Abstract
Description
Technical Field
[0001] This disclosure pertains to the pharmaceutical field and relates to pharmaceutically acceptable salts of pyrrolidine compounds, crystallization of said pharmaceutically acceptable salts, and corresponding preparation methods and uses. Prior Technology
[0002] Estrogen (E2) and estrogen α receptor (ERα) are important drivers of breast cancer development and progression. More than two-thirds of breast cancer patients express ER transcription factors, and ER remains a key driver in most ER-positive patients, even in tumors that have progressed after early endocrine therapy. Therefore, ER is a major target for breast cancer treatment (Pharmacology & Therapeutics 186 (2018) 1–24). Endocrine therapy aims to reduce ER activity, primarily through three classes: selective estrogen receptor modulators (SERMs), such as tamoxifen, which is an allosteric regulator of ER and inhibits its transcriptional activity upon binding; aromatase inhibitors (AIs), which reduce estrogen levels in the body by inhibiting the conversion of androgens to estrogens; and selective estrogen receptor downregulators, such as fulvestrant, which not only acts as an ER antagonist but also induces ER protein degradation. Although endocrine therapy is the first-line treatment for estrogen receptor-positive breast cancer patients, about 30% of patients will relapse after treatment, and almost all patients with metastatic breast cancer will develop drug resistance and progress.
[0003] Clinically, approximately 70-80% of breast cancers test positive for estrogen receptor (ER). The proliferation of these breast cancer cells is heavily dependent on ER, and this subtype accounts for 50% of breast cancer deaths. Early-stage ER-positive breast cancer has a relatively good prognosis, with a 5-year survival rate exceeding 90%. About 30% of patients who undergo postoperative endocrine therapy (TAM or AI drugs) experience recurrence within 10 years, but can still receive standard endocrine therapy.
[0004] Fulvestrant is the first and only clinically approved SERD (Sequencing Agent of Disease) drug for the treatment of postmenopausal patients with ER-positive metastatic breast cancer who have progressed after treatment with tamoxifen or aromatase inhibitors. Multiple studies have shown that ER degradation is not fully achieved in patients treated with fullvestrant. Furthermore, the intramuscular injection method causes significant reactions such as pain, swelling, and redness at the injection site, and its slow absorption and limited exposure restrict its clinical application. Therefore, new treatment options are urgently needed for patients with ER-positive breast cancer.
[0005] PCT / CN2021 / 093736 (application date May 14, 2021) describes a compound N-((S)-1-(3-fluoropropyl)pyrrolidine-3-yl)-6-((1S,3R)-3-methyl-2-(2,2,2-trifluoroethyl)-2,3,4,9-tetrahydro-1H-pyrido[3,4-b]indol-1-yl)pyridine-3-amine (compound of formula (I)), which is a selective estrogen receptor downregulator. In vitro and in vivo models show that compound (I) has a broad inhibitory effect on ER-positive cells. Summary of the Invention

[0115] The term “about” is used in this disclosure to mean approximately, around. When the term “about” is used in conjunction with a digit range, the range is modified by expanding the upper and lower limits of the stated digit range. Unless otherwise stated, the term “about” is used herein to modify the upper and lower limits of the stated value by a digit value with a 10% deviation.
[0116] Unless otherwise stated, the terms “comprise,” “comprise,” or “comprise,” and their English variations such as “comprises” or “comprising,” shall be understood in an open, non-exclusive sense, meaning “including but not limited to.”

Therefore, the phrases "some embodiments," "optional embodiments," or "implementation" appearing in different places throughout the specification do not necessarily all refer to the same embodiment. Furthermore, specific elements, structures, or features may be combined in one or more embodiments in any suitable manner.
[0118] The room temperature mentioned in this disclosure refers to 20 ± 5 °C.
[0119] The range "m~n" as described in this disclosure is a shortened representation of any combination of real numbers between m and n, where m and n are real numbers. The numerical range "0.5~2" includes, but is not limited to, 0.5, 0.6, 0.7, 0.8, 0.9, 1.0, 1.1, 1.2, 1.3, 1.4, 1.5, 1.6, 1.7, 1.8, 1.9, or 2.0, which have been listed herein.
[0120] The "X-ray powder diffraction pattern" described in this disclosure was obtained using Cu-Kα radiation measurements.
[0121] The “X-ray powder diffraction pattern or XRPD pattern” mentioned in this disclosure refers to the X-ray powder diffraction pattern obtained when X-rays are incident on an atomic surface of a crystal or part of a crystal sample with a lattice spacing of d at a grazing angle θ (the complementary angle of the incident angle, also known as the Bragg angle) with a grazing angle θ.
[0122] The crystal form disclosed in this disclosure is a substantially pure crystal. The term "substantially pure" as used in this application means that the purity of the crystal form is at least 85 wt%, preferably at least 90 wt%, and more preferably at least 95 wt%.
[0123] For the same compound and the same crystal form, the peak positions of their XRPD spectra are generally similar, but the relative intensity error may be large. It should also be noted that in the identification of mixtures, factors such as decreased content may cause the absence of some diffraction lines. In this case, it is not necessary to rely on all diffraction peaks observed in the high-purity sample; even a single diffraction peak may be characteristic for a given crystal.
[0124] The “2θ or 2θ angle” mentioned in this disclosure refers to the diffraction angle, where θ is the Bragg angle, and the unit is º or degrees; the error range of 2θ for each diffraction peak is ±0.20º.

[0135] Those skilled in the art will recognize that measurements of XRPD peak positions and / or intensities for a given crystalline form of the same compound will vary within a range of error. The 2θ value allows for an appropriate range of error. Typically, the range of error is expressed in "±". For example, 5.92 ± 0.2º 2θ represents a range of approximately 6.12 to 5.72. Depending on sample preparation techniques, calibration techniques applied to the instrument, human operational bias, etc., those skilled in the art will recognize that an appropriate range of error for XRPD diffraction angles (2θ) can be approximately ±0.20º, ±0.15º, ±0.10º, ±0.05º, or less. When used to describe XRPD plots, the terms "substantially consistent" or "substantially as shown" refer to a plot that includes at least 50%, at least 60%, at least 70%, at least 80%, at least 90%, at least 90%, or at least 99% of diffraction angles within a standard deviation of ±0.2º 2θ.
[0136] Because those skilled in the art recognize that measurements from DSC thermograms of a given crystalline form of the same compound will vary within error tolerances, the single-peak value (expressed in degrees Celsius) allows for an appropriate range of error. Typically, this range is expressed in "±". For the same crystalline form of the same compound, in successive analyses, errors in thermal transition temperature and melting point are typically within approximately ±5.0 °C. For example, a peak value of "186.82 ± 5 °C" indicates a range from 181.82 to 191.82. Depending on sample preparation techniques, calibration techniques applied to the instrument, human operational bias, etc., those skilled in the art recognize that an appropriate range of error for a single-peak value can be ±5.0, ±4.0, ±3.0, ±2.0, or less.

[0161] Identification of the absolute configuration of compounds of formula (I)
[0162] The NOESY spectrum (Figure 1) shows that the methyl hydrogen at the 3-position and the hydrogen at the 1-position of compound (I) have a significant NOE effect, proving that they are on the same side. The relative configuration of the pyridyl group at the 1-position and the methyl group at the 3-position on the 6-membered guanidine ring is trans. Since the absolute configuration of the carbon atom at the 3-position is R, the absolute configuration of the carbon atom at the 1-position is S.

[11] [:Mode] [(I)] [Study on Single Crystals of Succinate Compounds] []
[0186] Single crystals of succinate of formula (I) can be prepared by the following method: about 10 mg of the above-mentioned succinate of formula (I) (prepared by Example 9) is placed in a 3 mL sample bottle, 1500 µL of isopropyl ether and 900 µL of tetrahydrofuran are added, and after ultrasonic dissolution, the bottle mouth is sealed with a sealing film and a small hole is punched in the sealing film with a needle. After standing at room temperature for 3 days, crystals are precipitated. The obtained single crystal sample is identified as crystal type A by X-ray powder diffraction.
[0187] Single-crystal X-ray diffraction data were collected and the single-crystal structure was analyzed. The single-crystal structure data are shown in Table 5.

[0190] Testing of biological activity and related properties
[0191] Test Example 1: Saturated solubility test of compound of formula (I) and its pharmaceutically acceptable salt
[0192] Experimental instruments: rotary water bath constant temperature shaker (Taicang Wenerhuijin, model DSHZ-300A); high performance liquid chromatograph (Agilent 1260).
[0193] Experimental method: Accurately weigh about 10 mg of the test sample and place it in a 10 ml centrifuge tube. Add 1 ml of water and shake in a water bath constant temperature shaker (37℃, 150 rpm). If the sample is completely dissolved, add another 10 mg of sample and shake for about 24 hours. Take the supernatant, dilute it appropriately with acetonitrile, and perform HPLC detection. Determine its concentration according to the external standard method.
[0194] Experimental results: The saturated solubility of the test compounds is shown in Table 6. It can be seen that the 24-hour saturated solubility of compounds (I) succinate, benzoate and adipate is significantly better than that of free base. Table 6: Results of Saturated Solubility Test compound samples solvent Solubility at 24 hours (mg / ml) Formula (I) compound succinate (Example 10-1) water 3.27 Compound (I) benzoate (Example 8) 0.70 Compound (I) adipate (Example 7) 2.48 Compound of formula (I) (Example 1) 0.05
[0195] Test Example 2: Impurity Investigation Test of Compound (I) and its Pharmaceutical Salt Isomers
[0196] Experimental Methods: Compound (I) succinate (Example 10-1, crystal form A), compound (I) adipic acid salt (Example 7, crystal form C), and compound (I) (Example 1) were all packaged using two layers of polyethylene and one layer of pharmaceutical composite film. The changes in the content of isomer impurities of compound (I) under conditions of high temperature, light, high humidity, and accelerated reaction were investigated. The results are shown in Table 7. The isomer impurities are isomer 1 and isomer 2, with structures shown below. ND indicates that the impurity was not detected.
[0197] Experimental Results: Under high temperature, light, accelerated, and high humidity conditions, the isomer impurities of compounds (I) succinate and (I) adipate did not show significant growth. However, under 5 days of high temperature conditions, the content of isomer 1 impurity in the free state of compound (I) showed a significant increase. Furthermore, a new isomer 2 impurity was detected in the free state of compound (I) under high temperature, light, accelerated, and high humidity conditions. This experiment demonstrates that the above-mentioned salt forms of the compounds exhibit superior thermal stability and can more effectively suppress the formation of isomer impurities compared to the free base. [0198] Test Example 3: Stability Study of Pharmaceutical Salts of Compound (I)
[0199] Experimental method: Succinate (Example 10-1, crystal form A), benzoate (Example 8, crystal form B), and adipate (Example 7, crystal form C) of formula (I) were packaged with two layers of polyethylene and one layer of pharmaceutical composite film. The changes in the number and total amount of impurities under conditions such as high temperature, light, acceleration and high humidity were investigated. The results are shown in Table 8.

[0201] [Test Example] [4] [:Mode] [(I)] [Pharmacokinetic evaluation of compounds and their pharmaceutically acceptable salts in rats]
[0202] Experimental materials: Female SD rats were purchased from Beijing Vital River Laboratory Animal Technology Co., Ltd. MC (methylcellulose) was purchased from Sigma. Acetonitrile was purchased from Merck (USA).
[0203] Experimental methods:
[0204] Twelve female SD rats (200-300g, 6-8 weeks old) were randomly divided into four groups of three. Each group was orally administered compound (I), compound (I) benzoate, compound (I) succinate, and compound (I) adipic acid salt, respectively, at a dose of 24 mg / kg, in a 0.5% methylcellulose aqueous solution. Before the experiment, the rats were given normal water and fasted overnight. Food was resumed 4 hours after drug administration. Blood samples were collected from the jugular vein of each group before drug administration and at 0.25, 0.5, 1, 2, 4, 6, 8, 24, and 48 hours after drug administration. The collected whole blood samples were placed in K2EDTA anticoagulant tubes, centrifuged for 5 min (12000 rpm, 4ºC), and plasma was collected for analysis.
[0205] Take 10 μL of rat plasma sample [,] Add 150 μL of acetonitrile solvent (containing internal standard compound) to precipitate the protein, vortex for 5 min, centrifuge (14000 rpm) for 5 min, and dilute the supernatant 2-fold with water containing 0.1% (v / v) FA. Quantitative detection was performed using an LC-MS / MS system (AB Sciex Triple Quad 6500+). SD rat plasma standard curves and quality control samples were used concurrently with the sample concentration determination. For 10x diluted samples, 2 μL of sample was added to 18 μL of blank plasma, vortexed for 0.5 min, and then 300 μL of acetonitrile solvent (containing internal standard compound) was added to precipitate the protein. The remaining processing steps were the same as for the undiluted sample.

[0218] Experimental methods:
[0219] Animal Information: Balb / c nude mice, female, 6-8 weeks old, weighing approximately 18-22 grams. The animals were purchased from Shanghai Lingchang Biotechnology Co., Ltd. The mice were housed in an SPF-grade environment with individual ventilation in each cage. All animals had free access to standard certified commercial laboratory food and water.
[0220] Cell culture: Human breast cancer MCF-7 cell line was cultured in vitro under the following conditions: EMEM (cell culture medium) with 10% fetal bovine serum and 1% penicillin-drug antibiotics, at 37°C in a 5% CO2 incubator. Cells were passaged twice a week using 0.25% trypsin-EDTA digestion solution. When cell confluence reached 80%-90% and the required number was achieved, cells were harvested and counted.
[0221] Cell inoculation: 0.2 ml of MCF-7 cell suspension (containing 1 × 10⁷ cells) (DPBS:Matrix gel, volume ratio 1:1) was subcutaneously inoculated into the right posterior back of each mouse. Two days prior to cell inoculation, 17β-estradiol tablets were subcutaneously administered. On day 6 post-inoculation, when the average tumor volume reached 173.65 mm³, drug administration was initiated based on tumor volume, with grouping determined by randomization. Day 0 was the day of group administration.
[0222] Administration: The succinate of formula (I) (Example 10-1, crystal form A) was administered at a dose of 0.3, 1, 3, or 10 mg / kg (based on free base) orally (PO) once daily (QD) x 21 times. Fulvestrant was administered at a dose of 250 mg / kg subcutaneously (SC) once weekly (QW) x 4 times. Eight mice were used in each group.
[0223] Tumor measurements and experimental indicators:
[0224] The tumor diameter was measured twice weekly using calipers. The tumor volume was calculated using the formula: V = 0.5axb², where a and b represent the major and minor diameters of the tumor, respectively. The mouse's body weight was measured twice weekly.
[0225] The antitumor efficacy of the compound was evaluated using the tumor growth inhibition rate (TGI) (%). TGI (%) = [(1 - (mean tumor volume at the end of treatment - mean tumor volume at the start of treatment) / (mean tumor volume at the end of treatment in the solvent control group - mean tumor volume at the start of treatment in the solvent control group)] x 100%.
[0226] [Experimental Results:] []
[0227] See Table 12, Figures 13 and 14. Although a few mice experienced a weight loss exceeding 10%, this was unrelated to the dose of the test compound (I) succinate, and the weight of the mice in the test drug group was not significantly lower than that in the solvent control group. Considering that the estrogen patch in this model may affect the mouse's condition and weight, these individual weight losses were not related to the test compound (I) succinate. No mice died in any of the experimental groups.
[0228] The results showed that, on Day 20 after the start of administration, subcutaneous injection of 250 mg / kg of Fulvestrant (a control compound) into human breast cancer MCF-7 tumor-bearing mice once a week had a weak inhibitory effect on tumor growth in the MCF-7 xenograft tumor model (P<0.05). Oral administration of 0.3 mg / kg of the test compound (I) succinate to tumor-bearing mice once a day showed a certain inhibitory effect on tumor growth compared to the solvent control group (P<0.0001). Oral administration of 1 mg / kg, 3 mg / kg, or 10 mg / kg of the test compound (I) succinate to tumor-bearing mice once a day showed a significant inhibitory effect on tumor growth compared to the solvent control group (P<0.0001), exhibiting a good dose-dependent effect. At doses of 1 mg / kg and above, tumor regression began. In the human breast cancer MCF-7 subcutaneous tumor model, the compound (I) succinate at doses of 1 mg / kg and above (PO, QD) showed significantly superior antitumor activity compared to the control Fulvestrant (250 mg / kg, SC, QW) (P<0.0001). [10] [:Mode] [(I)] [Compound Pairs] [MCF7] [Study on the Degradation Effect of Intracellular Estrogen Receptors] [] 1. Experimental Objective The purpose of this experiment is to determine the degradation activity of compound (I) on endogenously expressed estrogen receptors in MCF7 cells, and to evaluate the activity of the compound based on DC 50 and maximum degradation efficiency. 2. Experimental Methods MCF7 cells (ATCC, HTB-22) were cultured in DMEM (Gibco, 11995-065) complete medium containing 10% fetal bovine serum. On day 1, MCF7 cells were seeded at a density of 3000 cells / well in 384-well plates using complete medium and cultured at 37ºC in a 5% CO2 cell incubator. The test compound was dissolved in DMSO at a storage concentration of 10 mM, diluted with Echo 550 (Labcyte Inc.), and added to the cell culture plates. The initial concentration of the test compound was 100 nM, with 3-fold serial dilutions, resulting in 9 concentration points. A blank control containing 0.5% DMSO was included, and duplicate wells were set up for each concentration point. The cells were cultured at 37ºC in a 5% CO2 cell incubator for 24 hours. Add paraformaldehyde to each cell culture well to the cell culture medium, with a final paraformaldehyde concentration of approximately 3.7% to fix the cells. After 30 minutes of treatment, discard the supernatant and wash each well once with 50 μL of PBS. Add PBS containing 0.5% v / v Tween-20 and treat the cells for 30 minutes, followed by washing with PBS once. Add blocking buffer (PBS containing 5% BSA and 0.05% Tween-20) and incubate at room temperature for 1 hour. Remove the blocking buffer and add a primary antibody mixture (anti-ER monoclonal antibody, Estrogen Receptor α (D8H8) Rabbit mAb, GST, #8644S, 1:1000 dilution; anti-GAPDH monoclonal antibody, GAPDH (D4C6R) Mouse mAb, GST, #97166S, 1:2000 dilution) and incubate at room temperature for 3 hours. Finally, incubate with PBST (PBS containing 0.05% BSA and 0.05% Tween-20). Wash three times with Tween-20; add detection secondary antibodies (800CW-goat anti-rabbit IgG, LI-COR, P / N: 926-32211, 1:1000 dilution; 680RD-goat anti-mouse IgG, LI-COR, #925-68070, 1:1000 dilution), incubate at room temperature in the dark for 45 minutes; wash three times with PBST, read the fluorescence signal of each well using an Odyssey CLx, and calculate the Chanel 800(ER) / Chanel 680 (GAPDH) values. Wells treated with 0.1 μM fulvestrant were used as a 100% degradation control, and the degradation rate at each concentration point was calculated. Data were analyzed using XLfit to calculate the degradation activity DC50 and maximum degradation rate Imax of the compounds. Data analysis is shown in Table 15. [surface]

[11] [:Mode] [(I)] [Compound Pairs] [MCF7] [Study on the inhibitory effect on cell proliferation] 1. Experimental Objective The purpose of this experiment was to determine the inhibitory effect of compound (I) on the in vitro proliferation of MCF7 cells and to evaluate the activity of the compound based on IC50. 2. Experimental Methods MCF7 cells (ATCC, HTB-22) were cultured in DMEM (Gibco, 11995-065) complete medium containing 10% fetal bovine serum. On day 1, MCF7 cells were seeded at a density of 500 cells / well in 384-well plates using complete medium and cultured overnight at 37ºC in a 5% CO2 cell incubator. On day 2, the test compound was added for treatment. A 10 mM solution of the compound was diluted and transferred to each well using an Echo550 (Labcyte Inc.). The initial concentration of the test compound in the cells was 100 nM, serially diluted 3-fold, resulting in 9 concentration points. A blank control containing 0.3% DMSO was included, and duplicate wells were set at each concentration point. Cells were cultured at 37ºC in a 5% CO2 cell incubator for 7 days. On day 8, the cell culture plates were removed. Add CellTiter-Glo® Luminescent Cell Viability Assay (Promega, G7573), incubate at room temperature for 10 minutes, and then read the luminescence signal value using an EnVision (PerkinElmer) multilabel microplate reader. Calculate the IC50 of the inhibitory activity of each compound using XLfit based on the compound concentration and luminescence signal value. 3. Data analysis is shown in Table 16. [0255] For an orally administered compound to reach its site of action and for effective absorption in the intestines, the compound needs to be in solution. Therefore, compounds with high inherent solubility may be more suitable for pharmaceutical use.
[0256] I. Materials and Reagents
[0257] The test compound was prepared according to the described method. Progesterone, the control drug, was purchased from Sigma. Phosphate buffer (pH 7.4) was prepared in our laboratory. Acetonitrile and methanol were purchased from Fisher. Other reagents were purchased commercially.
[0258] 1.5 ml flat-bottomed glass vial (BioTech Solutions); PTFE / silicone bottle stopper (BioTech Solutions); PTFE-coated stir bar; MultiScreenHTS HV (0.45 µm) 96 well plate filter (Millipore, MSHVN4510 or MSHVN4550); Eppendorf Thermomixer Comfort; Vacuum Manifold ORVMN96 (Orochem).
[0259] II. Experimental Procedure
[0260] Preparation of stock solution
[0261] Prepare 10 mM stock solutions of the analyte and the control drug progesterone using DMSO.
[0262] Apparent solubility determination steps
[0263] Add 30 µL of 10 mM analyte stock solution to the corresponding wells of the 96-well plate in the specified order. Add 970 µL of pH 7.4 phosphate buffer to the corresponding vial of the sample plate. The experiment is double-replicated. Add a stir bar to each vial and cap it with a PTFE / silicone stopper. Then place the sample tray in an Eppendorf Thermomixer Comfort and shake at 1100 rpm at 25°C for 2 hours. After 2 hours, remove the stopper, remove the stir bar with a large magnet, and then transfer the sample from the sample plate to the filter plate. Filter the sample using a vacuum pump to create negative pressure. Transfer 5 µL of the filtrate to a new sample plate, and then add 5 µL of DMSO and 490 µL of 50% ACN(IS).H₂O (acetonitrile:water = 1:1 internal standard). Depending on the peak shape, the sample diluent may be diluted with a certain proportion of 50% ACN(IS).H₂O to obtain a better peak shape. The dilution factor may be adjusted depending on the solubility of the analyte or the strength of its liquid chromatography response signal.
[0264] Sample analysis steps
[0265] The sample plate is placed into the sample tray of the autosampler, and the sample is evaluated by liquid chromatography-mass spectrometry.
[0266] III. Experimental Procedure
[0267] All calculations were performed using Microsoft Excel. Analysis and quantification of the sample filtrate were accomplished through qualitative and quantitative analysis of standard peaks of known concentrations using LC-MS. The apparent solubility of compound (I) in phosphate buffer at pH 7.4 was calculated. [13] [:Mode] [(I)] [The compound's effect on the blood-brain barrier in rats ()] [BBB] [Through ability] []
[0269] The ability of a drug to cross the blood-brain barrier and achieve sufficient exposure in the brain is crucial for its effectiveness against brain metastases. Therefore, by measuring the drug concentration in plasma and brain tissue after administration to animals, we can assess the distribution of the drug in the brain and determine whether the drug can inhibit tumor growth in an in situ brain model.
[0270] Experimental materials
[0271] Female SD rats were purchased from Beijing Vital River Laboratory Animal Technology Co., Ltd. MC (methylcellulose) was purchased from Sigma; acetonitrile was purchased from Merck (USA). PBS (phosphate buffered saline) was purchased from Sangon Biotech.
[0272] Experimental methods
[0273] Six female SD rats (200-300g, 6-8 weeks old) were randomly divided into two groups of three each. Each group was administered compound (I) and 0.5% methylcellulose aqueous solution as solvent. Before the experiment, the rats were given normal water and fasted overnight. Feeding resumed four hours after drug administration. Plasma and brain tissue were collected from each group two hours after drug administration. Whole blood samples were placed in K2EDTA anticoagulant tubes, centrifuged for 5 min (12,000 rpm, 4ºC), and plasma was collected for analysis. Tissue samples were blotted dry with filter paper and stored at -80°C for analysis.
[0274] Take 10 μL of rat plasma sample, add 150 μL of acetonitrile solvent (containing internal standard compound) to precipitate protein, vortex for 5 min, centrifuge (14,000 rpm) for 5 min, and dilute the supernatant 2-fold with water containing 0.1% (v / v) FA. Quantitatively detect the protein using an LC-MS / MS system (AB Sciex Triple Quad 6500+). For 10x diluted samples, take 2 μL of sample and add 18 μL of blank plasma, vortex for 0.5 min, add 300 μL of acetonitrile solvent (containing internal standard compound) to precipitate protein, and follow the same steps as the undiluted sample. A standard curve of SD rat plasma and plasma quality control samples were used concurrently with the determination of plasma sample concentrations.
[0275] Rat brain tissue samples were first homogenized with 4 times their volume of PBS homogenate. 20 μL of the homogenate was diluted with 20 μL of blank mouse plasma, and then 600 μL of acetonitrile (containing an internal standard compound) was added to precipitate the protein. After vortexing for 5 min, the sample was centrifuged (14,000 rpm) for 5 min. The supernatant was diluted 2-fold with water containing 0.1% (v / v) FA and quantitatively analyzed using an LC-MS / MS system (AB Sciex Triple Quad 6500+). The compound of formula (I) exhibited excellent blood-brain barrier permeability, resulting in high drug exposure in rat brain tissue.
