Abstract
Description
[0001] PERSONAL CARE COMPOSITION
[0002] Field of the Invention
[0003] The present invention relates to a personal care composition comprising moringa oil and coconut oil with a specific weight ratio. Surprisingly, it was found the expression of Heme Oxygenase 1 (HM0X1) gene was synergically upregulated within such ratio.
[0004] Background of the Invention
[0005] Human skin may provide physicochemical protection against environmental insults through its barrier function, mechanical strength and imperviousness to water. It is constantly exposed to harmful environments and extreme conditions, such as ultra-violet radiation, blue light, pollution etc. Such harmful environmental applications may cause excessive generation of reactive oxygen species (ROS) thereby resulting in an oxidative stress condition. Oxidative stress may affect our skin in many ways including signs of premature aging such as wrinkles, fine lines, uneven skin tone.
[0006] Heme Oxygenase 1 (coded by the HM0X1 gene) is known to demonstrate a protective role against oxidative stress. It is an enzyme that catalyzes the degradation of heme to biliverdin a precursor of a powerful antioxidant metabolite, bilirubin which has antioxidant and antiinflammatory properties. Heme Oxygenase 1 , primarily functioning in the catabolism of heme, is a very important cytoprotective enzyme that has antioxidant, anti-inflammatory and anti-apoptotic properties.
[0007] Stimulating HM0X1 gene would, in turn, stimulate the degradation of heme thereby provide antiaging, anti-oxidation and anti- inflammatory benefits. Human skin is also a place where inflammation, oxidative stress and cellular damage occurs regularly.
[0008] Therefore, the present inventors have recognized there is a need to develop a personal care composition which is capable of improving the expression of HM0X1 gene and developed a personal care composition comprising moringa oil and coconut oil, it was surprisingly found that the expression of HM0X1 gene is capable of significantly upregulated when the weight ratio of the moringa oil to the coconut oil is at least 1.4:1. [0050] Example 2
[0051] This example demonstrates the performance of upregulating the expression of HM0X1 gene by different personal care compositions.
[0052] ArchSkin living skin equivalent (LSE), is a 3D reconstructed human epidermis model with Chinese human keratinocytes through air-liquid interface culture in a defined serum free medium, supplied by Archgene (Shanghai, China). Squalene monohydroperoxide (SQOOH), a stress surrogate, was prepared in house, by subjecting squalene to solar irradiation in the presence of a sensitizer.
[0053] The samples were diluted in phosphate buffered saline (PBS) at volume ratio of 1 :1. The SQOOH was diluted using squalene to be 1 % by weight. 10 pL of diluted SQOOH was applied topically on the LSEs and left for 30 seconds. Then, the LSEs were topical treated by 10pL of the diluted samples for another 30 seconds and gently rinsed by 20mL PBS. After 24 hours, the tissues were collected for gene expression analysis.
[0054] The extracted RNA was quantified using Nanodrop spectrometer (Thermo Fisher Scientific, Waltham, MA, US) and reverse transcribed to generate the template cDNA using the Evo M-MLV RT Premix for qPCR (Accurate Biotechnology, Cat: AG11706) according to manufacturer’s protocol. Gene expression of HM0X1 gene was analyzed by polymerase chain reaction (PCR) using SYBR® Green Premix Pro Taq HS qPCR Kit (Accurate Biotechnology, AG11701). The actin beta (ACTS) gene was selected as the housekeeping gene and all data on relative expression of the target genes was normalized to ACTB gene. The fold changes in expression were calculated relative to the blank control (medium having no active). All tests were conducted at least three times and Table 1 shows the fold changes in gene expression of HM0X1.
[0055] The results were summarised in Table 2.
[0056] Table 2 a: significantly better than any one of sample A, B, or C (p<0.05) b: significantly better than sample 3 (p<0.05) c: significantly better than sample 1 (p<0.05)
[0057] It is evident from the data in Table 1 that samples 1 , 2 and 3 are capable of significantly elevating the expression of HM0X1 gene than that of samples A, B, or C. Sample 1 performs significantly better in elevating the expression of HM0X1 gene than sample 3 and sample 2 performs significantly better in elevating the expression of HM0X1 gene than both samples 1 and 3.
