Abstract

This disclosure relates to a truncated chromatin open element and combinations thereof, as well as an expression system for eukaryotic cells containing the chromatin open element or combinations thereof, and its applications. The chromatin open element is a truncated sequence of two chromatin open elements from different sources, which can be used alone or in combination. This chromatin open element significantly shortens the effective sequence, thus facilitating the integration of vectors containing the element or combinations thereof with other elements, such as ITR integration. Semi-site-specific integration is achieved in the presence of PiggyBac transposase, followed by a rapid bulkpool screening process, thereby shortening the protein production and cell line construction cycle by approximately 4-5 weeks while maintaining comparable yield.
