Abstract

A controlled-release drug-releasing microneedle with good biocompatibility and a preparation method therefor. The preparation method comprises: step S1, dissolving a sustained-release drug in a molding material solution to obtain a sustained-release needle body solution; and dissolving an instantaneous drug in a molding material solution to obtain an instantaneous drug layer solution, wherein the molding material solution comprises: 16 wt%-19 wt% of polyvinyl alcohol solution; step S2, providing a microneedle mold, wherein the microneedle mold comprises needle body cavities distributed in an array and backing cavities in communication with opening ends of the needle body cavities; and adding the sustained-release needle body solution into the needle body cavities, such that the sustained-release needle body solution fills the needle body cavities, and then adding a backing layer solution into the backing cavities to obtain a drug-releasing microneedle precursor, wherein the backing layer solution comprises: 20 wt%-22 wt% of polyvinyl alcohol solution; and step S3, repeatedly freezing and thawing the drug-releasing microneedle precursor m times, wherein m > 1; and removing the microneedle mold, applying the instantaneous drug layer solution on a surface of the sustained-release needle body, and performing freeze-crosslinking to obtain a controlled-release drug-releasing microneedle with good biocompatibility. The controlled-release drug-releasing microneedle with good biocompatibility can flexibly contain different drugs according to flexible clinical drug requirements, and enable the drugs to be released in different sequences and rates, thereby solving the technical problem of a single drug delivery mode in existing microneedles.
