Abstract
Description
BIOACTIVE COLLAGEN PEPTIDES AND VITAMIN C FORMULATIONSFIELD OF THE INVENTION
[0001] This disclosure relates to formulations containing bioactive collagen peptides and vitamin C as well as their use in treating osteoporosis.BACKGROUND OF THE INVENTION
[0002] According to the Mayo Clinic, “osteoporosis causes bones to become weak and brittle -so brittle that a fall or even mild stresses such as bending over or coughing can cause a fracture. Osteoporosis-related fractures most commonly occur in the hip, wrist or spine. ” The National Institute of Arthritis and Musculoskeletal and Skin Disease states that, “osteoporosis is a bone disease that develops when bone mineral density and bone mass decreases, or when the structure and strength of bone changes. This can lead to a decrease in bone strength that can increase the risk of fractures (broken bones) . ” The National Institute of Arthritis and Musculoskeletal and Skin Disease also calls osteoporosis a silent disease that does not have any symptoms and patients suffering from the disease may not be even aware of the disease until a bone fracture, such as e.g., most commonly hip bones, vertebrates in the spine, and wrist; while osteoporosis can occur at any age, the risk increases with age.
[0003] Currently available treatments for osteoporosis include the drug alendronate. In addition, supplements including vitamins and collagen peptides are used to promote healthy bones and reduce calcium loss. According to WebMD, “ [c] ollagen peptides are used for aging skin and osteoarthritis. They are also used for osteoporosis, brittle nails, muscle strength, and many other purposes, but there is no good scientific evidence to support most of these uses. ”
[0004] Alendronate has side effects including an increased risk of breaking a thigh bone. While supplements may provide some benefit, they are unable to treat, reduce, and / or prevent osteoporosis. What is needed is a more effective treatment for osteoporosis.SUMMARY OF THE INVENTION

[0026] The general description and the following detailed description are exemplary and explanatory only and are not restrictive of the invention, as defined in the appended claims.
[0027] This is disclosure is based on the discovery that the combination of bioactive collagen peptides and vitamin C (ascorbic acid) have a synergistic effect on the treatment of osteoporosis. Specifically, it has been discovered that a combination of bioactive collagen peptide and vitamin C is effective in restoring osteoporosis and / or preventing formation of osteoporotic bones. In particular (as shown in Example 1 below) , collagen peptide combined with vitamin C, can provide better efficacy than collagen peptide and vitamin C alone. Specifically, the combination showed on par efficacy with a drug (alendronate sodium) but in dietary solution.
[0028] Accordingly, the disclosure provides for formulations containing bioactive collagen peptides and vitamin C. These formulations support bone growth, restore osteoporosis, and prevent osteoporotic bone formation. These formulations can treat osteoporosis and showed on par efficacy with drug (alendronate sodium) , but in a dietary supplement, such as e.g., a solution.

[0077] Examples
[0078] Example 1: Impact of bioactive collagen peptides and vitamin C on osteoporosis
[0079] Existing dietary ingredients (vitamin C and collagen peptide) are believed to have some health benefits on collagen formation in our body, which can link to bone health. However, the direct health benefit on osteoporosis of the ingredient combination is unknown. Accordingly, the study in this Example investigated the protective effects of cattle bone-derived bioactive collagen peptides (BCPs) combined with vitamin C (VC) on dexamethasone-induced osteoporosis (OP) in zebrafish.
[0080] Methods
[0081] Anti-osteoporosis: dexamethasone-induced osteoporotic zebrafish (wildtype, AB line, 72 hours post-fertilization) were treated with different concentration of BCPs and / or vitamin C for forty-eight hours. BCPs were derived from cattle bone under a specific hydrolysis process with a signatured peptide pattern and average molecular weight of 5kDa. Vitamin C was extracted from acerola cherry. Alizarin red stained was performed at different time points within 48 hours. The fluorescence intensity of vertebrae was analyzed.
[0082] Prevention of osteoporosis: 24 hours post-fertilization or 48 hours post-fertilization zebrafish (wildtype, AB line) was pre-treated with BCPs and / or vitamin C for 48 hours or 24 hours respectively, followed by a co-treatment of dexamethasone. Alizarin red stained was performed 48 hours after co-treatment. Fluorescence intensity of vertebrae was analyzed. A control group (wildtype, AB line, without any treatment) , a model group (wildtype, AB line, treated with dexamethasone) and a group with alendronate sodium (5 μg / mL) as positive control were set up in both sets.
[0083] Results
[0084] Dexamethasone treatment significantly reduced the fluorescence intensity of zebrafish vertebrae, while alendronate sodium effectively restored dexamethasone-induced bone loss.
[0085] Anti-osteoporosis: compared with the control group, both BCPs and vitamin C treatment significantly increased the fluorescence intensity of vertebrae (p<0.05) . The protective effects were observed to be dose-dependent and better achieved when BCPs combined with vitamin C, suggesting a synergized effect of BCPs and vitamin C.
[0086] Prevention of osteoporosis: compared to unpretreated groups, pre-treatment of BCPs and vitamin C showed a trend (p=0.16) of recovering the fluorescence intensity of vertebrae in zebrafish treated with dexamethasone.
[0087] Specifically, it was observed that BCPs synergized with vitamin C to improve dexamethasone-induced osteoporosis in zebrafish (see FIG. 1A and FIG. 1B) . Compared with the control group, both BCPs and vitamin C treatment significantly increased the fluorescence intensity of vertebrae (p<0.05) . Furthermore, the protective effects were dose-dependent and better achieved when BCPs combined with vitamin C, suggesting a synergized effect of BCPs and vitamin C.
[0088] In addition, vitamin C accelerated BCPs’ protective effect on dexamethasone-induced bone loss in zebrafish (see FIG. 2A and FIG. 2B) . Protective effect on bone loss was observed in both BCPs &BCPs +VC group as early as 48 hours after dexamethasone treatment. Better protective effect on dexamethasone-induced bone loss was achieved in BCPs + VC group than in BCPs group at every timepoint. The BCPs +VC group recovered same content of boss loss earlier than BCPs group during treatment (BCPs + VC 12 hours vs. BCPs 24 hours; BCPs + VC 24 hours vs. BCPs 36 hours) .
[0089] Additionally, BCPs + vitamin C pre-treatment showed a trend to prevent bone loss in dexamethasone-treated zebrafish (see FIG. 3A and FIG. 3B) . Pre-treatment of BCPs and vitamin C showed a trend (p=0.16) of recovering the fluorescence intensity of vertebrae in zebrafish treated with dexamethasone. Furthermore, the treatment with the combination of BPCs and vitamin C was almost as effective as treatment with alendronate sodium.
[0090] The testing in this example demonstrated that bioactive collagen peptides synergized with vitamin C to improve dexamethasone-induced osteoporosis in zebrafish. Specifically, the combination of bioactive collagen peptides and vitamin C was verified to have superior efficacy that the single ingredients in dexamethasone-induced osteoporotic zebrafish. The observed effects were on par with the osteoporosis treatment drug alendronate, in restoring from osteoporosis and preventing formation of osteoporotic bones.
[0091] Example 2: Conversion of Tested Dosage to Dosage in Humans
[0092] In Example 1, various combinations of bioactive collagen peptides and vitamin C were evaluated in a zebrafish model. These dosages equate to the following dosages in a human.
