Description

High-protein vegan nutritional composition based on soy proteinFIELD OF THE INVENTION

[0001] The present disclosure relates to heat sterilized liquid vegan nutritional compositions offering good mouthfeel and viscosity. The nutritional compositions herein provide high caloric densities of at least 1 ,4kcal / ml and high amounts of protein of at least 6.5g / 100ml in order to be used in therapy such as the prevention and treatment of nutrient deficiencies associated with malnutrition especially in patients for whom, e.g., proteins of animal origin cannot be used, may it be by personal choice or due to food allergies.BACKGROUND OF THE INVENTION

[0002] There is an increasing global demand for plant-based nutrition due to several aspects, e.g., environmental considerations like a reduction of the world-wide meat consumption and production as well as the need for lactose-free diets due to widespread incidence of lactose- intolerance. Personal preferences like animal-free diets, alternative protein sources, or health concerns are aspects of upcoming importance and play a major role in the consumer behavior as well.

[0003] Additionally, there is a general medical need for nutritional compositions high in protein and calories in a low volume of formula for malnourished people, such as compositions having a protein content of at least 6.5g / 100ml of the composition and caloric densities of at least 1.4kcal / ml.

[0004] High-protein, high caloric compositions are difficult to produce due to several technical problems, especially on industrial scale. Additionally, plant-based ingredients - especially plant-proteins - often have an unpleasant taste and develop unpleasant mouthfeel when used in high concentrations. Both make the consumption of the respective formula less attractive resulting in low appetite and a risk of consuming only insufficient amounts of nutrients.

[0005] Hence, following a plant-based diet can be especially challenging for patients with or at risk of malnutrition due to low protein intake and low intake of micronutrients.

[0006] A commercially available plant-based liquid nutritional composition provides 6g / 100ml protein and a caloric density of 1.5kcal / ml equaling 16EN% of protein. The protein component predominantly consists of pea protein with a smaller fraction of soy protein.

[0007] Pea protein however is often considered a nutritionally less preferred source of plant protein compared to soy protein. It has a lower protein digestibility-corrected amino acid score (PDCAAs) indicating a lower protein quality due to a lower digestibility and amino acid score. Malnourished patients have increased protein requirements. Hence, provision of high-qualityprotein is essential to support protein synthesis in their bodies. Furthermore, allergies to pea protein have been emerging and reported in several studies and reports. There is increasing evidence for the allergic risk caused by the exposure of consumers to pea protein due to emerging use of pea protein as vegan protein source in vegan diets. Therefore, in order to minimize the likelihood of potential allergic reactions, it is preferable to provide products with proteins from single plants as a source (e.g. comprising only soy or pea, instead of both).

[0008] Consequently, a need to be addressed by the present disclosure is the provision of a high-energy, high-protein vegan liquid enteral nutritional composition with acceptable taste and mouthfeel that contains a high-quality protein component and minimizes allergic potential.SUMMARY OF THE INVENTION

[0009] In a first aspect, the present disclosure relates to a heat sterilized liquid vegan nutritional composition according to claim 1. The nutritional composition is inter alia characterized by comprising a protein component consisting of at least 6.5g of soy protein per 100ml of the nutritional composition, and in that the nutritional composition comprises at least 13mg phosphorus per gram of protein.

[0010] The inventors found that such nutritional compositions offer excellent taste, mouthfeel, stability, and viscosity properties.

[0011] These properties render them particularly suitable for use in therapy such as prevention and treatment of nutrient deficiencies associated with malnutrition (according to a second aspect of the present disclosure.

[0012] The inventors found that the soy protein blend characterized by the amount of phosphorus per gram of protein influences viscosity, taste, and mouthfeel of the nutritional compositions herein.DESCRIPTION OF THE FIGURES

The mixture is homogenized at a pressure, e.g., of 100 / 50 bar (total pressure 150 bar), followed by a sterilization, e.g., at 138-140°C for 6-15 sec. Finally, a second homogenization step, e.g., at 80 / 20 bar (total pressure 100 bar) is applied.EXAMPLESSoy protein sources

[0109] SUPRO® 1500 IP and SUPRO® XT 34N IP. Both isolated soy proteins, obtained from Solae Europe. Characteristics, such as content of phosphorus, are listed in Table 2.METHODSViscosity

[0110] Determination of the shear viscosity by rotational rheometry.

[0111] Shear rheological measurements were performed at 20 °C , using a rotational rheometer (Haake RheoStress 1, Thermo Scientific GmbH, Germany) with double gap cylinder geometry (DG43). All the samples had a resting time in the measuring geometry prior to testing. The flow curves of the product were determined at shear rates between 0.1 and 500 s-1. The experimental data were fitted with the Oswald de Waele model. The power law index (n), the consistency index (K) were extracted and the viscosity at 50 s-1determined.

[0112] The person skilled in the art knows how to determine the shear viscosity in nutritional compositions of the present disclosure by rotational rheometry.Particle size distribution

[0113] Particle size distribution was determined by laser light scattering using a Mastersizer 3000 (Malvern). The methods are well established and known to the person skilled in the art. Results are provided in Volume% against particle size in pm (diameter).Sample preparation

[0114] Vegan liquid enteral nutritional compositions according to table 1 were prepared.Process description

[0115] Water was provided in a mixing vessel and heated to 60-70°C. The first fraction of the carbohydrate component (sucrose) and, optionally, the dietary fibres were added followed by the addition of soy protein. The obtained mixture was stirred for 5-10 min. and the remaining fraction of the carbohydrate component (glucose syrup) was added. The mixture of the lipid component with emulsifier (pre-heated to 50-80°C) was added and the obtained mixture stirred for 5-10 min. Minerals, vitamins and, optionally, additives were added followed by the addition of aroma. The pH was adjusted to 8.3 by adding a sodium hydroxide solution and the obtained mixture stirred for 5-15 min. The temperature of the mixing vessel was decreased to 15-20°C. The mixture was homogenized at a pressure of 100 / 50 bar (total pressure 150 bar), followed by a sterilization at 138-140°C for 6-15 sec. Finally, a second homogenization step at 80 / 20 bar (total pressure 100 bar) was applied.

[0116] Examples 1 , 2, 3, and 4 were prepared accordingly, using the same flavor. The nutrient profile only varied in terms of the composition of the soy protein component as indicated in table 1. Example 1 is a comparative example. In example 4, the amounts of macronutrients was adapted to provide an energy density of 2.0 kcal / ml.

[0117] An evaluation of the taste and mouthfeel of Examples 1 , 2 and 3 was undertaken by a test panel. The test was carried out under controlled conditions: The products were served at room temperature. Samples of each product were provided to panellists at the same time in random presentation. Water and white bread were served to allow for taste bud neutralisation in between testing samples. The panellists were required to identify the most preferred sample in taste preference and mouthfeel (e.g., pleasant, chalky, creamy, oily) in two evaluation cycles.

[0118] Results are displayed in Figure 1 (taste) and Figure 2 (mouthfeel). In terms of taste, most of the panellists (50%) selected Example 3 as highly acceptable, followed by Example 2 (33%) which was considered moderately acceptable, and Example 1 (17%). In terms of mouthfeel, Example 1 was often described as chalky, to viscous and having the least pleasant mouthfeel. Example 3 was chosen to have the most pleasant mouthfeel by almost 70% of the panellists.

[0119] The examples show that it is possible to prepare heat-sterilized, high-energy, high- protein, vegan, liquid enteral compositions with improved properties in terms of taste, mouthfeel, viscosity, and emulsion stability. These properties can be influenced by selecting the protein source.

[0120] Without wishing to be bound by theory, it is assumed that taste and I or mouthfeel in the compositions are influenced by emulsion properties such as viscosity and particle size distribution of the nutritional emulsions. This is supported by the empirical finding when comparing taste and mouthfeel panel test results for Examples 1 , 2 and 3 with particle size distributions and viscosity data.TABLESTable 1 - Examples*(-) very low acceptance, (- / +) low acceptance, (+) good acceptance, (++) excellent acceptance, see Fig. 1 and 2Table 2 - Soy protein examplesEMBODIMENTSNutritional composition1.
