[0005] Systemic lupus erythematosus (SLE) is a rheumatic immune disease that can cause irreversible damage to organs. Lupus nephritis is the main complication of SLE, manifested as proteinuria, hematuria, tubular urine, and progressive decline in renal function, and eventually even requires renal transplantation or long-term dialysis treatment, which seriously affects the quality of life of patients and is called “immortal cancer”. Currently, there are limited treatment options, and more therapeutic drugs are urgently needed to meet clinical needs.

[0006] IL-17A is the main pro-inflammatory factor of the IL17 family. IL-17A is secreted by Th17, CD8+ (Tc17), γδT, NKT and ILC3s cells. IL36 is a member of the ILI superfamily and consists of three agonists IL36a, IL36B, and IL36γ and a natural receptor antagonist IL-36Ra. Once activated, this pathway promotes the production of a plurality of inflammatory factors. Literature reports that IL17 is highly expressed in patients with active SLE, and animal models and clinical trial data show that blocking IL17 has the potential to treat SLE and lupus nephritis (LN). Currently, Novartis and Jiangsu Hengrui Pharmaceuticals Co., Ltd. are conducting Phase III clinical trials to investigate the efficacy of IL17 monoclonal antibodies in the treatment of LN. The levels of IL36a and IL36γ in the serum of SLE patients are significantly increased and positively correlated with the SLE disease activity index (SLEDAI).

[0007] However, at present, there is still an urgent need to develop more effective treatment options to reduce the mortality rate of patients, slow down the rate of increase in urine protein levels, and reduce renal pathological damage.SUMMARY

[0008] The present application provides a method for preventing and / or treating systemic lupus erythematosus by targeting IL-17A and IL-36R. In the present application, by jointly administering an antigen binding protein targeting IL-17A and an antigen binding protein targeting IL-36R, or by administering a bispecific antigen binding protein targeting IL-17A and IL-36R, the synergistic effect of the two targets can be exerted, thereby more effectively treating systemic lupus erythematosus compared with monotherapy targeting a single target alone. By targeting IL-17A and IL-36R, one or more of the following therapeutic effects can be achieved on systemic lupus erythematosus or diseases and / or conditions caused by systemic lupus erythematosus: (1) the increase in urine protein level of the subject can be inhibited; (2) the renal pathological damage in the subject can be significantly improved; and (3) the renal fibrosis in the subject can be inhibited.

[0432] This example uses the stable cell line CHO-hIL-36R OE (from Huabo Biopharm) to investigate whether the bispecific antibody HB0043 can enrich IL-17A on and around the cell surface by targeting cell surface-expressed IL-36R. The functional difference compared to the single-target antibody HB0034 is also evaluated simultaneously.

[0433] CHO-hIL-36R OE cells were cultured to the logarithmic growth phase, resuspended in serum-free RPMI-1640, and starved for 12-16 hours. Cells were then counted and adjusted to a density of 1×106 / mL (cell viability >95%). After grouping according to experimental needs, 100 nM of antibodies (HB0043 or HB0034) were added to each group under ice-bath conditions and incubated for 30 minutes. After incubation, cells were washed twice with PBS, resuspended in RPMI-1640 containing 100 ng / ml IL-17A, plated in a low-attachment round-bottom 96-well plate, and cultured for 12-16 hours at 37° C. under 5% CO2. A bispecific antibody comprises a first binding domain targeting IL-17A, and a second binding domain targeting IL-36R,wherein said first binding domain targeting IL-17A comprises HCDR1-3 and LCDR1-3, said HCDR1 comprises an amino acid sequence as set forth in SEQ ID NO: 15, said HCDR2 comprises an amino acid sequence as set forth in SEQ ID NO: 14, said HCDR3 comprises an amino acid sequence as set forth in SEQ ID NO: 13, said LCDR1 comprises an amino acid sequence as set forth in SEQ ID NO: 19, said LCDR2 comprises an amino acid sequence as set forth in SEQ ID NO: 18, and said LCDR3 comprises an amino acid sequence as set forth in SEQ ID NO: 17; andwherein said second binding domain targeting IL-36R comprises HCDR1-3 and LCDR1-3, said HCDR1-3 and LCDR1-3 comprise an amino acid sequence as shown in any one of the groups consisting of: (1) HCDR 1: SEQ ID NO:3, HCDR 2: SEQ ID NO:2, HCDR3: SEQ ID NO: 1, LCDR 1: SEQ ID NO:7, LCDR 2: SEQ ID NO:6 (STS), LCDR 3: SEQ ID NO:5; and(2) HCDR 1: SEQ ID NO:29, HCDR 2: SEQ ID NO:28, HCDR3: SEQ ID NO: 27, LCDR 1:SEQ ID NO:33, LCDR 2: SEQ ID NO:32 (AAS), LCDR 3: SEQ ID NO:31.

2. The bispecific antibody according to claim 1, wherein said first binding domain comprises VH and VL, said VH comprises an amino acid sequence as set forth in SEQ ID NO: 16, and said VL comprises an amino acid sequence as set forth in SEQ ID NO: 20.

3. The bispecific antibody according to claim 1, wherein said second binding domain comprises VH and VL, said VH and VL comprise an amino acid sequence as shown in any one of the groups consisting of: (1) VH: SEQ ID NO: 4, VL: SEQ ID NO: 8; and (2) VH: SEQ ID NO: 30, VL: SEQ ID NO: 34.

4. The bispecific antibody according to claim 1, wherein said first binding domain comprises an antibody heavy chain constant region and an antibody light chain constant region.

5. The bispecific antibody according to claim 4, wherein said antibody heavy chain constant region is derived from a heavy chain constant region of IgG and said light chain constant region is derived from IgK.

6. The bispecific antibody according to claim 4, wherein said antibody heavy chain constant region comprises an amino acid sequence as set forth in SEQ ID NO: 21, and the light chain constant region comprises an amino acid sequence as set forth in SEQ ID NO: 10.

7. The bispecific antibody according to claim 1, wherein said second binding domain is scFv.

8. The bispecific antibody according to claim 7, wherein said scFv comprises an amino acid sequence as set forth in SEQ ID NO: 24.

9. The bispecific antibody according to claim 1, wherein said second binding domain is directly or indirectly linked to said first binding domain.

10. The bispecific antibody according to claim 9, wherein said second binding domain is linked to said first binding domain via a linker.

11. The bispecific antibody according to claim 1, wherein said bispecific antibody is a dual-chain antibody or a single-chain antibody.

The bispecific antibody according to claim 1, wherein said bispecific antibody has a structure represented by formula I from N-terminus to C-terminus:wherein,VH represents the heavy chain variable region of the anti-IL-17A antibody;VL represents the light chain variable region of the anti-IL-17A antibody;CH1, CH2 and CH3 represent the heavy chain constant regions CH1, CH2 and CH3 of the anti-IL-17A antibody, respectively;CL represents the light chain constant region of the anti-IL-17A antibody;ScFv represents the ScFv of the anti-IL-36R antibody;L is a linker element;“~” represents a disulfide bond;“—” represents a peptide bond;wherein, the bispecific antibody has an activity of binding to IL-17A and IL-36R simultaneously.

The bispecific antibody according to claim 12, wherein said bispecific antibody is formed by fusion of the anti-IL-17A antibody and the ScFv of the anti-IL-36R antibody, and has two pairs of peptide chains that are symmetrical to each other; each pair of peptide chains comprises a light chain L chain and a heavy chain H chain; all the peptide chains are linked via disulfide bonds, and any pair of peptide chains has a structure of the H chain and the L chain represented by formula I from N-terminus to C-terminus.

14. The bispecific antibody according to claim 12, wherein said bispecific antibody is a homodimer of the peptide chain of the structure represented by formula I.

15. The bispecific antibody according to claim 12, wherein the linker element is a linker peptide.

16. The bispecific antibody according to claim 15, wherein the amino acid sequence of said linking peptide is (G4S) n, wherein said n is a positive integer.

17. The bispecific antibody according to claim 13, wherein said H chain of the bispecific antibody has an amino acid sequence as set forth in SEQ ID NO: 25; and said L chain of the bispecific antibody has an amino acid sequence as set forth in SEQ ID NO: 23.

The bispecific antibody according to claim 1, wherein said bispecific antibody has one or more of the following properties:(1) binding to IL-36R with high affinity;(2) binding to IL-1Rrp2 with high affinity;(3) blocking the binding of IL-36α, IL-36β, and IL-36γ to IL-36R; and(4) blocking IL-36α-, IL-36β-, and IL-36γ-mediated cytokine secretion.

19. A method for preventing and / or treating systemic lupus erythematosus, comprising administering the bispecific antibody according to claim 1 to a subject in need thereof.

20. The method according to claim 19, wherein said systemic lupus erythematosus comprises lupus erythematosus nephritis.
