Abstract

The invention relates to a process for propagating the multi-petaled yellow jasmine (Ochna integerrima (Lour.) Merr.) using tissue culture techniques from leaf callus tissue samples, in the field of high-tech agriculture. This process aims to overcome the limitations of traditional propagation methods such as seed sowing and grafting, which have low survival rates, genetic inhomogeneity, and difficulty in scaling up production. The proposed process includes the following main steps: (i) selection and sterilization of young leaves; (ii) induction of callus formation on MS medium supplemented with 2,4-D and BA; (iii) shoot regeneration from callus on medium containing kinetin and BA; (iv) rapid shoot multiplication using BA-rich medium; (v) root formation with IBA to form complete plants; and (vi) acclimatization of seedlings in greenhouse conditions with suitable substrate. Experimental results showed that the process was highly effective with a large shoot multiplication coefficient, increasing from an average of 7.42 to 14.9 shoots after each cycle, while also demonstrating good rooting ability with an average of 4.49 roots.As a result, the seedlings develop uniformly, healthily, and achieve a survival rate of up to 96.30% after the training period. Furthermore, the seedlings maintain the morphological and genetic characteristics of the mother plant, ensuring seed quality. Based on this, the invention offers outstanding efficiency in producing a large quantity of seedlings in a short time, with consistent quality, disease-free, and genetically stable. Simultaneously, this process contributes to standardizing in vitro propagation techniques for the multi-petaled yellow apricot tree, opening up a new direction in asexual propagation of economically valuable woody plant species. With these advantages, the invention has great practical significance, meeting the needs of industrial-scale seedling production in Vietnam.
