Universal dendritic cell and preparation method and application thereof

Abstract

This application discloses a universal dendritic cell (DC) and a preparation method and application thereof, belonging to the technical field of immune cells. The universal DC is a universal DC that does not express HLA-I class molecules, obtained by knocking out HLA-I class genes in a starting DC. In its application, according to an HLA-I class molecule subtype of a target antigen-specific T cell to be obtained, HLA-I class molecules that match the target antigen-specific T cells can be re-expressed in the universal DC through gene editing, viral, or mRNA techniques, thereby overcoming the HLA-I class molecule-specific matching restriction that exists when using allogeneic DCs, and solving the problem of insufficient HLA-I class molecule subtype coverage of a single DC.
