Abstract
Description
Composition for improving skin condition
[0001] The present application relates to a composition for improving skin condition comprising Lactobacillus plantarum CJLP55 or a culture solution thereof.
[0002] To improve skin health and enhance its beauty, interest in inner beauty, which is achieved through the consumption of various materials and products, has grown in addition to the existing perception that focused solely on outer beauty, such as cosmetics applied to the skin surface. Skin troubles such as acne and dermatitis that occur on the skin's surface are known to be caused by increased sebum secretion, hyperkeratinization of hair follicles, colonization of harmful skin bacteria, and inflammation. To address the root causes of these skin troubles, research is also being conducted on the "gut-skin axis," a theory about the correlation between the gastrointestinal tract and the skin.
[0003] Meanwhile, there have been attempts and studies to utilize lactic acid bacteria, known as beneficial microorganisms found in various fermented foods, to improve skin health. Research has shown that applying lactic acid bacteria cultures or compositions containing them to the skin as cosmetics, for example, has antibacterial activity against harmful bacteria living on the skin and can improve wrinkles and aging. However, the effects of ingesting lactic acid bacteria, rather than directly applying them, on skin condition are unknown, and no research has been conducted.
[0004] Accordingly, the inventor of the present application has completed the invention of the present application by discovering a new effect and use of a lactic acid bacteria strain that can function as a probiotic and is beneficial to health, and further has the effect of improving skin condition and can more significantly inhibit the growth of skin-harmful bacteria compared to other strains.
[0005] The purpose of this application is to provide a composition for improving skin condition that can suppress the occurrence and progression of skin troubles such as acne, reduce oil in the skin, and maintain moisture.
[0006] In order to achieve the above purpose, the present application provides a composition for improving skin condition, comprising Lactobacillus plantarum CJLP55 or a culture solution thereof.
[0007] The composition for improving skin condition of the present application has the advantage of being able to improve the severity of acne, reduce oil and sebum in the skin, maintain moisture content, and improve skin acidity based on the growth inhibitory effect on skin harmful bacteria such as Propionibacterium genus, thereby improving the condition of the skin and inhibiting the occurrence and progression of skin troubles.
[0008] However, the effects of the present application are not limited to the effects mentioned above, and other effects not mentioned will be clearly understood by those skilled in the art from the description below.
[0009] Figures 1a and 1b show a comparison of the number of subjects with improved oiliness measured after consuming each food for 12 weeks in the lactic acid bacteria (Lactobacillus plantarum CJLP55) intake group and the placebo group in a human application test. Figure 1a is a graph comparing the number of subjects with improved oiliness by 30% or more, and Figure 1b is a graph comparing the number of subjects with improved oiliness by 50% or more. In each graph, the left side represents the ITT analysis results, and the right side represents the PP analysis results.
[0010] Figure 2 is a graph showing the ratio of each species of microorganism in the genus Propionibacterium based on the results of ITT (Figure 2A) and PP (Figure 2B) analyses of the base sequence of skin flora in a human application test. In both graphs, P. acnes was 94%, P. unassigned was 5%, and P. granulosum was 1%.
[0011] Hereinafter, the present application will be described in detail.
[0012]
[0013] The present application provides a composition for improving skin condition, comprising Lactobacillus plantarum CJLP55 or a culture solution thereof.
[0014] In this application, the term "culture solution" means the culture solution itself obtained by culturing a strain, or the culture supernatant obtained by removing the strain therefrom, and the filtrate, concentrate or dried product thereof, and may be used interchangeably with "culture supernatant", "conditioned culture solution" or "conditioned medium".
[0015] In this application, the term “improvement” means any action that includes amelioration, suppression, or delay of symptoms, and may be used interchangeably with prevention or treatment.
[0016] The above prevention may be any act of administering a composition to a subject to suppress or delay the onset of the disease, and the above treatment may be any act of administering a composition to a subject to improve the symptoms of the disease that has already been infected.
[0017] In this application, the term "improving skin condition" means any act of improving skin condition regardless of cosmetic or medical aspects, and may be used interchangeably with improving skin health or improving hypersensitive skin condition.
[0018] The above Lactobacillus plantarum CJLP55 is a type of lactic acid bacterium, a Gram-positive bacterium, and has the characteristic of being able to grow under both aerobic and anaerobic conditions. Specifically, it may be a microorganism deposited with the accession number KCTC 11401BP (Deposit institution: Korea Research Institute of Bioscience and Biotechnology Gene Bank, Deposit date: 2008.10.16), and may be isolated from fermented foods such as kimchi, fermented vegetables, soybean paste, soy sauce, cheonggukjang, and salted seafood. The above Lactobacillus plantarum CJLP55 has excellent characteristics of acid resistance to gastric acid, bile acid resistance, and adhesion to intestinal epithelial cells, and thus can have a beneficial effect on the intestinal flora in the gastrointestinal tract of animals, including humans, and thus can be used as probiotics.
[0019] The above culture solution is obtained by culturing the Lactobacillus plantarum CJLP55, and may be the culture solution itself containing the lactic acid bacteria cells, or a culture supernatant obtained by removing the lactic acid bacteria cells therefrom, and may also be a filtrate, concentrate, or dried product thereof. The culture solution from which the lactic acid bacteria cells have been removed contains components produced and secreted by the Lactobacillus plantarum CJLP55, and thus may have skin condition improvement activity.
[0020] The above filtrate is obtained by removing floating solid particles from a culture solution of Lactobacillus plantarum CJLP55 to obtain only a water-soluble supernatant excluding the sediment. The particles may be filtered out using a filter such as cotton or nylon, for example, a filter of 0.2 ㎛ to 5 ㎛, or a cryofiltration method, centrifugation method, etc., but is not limited thereto.
[0021] The above concentrate increases the solid concentration of the culture medium, and may be a concentrate of the culture medium containing the lactic acid bacteria cells or a concentrate of the culture supernatant from which the lactic acid bacteria cells have been removed. The concentrate may be concentrated by vacuum concentration, plate concentration, thin film concentration, etc., but is not limited thereto, and may be performed at a temperature of 40°C to 60°C using a known concentrator, for example. Depending on the concentration of the concentrate, the content of the culture medium included in the composition of the present application can be appropriately adjusted.
[0022] The above-mentioned dried product includes, but is not limited to, those dried by methods such as freeze-drying, vacuum drying, hot air drying, spray drying, reduced pressure drying, spray drying, foam drying, high-frequency drying, and infrared drying. For example, when a freeze-dried product obtained by freeze-drying a culture solution containing Lactobacillus plantarum CJLP55 cells is used, there is an advantage in orally consuming, formulating, packaging, storing, etc. a composition containing the same, and the strain can be preserved for a long period of time, so there is an advantage in that the strain that was freeze-dried in a subject to which the composition of the present application is administered, particularly in the intestine, can exhibit normal physiological activity again and perform growth and metabolism.
[0023] The Lactobacillus plantarum CJLP55 or its culture solution may be included in the composition of the present application in an amount of 3 wt% to 15 wt%. Specifically, the Lactobacillus plantarum CJLP55 or its culture solution may be included in an amount in a range consisting of a lower limit selected from 3 wt%, 3.5 wt%, 4 wt%, 4.5 wt%, 5 wt%, 5.5 wt%, 6 wt%, 6.5 wt%, and 7 wt% and / or an upper limit selected from 15 wt%, 14 wt%, 13 wt%, 12 wt%, 11 wt%, 10.5 wt%, 10 wt%, 9.5 wt%, and 9 wt%. For example, the content may be 3 to 15 wt%, 3.5 to 14 wt%, 4 to 13 wt%, 4.5 to 12 wt%, 5 to 11 wt%, 5.5 to 10.5 wt%, 6 to 10 wt%, 6.5 to 9.5 wt%, or 7 to 9 wt%. When the content of Lactobacillus plantarum CJLP55 or its culture solution is within the above range, when ingested, it can perform functions such as proliferation of beneficial lactic acid bacteria in the intestines, inhibition of harmful bacteria, and smooth bowel movements, and can sufficiently improve the effect of improving skin condition, while not feeling rejection in terms of taste and smell when ingesting the composition of the present application.
[0024] The composition may further include maltodextrin and glucose. The maltodextrin and glucose may be included as excipients. The composition may include, without limitation, any ingredient that can be commonly used as an excipient in the technical field of the present application, as long as it does not inhibit the skin condition improvement effect by Lactobacillus plantarum CJLP55 or its culture medium included in the composition of the present application. The maltodextrin and glucose, when included in the composition of the present application, may exhibit the effect of inhibiting the secretion of free fatty acids in epidermal cells and / or sebaceous glands of a person ingesting the composition. The content of the maltodextrin and glucose may be set without limitation at a level that does not inhibit the skin condition improvement effect by Lactobacillus plantarum CJLP55 or its culture medium. For example, the maltodextrin may be included in the composition of the present application in the same amount as the glucose.
[0025] The composition of the present application may be for oral administration. The term "for oral administration" refers to the composition of the present application being administered / ingested through the oral cavity of a human or animal, and thus, the composition of the present application may be used in the form of a pharmaceutical or food product. When the composition is administered orally, the Lactobacillus plantarum CJLP55 contained therein may function as a probiotic in the gastrointestinal tract of a human or animal, and may also have an effect on the skin, thereby improving its condition.
[0026] The improvement in the skin condition may be at least one selected from the group consisting of sebum suppression, sebum component improvement, acne improvement, and dermatitis improvement. The composition of the present application can suppress sebum by reducing the amount of sebum or secretion. In addition, the sebum component can be improved by reducing the content of components in sebum that may cause skin trouble, such as neutral fat, cholesterol, cholesterol ester, and free fatty acid, or increasing the content of components in sebum that may improve skin condition by showing a skin moisture retention effect, such as ceramide. The improvement in acne may be a reduction in nodules, papules, pustules, and comedones, and a reduction in the severity of acne. In addition, the composition of the present application can improve dermatitis that may be caused by sebum by reducing the amount of sebum by suppressing sebum secretion and improving components in sebum.
[0027] The composition may reduce at least one selected from the group consisting of neutral fat, cholesterol, cholesterol ester, and free fatty acid in sebum, or increase ceramide. The neutral fat may be secreted from a sebaceous gland, and the cholesterol, cholesterol ester, and free fatty acid may be secreted from both the sebaceous gland and the epidermis. The ceramide may be ceramide 1 to 7, and specifically, ceramide 2, and an effect of improving skin moisturization may be exhibited according to an increase in ceramide 2.
[0028] The composition described above can reduce skin oil and increase skin moisture. Specifically, the composition of the present application can reduce the amount of oil (e.g., neutral fat, cholesterol, cholesterol ester, free fatty acid, etc.) on the skin surface or reduce the amount of sebum. Furthermore, the composition can maintain skin moisture by increasing skin moisture or inhibiting moisture loss, thereby exhibiting a moisturizing effect on the skin.
[0029] The composition described above can improve skin acidity. Improving acidity refers to reducing the pH of the skin or inhibiting an increase in the pH of the skin. Skin acidity can be used as a key indicator of skin health. While healthy skin generally maintains a slightly acidic pH of 4 to 6, skin pH can increase when skin diseases such as atopic dermatitis or acne occur. Therefore, reducing the pH can improve skin condition. Skin acidity can be determined by the total content of lactic acid, free amino acids, and free fatty acids produced in the epidermis, and the composition described above can increase the lactic acid content of the skin.
[0030] The composition may have antibacterial activity against microorganisms of the genus Propionibacterium (Cutibacterium) or the genus Staphylococcus. The microorganisms of the genus Propionibacterium or the genus Staphylococcus may cause skin diseases such as acne and / or dermatitis. The composition of the present application may have antibacterial activity by inhibiting the growth and development of the microorganisms, and may reduce the number of the microorganisms or inhibit the increase in the number of the microorganisms. For example, the composition may have antibacterial activity against the microorganisms distributed on the skin surface, and thus may prevent, treat, and improve skin diseases caused by the microorganisms.
[0031] The above Propionibacterium genus microorganism may be Propionibacterium acnes (or Cutibacterium acnes). Propionibacterium acnes is a skin-harmful bacterium that can cause acne or dermatitis, and can grow using fatty acids secreted from sebaceous glands. The composition of the present application may have antibacterial activity by inhibiting the growth and development of Propionibacterium acnes, and thus may prevent, treat, and improve skin diseases caused by Propionibacterium acnes.

[0050]
[0051] Hereinafter, the present application will be described in detail through experimental examples.
[0052] However, the following experimental examples are intended to specifically illustrate the present application, and the contents of the present application are not limited by the following examples.
[0053]
[0054] [Experimental Example 1]
[0055] Confirmation of the growth inhibitory effect of Lactobacillus plantarum CJLP55 against P. acnes, the causative agent of acne.
[0056] In order to confirm the growth inhibitory effect of Lactobacillus plantarum CJLP55 strain on Propionibacterium acnes (or Cutibacterium acnes, hereinafter referred to as 'P. acnes'), which is known to be the causative agent of skin troubles such as acne, various types of lactic acid bacteria were inoculated together in the culture medium of P. acnes and it was confirmed in vitro whether the growth of P. acnes was inhibited.
[0057] The Lactobacillus plantarum CJLP55 strain is a strain disclosed in Korean Patent Publication No. 10-1255050 B1, and was deposited with the Korea Research Institute of Bioscience and Biotechnology Gene Bank on October 16, 2008 as KCTC 11401BP.
[0058] As a lactic acid bacteria strain, the standard strain KCTC 3108 of Lactobacillus plantarum was used, and Lactobacillus rueteri KCTC 3594, which is known to have an activity of inhibiting the growth of P. acnes (Mi-Sun Kang et al., 2012, The Journal of Microbiology, 50(1), 137-142), and the Lactobacillus plantarum CJLP55 strain of the present application were used. In the case of the acne-causing bacterium P. acnes (accession number: KCTC 3314, culture medium: Reinforced Clostridial Medium, RCM medium, culture temperature: 37 ℃), it was cultured under anaerobic conditions by sealing it in an anaerobic culture tank using a gas pack system (AnaeroPack, MITSUBISHI GAS CHEMICAL).
[0059] The Lactobacillus plantarum CJLP55 strain of the present invention, and the standard strains Lactobacillus plantarum KCTC 3108 and Lactobacillus reuteri KCTC 3594 strains were inoculated into MRS medium (Difco 0881) at a concentration of 1.0 × 10 7 After culturing for 24 hours by spotting at CFU / ㎖, the P. acnes strain was inoculated at 1.0 × 10 on RCM medium containing 0.8% agar. 7 CFU / ㎖ were inoculated and overlaid. After additional incubation for 24 h, the diameter of the clear zone, which appears as the growth of P. acnes strains is inhibited around the colonies of each experimental strain and control strain, was measured.
[0060] As a result, as shown in Table 1 below, no inhibition zone was formed at all in the colony of the standard strain of Lactobacillus reuteri, but inhibition zones were formed in the colonies of the standard strain of Lactobacillus plantarum (KCTC 3108) and the Lactobacillus plantarum CJLP55 strain of the present invention. At this time, the diameter of the inhibition zone formed around the colony of the Lactobacillus plantarum CJLP55 strain was found to be larger than that of the colony of the standard strain. Since the size of the diameter of the inhibition zone is larger when the antibacterial activity exhibited by the strain of the colony is excellent, it was confirmed that the Lactobacillus plantarum CJLP55 strain of the present invention exhibits antibacterial activity that inhibits the growth of P. acnes, and has better antibacterial activity against P. acnes compared to the general Lactobacillus plantarum standard strain.
[0061] Diameter of inhibition zone (mm) Lactobacillus reuteri standard strain (KCTC 3594) - Lactobacillus plantarum standard strain (KCTC 3108)++ Lactobacillus plantarum CJLP55+++
[0062] (-: no inhibition zone, +: less than 10.0 mm, ++: 10.0~20.0 mm, +++: more than 20.0 mm)
[0063] In view of the above results, the Lactobacillus plantarum CJLP55 strain of the present invention exhibits better activity in inhibiting the growth of P. acnes than other lactic acid bacteria strains that have been known to be able to inhibit the activity of acne-causing bacteria to some extent, and in particular, it was newly confirmed that it has a characteristic in which the growth inhibition activity of acne-causing bacteria is more excellent than that of KCTC 3108, which is a standard strain of Lactobacillus plantarum. Therefore, the Lactobacillus plantarum CJLP55 of the present invention or a culture solution containing the same can be used as an ingredient for improving acne that may be caused by the P. acnes strain, and thus can be usefully used as an effective ingredient of a composition for improving skin conditions.

[0065] [Experimental Example 2]
[0066] Confirmed effects of Lactobacillus plantarum CJLP55 on reducing sebum and improving skin problems.
[0067] To confirm the effects of Lactobacillus plantarum CJLP55, which has been shown to be effective in suppressing the growth of P. acnes, the causative agent of acne, on reducing sebum and improving skin problems in humans, a human application study was conducted. The human application study was conducted with 90 adults and received approval from the Kyung Hee University Institutional Review Board (Approval Number: KHSIRB 18-016).
[0068]
[0069] [2-1] Selection of research subjects and assignment to experimental groups
[0070] Healthy men and women aged 18-39 years with acne were recruited and selected from the Department of Dermatology, Wonju Severance Christian Hospital, based on the sebum secretion standards by area (SM815 sebum meter, Courage-Khazaka, Germany) according to Table 2 below and the acne severity evaluation system (Investigator's Global Assessment (IGA), provided by the Korea Food and Drug Safety Evaluation Institute) according to Table 3 below. Subjects whose sebum secretion levels in the forehead and T-zone were determined to be 'oily' and whose acne severity was grade 2-3 were selected. The subjects were divided into a placebo group and a lactic acid bacteria (Lactobacillus plantarum CJLP55) intake group while maintaining a double-blinded study, and the test was conducted.
[0071] Separate statistical analysis was performed on the results of all subjects in the human application trial (ITT, intent to treat) and the results of subjects whose oil levels were relevant to the subjects at the time of selection but whose oil levels were measured as 85 or higher at week 0 or who took antibiotics or received dermatological procedures during the 12-week intake period (PP, per protocol).
[0072] Forehead, T-zone, scalp: Less sebum < 70, Normal 70 ~ 150, Oily > 150
[0073] Grade Characteristics 0 No lesions 1 Almost no lesions (Nodules: None, Papules / Pustules: 1 or less, Comedones: Few) 2 Mild (Nodules: None, Papules / Pustules: 5 or less, Comedones: Slight) 3 Severity (Nodules: 1 or less, Papules / Pustules: 6 or more, Comedones: Many) 4 Severe (Nodules: 5 or less, Papules / Pustules: Many, Comedones: Many)
[0074] [2-2] Manufacturing and consumption methods of food
[0075] The placebo (control food) administered to the placebo group was a product that did not contain Lactobacillus plantarum CJLP55, and was manufactured to contain 50% by weight of maltodextrin and 50% by weight of powdered glucose. The food administered to the lactic acid bacteria intake group was manufactured to contain 9% by weight of Lactobacillus plantarum CJLP55, 45.5% by weight of maltodextrin, and 45.5% by weight of powdered glucose. Each food manufactured as described above had the same shape, color, and flavor, and the intake amount and intake method was to take one sachet (2 g) once a day, regardless of meals, either by pouring it into the mouth as is, or mixing it with 20 to 40 ml of water or milk and taking it together for a total of 12 weeks, and to take it with a 2-hour interval when taking other medications.
[0076] In the case of the above lactic acid bacteria intake group, 10 was the minimum amount that the probiotics reach the intestines and show beneficial functions. 10 Lactobacillus plantarum CJLP55 was administered to animals for 1 CFU / day.
[0077]
[0078] [2-3] Statistical analysis method
[0079] Descriptive statistics of all data obtained from the human application trial were performed using the SPSS 23.0 program (IBM, USA). Student's paired t-tests were performed on the data from all subjects before and after intake (weeks 0-6 or 0-12). Between-group significance was assessed for the 12-week intake period using Student's unpaired t-tests. Significance was determined at a P value of <0.05.
[0080]
[0081] [2-4] Confirmation of the effect of improving oil, moisture, and pH by consuming lactic acid bacteria
[0082] First, the placebo group and the probiotic group were ingested the food manufactured according to the above Experimental Example 2-2 for 12 weeks according to the above consumption method, and then the changes in skin oil, moisture, and pH were observed. At weeks 0, 6, and 12, the subjects were asked to stay in a space maintained at a temperature of 18-26 ℃ and a humidity of 26-53% for more than 30 minutes after washing their faces, and then the oil, moisture, and pH were measured on the forehead and T-zone using a probe using a Sebum meter (SM815, Courage-Khazaka Electronic GmbH, Germany), a Corneum meter (CM825), and a pH meter (PH905), respectively. The moisture, oil, and acidity values for each week for each subject are the average of five repeated measurements.
[0083] Among the results of the oil, moisture, and pH measurements, the results of the 42 placebo group and the 42 lactic acid bacteria group corresponding to the ITT analysis are shown in Table 4 below, and the results of the 39 placebo group and the 39 lactic acid bacteria group corresponding to the PP analysis are shown in Table 5 below.
[0084] The results of the ITT analysis on changes in oil content showed that, unlike the placebo group, whose individual oil content range gradually widened, the individual oil content range in the probiotic group gradually narrowed as the intake period increased. The oil content in the probiotic group at weeks 6 and 12 was found to be significantly reduced compared to week 0, and there was also a significant difference between the groups when compared to the placebo group. The results of the PP analysis on changes in oil content also showed that, unlike the placebo group, the oil content range narrowed as the intake period passed, and the oil content at weeks 6 and 12 significantly decreased compared to week 0. There was also a significant difference between the groups when compared to the placebo group. Based on the results of the ITT and PP analyses above, the oil and sebum reduction effect of Lactobacillus plantarum CJLP55 intake was significantly observed from week 6, and the effect was significantly apparent by week 12. In particular, the number of subjects whose oil content improved by 30% or 50% or more compared to week 0 was statistically significantly higher in both the ITT and PP analyses (Fig. 1a and Fig. 1b).
[0085] In the case of ITT and PP analysis of changes in epidermal moisture, which is an indicator of skin hydration, moisturization improved in both the placebo group and the probiotic group until the 6th week, and there was no significant difference between groups. However, from the 12th week, moisturization increased more significantly in the probiotic group than in the placebo group, and there was significant difference between groups.
[0086] In the case of the ITT and PP analysis results on changes in skin acidity measured by pH, the acidity improvement effect was significant until the 6th week, but there was no significance between groups. On the other hand, in the 12th week, the acidity improved more significantly in the group that consumed lactic acid bacteria as the pH decreased, showing significance between groups.

[0089] Furthermore, skin acidity, a key indicator of skin health, can be determined by the total content of lactate and free amino acids (FAAs) produced in the epidermis. Accordingly, changes in the content of these components were also measured.
[0090] At weeks 0, 6, and 12, epidermal tissues were collected from the forehead and T-zone of the subjects using tape strips (22-㎜ D-SQUAME Tape, Cu Derm, USA). For lactic acid, the L-lactate component in the extract extracted by sonication with distilled water was measured at 565 nm using a kit (EnzyChrom™ L-lactate assay kit, Bioassay system, USA) according to the kit's instructions. For free amino acids, the extract extracted by sonication with a chloroform / methanol (2:1, v / v) solution on the tape was measured using a kit (L-amino acid quantitation colorimetric / fluorometric kit, Biovision Co., USA) according to the kit's instructions.
[0091] The results of ITT analysis on changes in lactic acid content and total free amino acid content of the epidermis measured according to the above method are shown in Table 6 below, and the results of PP analysis are shown in Table 7 below.
[0092] According to the results of the intention-to-treat (ITT) analysis on changes in epidermal lactate content, there was no significant difference between the placebo group and the probiotic group at both weeks 6 and 12. However, the results of the pre-treatment (PP) analysis showed that the lactate content in the probiotic group significantly increased at week 12, whereas the lactate content in the placebo group decreased, demonstrating a significant difference between the groups. Regarding total free amino acids in the epidermis, neither the ITT nor the PP analysis found significant differences between the groups at weeks 6 and 12.
[0093] Based on the PP analysis results of the changes in acidity and the content of acidity-related indicator components as described above, it was confirmed that the acidity improvement effect resulting from the intake of Lactobacillus plantarum CJLP55 was due to the increase in the content of lactic acid.

[0097] [2-5] Confirmation of changes in skin lipid content according to probiotic intake
[0098] The placebo group and the probiotic group consumed the food prepared according to Experimental Example 2-2 for 12 weeks according to the above consumption method, and then the lipid components of the skin were checked and their contents were determined. At weeks 0, 6, and 12, the subjects' sebum lipids were collected by attaching tape (22-mm D-SQUAME Tape, Cu Derm, USA) to the facial skin area. The collected tape was soaked in chloroform / methanol (2:1, v / v) and sonicated for 2 hours to extract the lipids. This was dried with nitrogen and dissolved again in chloroform solution, and neutral fat (TG, triglyceride), cholesterol esters (CE, cholesterol esters), free fatty acid (FFA, free fatty acid), cholesterol (Chol), and ceramide (Cer) were separated through HPTLC (developing conditions: 1st developing solution CHCl3 / methanol / water (10:2.5:0.25, v / v / v) up to 2 cm, 2nd developing solution CHCl3 / methanol / acetic acid (11.25:1.25:0.13, v / v / v) up to 5 cm, 3rd developing solution hexane / diethyl ether / acetone (2.5:10:0.63, v / v / v) up to 6 cm, 4th developing solution hexane / diethyl ether (12:0.73) up to 9.5 cm). Each separated fraction was scanned with a TLC Ⅲ scanner and quantitatively calculated using an external standard method, and the results of ITT and PP analysis are shown in Tables 8 and 9, respectively.
[0099] As a result, for total lipid content, both ITT and PP analyses showed a significant decrease in lipid content between groups in the probiotic intake group from week 6. Among them, for neutral fat, both ITT and PP analyses showed a significant decrease in neutral fat between groups in the probiotic intake group from week 6, and the effect was more significant at week 12. For cholesterol ester content, the decreasing effect in the probiotic intake group also showed a significant decrease between groups at week 12. There was no significant decrease in total free fatty acid content between groups, and the decrease in cholesterol content showed a significant decrease between groups in the probiotic intake group from week 6, and was more significant at week 12.
[0100] For ceramides, ceramides 1-7 were fractionated, and the ITT analysis showed no significant difference between groups. However, the PP analysis showed that the content of ceramide 2 (Cer2), a major ceramide, significantly increased in the probiotic intake group at week 12.

[0103] In view of the results of the above human application tests, it is expected that the intake of maltodextrin and powdered glucose included as excipients in the food consumed by the test subjects inhibits only the secretion of some lipids from epidermal cells and sebaceous glands only in the 6th week of intake, whereas the intake of Lactobacillus plantarum CJLP55 of the present application has the effect of reducing the total sebum content by inhibiting the content and secretion of lipids such as neutral fat, cholesterol, and cholesterol ester, and has the effect of maintaining skin moisture by selectively increasing the production of ceramide in epidermal cells, ultimately resulting in the effect of improving acne severity.
[0104]
[0105] [2-6] Confirmation of the effect of reducing harmful skin bacteria through lactic acid bacteria intake
[0106] In order to analyze the changes in the skin flora of harmful bacteria proliferating on the skin according to the ingestion of Lactobacillus plantarum CJLP55, a skin surface sample was collected by rubbing the same area on one cheek for 30 seconds and stored at -80℃. Each skin sample obtained at weeks 0 and 12 was subjected to next generation sequencing (NGS, MiSeq) for the 16S rRNA V3-V4 region by Macrogen, Inc., and the changes in the skin flora were analyzed through preprocessing and data analysis of the obtained sequence data.
[0107] Among the test subjects, 1 person in the placebo group and 3 people in the probiotic group could not create a DNA library, so the ITT analysis was conducted on 41 people in the placebo group and 39 people in the probiotic group, and the PP analysis was conducted on 38 people in the placebo group and 36 people in the probiotic group, and the results are shown in Table 10 below. Both the ITT and PP analysis results showed that the proportion of microorganisms in the Propionibacterium and Staphylococcus genera in the probiotic group was significantly different from the placebo group at week 12. In the placebo group, the Propionibacterium and Staphylococcus genera, known as harmful skin bacteria, significantly increased compared to week 0, whereas in the probiotic group, there was no significant change over 12 weeks, confirming that the proliferation of the harmful bacteria was suppressed.
[0108] In addition, species-level analysis of the sequences assigned to the genus Propionibacterium was performed, and an average of 94% of the sequences were confirmed to be P. acnes in both ITT and PP analyses (Fig. 2).

[0111] [2-7] Changes in skin roughness due to probiotic intake
[0112] The placebo group and the probiotic group were administered the food manufactured according to the above Experimental Example 2-2 according to the above intake method for 12 weeks, and then the amount of removed dead skin cells was measured to confirm the change in skin roughness. At weeks 0, 6, and 12, the stratum corneum was collected from the inner part of the arm (volar forearm) of the subjects using tape strips, and photographs were taken at a fixed position of a densitometer (SLB Myimager, Seoulin, Korea). The amount of removed dead skin cells was measured with the densitometer and quantified for comparison. The amount of dead skin cells for each week for each subject was calculated as the average of the measurements for five tape strips, and the results of the ITT analysis are presented in Table 11 below, and the results of the PP analysis are presented in Table 12 below.
[0113] In the ITT analysis results of skin roughness measured through digitization of the keratinocytes in each group, there was no significant difference between groups in the changes at 6 and 12 weeks, but in the PP analysis, it was confirmed that the skin roughness in the group taking probiotics was significantly reduced compared to the placebo group, and the improvement in roughness in the group taking probiotics was significant between groups from the 6th week, and the reduction effect was more significant at the 12th week.
[0114] As seen from the above results, the subjects who consumed Lactobacillus plantarum CJLP55 showed a decrease in keratin and a decrease in skin roughness, which means that the skin's moisturizing effect was improved by the consumption of the lactic acid bacteria.
