[0272] Example 1-1: Preparation of Compound of Formula I-1

[0273] The compound of formula I-1 is prepared by a process comprising the following steps:

[0274] (1) In a three-necked jacketed flask, 3,6-diaminopyrazine-2,5-diacid (Compound III) (10 g, 1 eq), D-serine benzyl ester hydrochloride (12.86 g, 1.1 eq) and L-serine benzyl ester hydrochloride (12.86 g, 1.1 eq) were added. DMF (35 times the volume of Compound III) was added and stirred evenly.

[0275] (2) After cooling to 0°C, DIPEA (14.34 g, 2.2 eq) was added. After 30 min, ethyl 2-oximecyanoacetate (15.77 g, 2.2 eq) was added. After 20 min, EDC*HCl (21.28 g, 2.2 eq) was added. The mixture was reacted for another 40 min, then warmed to room temperature and reacted for 18 h.

[0276] (3) Purified water (70 times the volume of compound III) was added, and the solid precipitated and then filtered.

[0277] (4) Through the chromatography column (Xinweier C184806C sand plate chromatography column, ), and the eluate was concentrated under reduced pressure at 33°C to obtain pure compound of formula II-1 (4.8 g).

[0278] (5) Add the compound of formula II-1 (2 g, 1 eq) to a reaction flask, followed by ethanol (32 ml, 16 volumes), formic acid-potassium hydroxide aqueous solution (HCOOH: 1.0 g, 6 eq; KOH: 0.4 g, 6 eq; purified water: 8 ml, 4 volumes), and Pd / C (0.2 g, 0.1 volumes). React at 60°C for 2 h. After completion of the reaction, monitor the reaction by TLC. Remove the Pd / C by filtration through celite while hot. Wash the filter cake with ethanol aqueous solution (36% H2O, v / v) until the filtrate is light yellow. Combine the filtrates. Add 1 mol / L hydrochloric acid (7 ml), stir for 15 min, and concentrate to dryness under reduced pressure.

[0279] (6) The obtained solid was slurried with 20 ml of acetonitrile for 2 h, filtered, and then slurried with water for 2 h. After filtration, the filter cake was dried under reduced pressure at 40°C for 12 h to obtain a solid compound of formula II-1 (0.79 g): the overall molar yield was 10% (based on the compound of formula III) and the purity was 98.39%.

[0336] Example 2-B: Preparation of Compounds of Formula I-2

[0337] The compound of formula I-2 is prepared by a process comprising the following steps:

[0338] (1) Compound III (10 g, 1 eq), D-serine benzyl ester hydrochloride (12.86 g, 1.1 eq) and D-serine ethyl ester hydrochloride (9.38 g, 1.1 eq) were added to a three-necked jacketed flask, and DMF (35 times the volume of compound III) was added and stirred evenly.

[0339] (2) After cooling to 0°C, DIPEA (14.34 g, 2.2 eq) was added.

[0340] (3) Purified water (70 times the volume of compound III) was added, and the solid precipitated and then filtered to obtain a crude product (10 g) of the compound of formula II-2.

[0341] Through the chromatography column (Hinwell C184806C sand plate chromatography column, ) or high pressure preparative chromatography (LAB-ZB-05, domestic Aonuo medium and high pressure equipment) to obtain pure compound of formula II-2.

[0342] (4) Add the compound of formula II-2 (5 g) to a high-pressure hydrogenation autoclave and dissolve it in 175 ml of ethanol (36% H2O, v / v). Then add Pd / C (0.5 g, 0.1 times the volume). After nitrogen displacement three times, the atmosphere was replaced with hydrogen three times (0.2 MPa) and the reaction was continued for 4 to 12 hours. After the reaction, the Pd / C was removed by filtration through diatomaceous earth, and the filtrate was spin-dried or concentrated to dryness under reduced pressure.

[0343] (5) The obtained crude product was first slurried with acetonitrile for 1 hour and then slurried with water for 1 hour. After filtering, the filter cake was dried under reduced pressure at 45°C to obtain the compound of formula I-2.
