Abstract

OCR Liposomal complexes containing cholesterol derivatives and plasmic DNA have been shown to inhibit cancer cells. Nanoparticles are formed depending on the ratio of liposomes to plasmid DNA. Liposome derivatives. Cholesterol toxicity depends on the ratio of components within the liposome and the quantity of liposomes. Cholesterol-derived liposomes deliver more plasmid DNA into oral cancer cells than non-liposome use. Cholesterol derivatives, cholesterol derivative liposomal complexes with plasmid DNA, inhibit growth. It causes oral cancer cells to die through apoptosis.

Claims

------08 / 09 / 2568------(OCR) OCR 10KL1. A cholesterol derivative liposome complex with plasmid DNA exhibiting anticancer activity, in which the cholesterol derivative liposome contains a cationic cholesterol derivative lipid, N'-(3-aminopropyl)-N-3'-(carbamoylcholesteryl)propyl-glycineamide, and a supplemental lipid, 1,2-dioleyl-SN-glycero-3-phosphoethanolamine, in molar ratios of 50:25, 50:50, or 50:75, fabricated by ethanol injection. The anticancer-acting plasmid DNA is a plasmid that encodes the tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) and a green fluorescent protein (pTRAIL-EGFP).

2. The process for synthesizing cholesterol-derived liposomal complexes with plasmid DNA exhibiting cancer-inhibiting activity, as claimed in claim 1, which includes the following steps: 2.

1. The fabrication of cholesterol derivative liposomes involves the following steps: a) Dissolve the cationic lipid N'(3-aminopropyl)-N-3'-(carbamoylcholesteryl)propyl-glycineamide in pure ethanol at room temperature. b) Dissolve the supplemental lipid 1,2-dioleyl-SN-glycero-3-phosphoethanolamine in pure ethanol at room temperature. c) The molar ratio of the substances in steps a) and b) is 50:25, 50:50, or 50:

75. d) Mix the solutions in steps a) and b) together. Aspirate the mixture from step d) using a micropipe and inject it into a HEPES buffer solution pH 7.4 (20 mM HEPES pH 7.4). f) Vortex the solution from step e) for 20-30 minutes continuously at room temperature. 2.2 The fabrication of cholesterol derivative liposome complexes with pTRAIL-EGFP plasmid DNA involves the following steps: a) Mix the liposome solution of N'-(3-aminopropyl)-N-3'(carbamoylcholesteryl)propyl-glycineamide obtained from step 2.

1. Mix equal volumes (5, 30, or 50 μL) of pTRAIL-EGFP plasmid DNA solution in purified water or cell culture medium (without fetal bovine serum) at room temperature. Mix thoroughly by gently pipetting up and down 15 times, and incubate at room temperature for 15-20 minutes.
