Abstract
This invention relates to methods for producing immune cells expressing a therapeutic antigen receptor. An immune cell is provided that comprises a first expression cassette. The first expression cassette comprises a first coding sequence for a production TCR. The immune cells comprising a production TCR collectively form a production cell bank. The production cell bank may be stored and used as the starting material for producing immune cells expressing a therapeutic antigen receptor. A second expression cassette is then inserted into the first expression cassette. The second expression cassette comprises a second coding sequence for a therapeutic antigen receptor. Insertion of the second expression cassette introduces a frameshift mutation into the first coding sequence, such that the first and second coding sequences are in different reading frames and the immune cells express the therapeutic antigen receptor encoded by the second coding sequence and do not express the production TCR encoded by the first coding sequence. Methods, reagents for use in the methods and immune cells produced by the methods are provided.
