy with Ljuvant. Combined with anjuvant following a live attenuated vaccine dose.
