SUBSTITUTED TETRAHYDROISOQUINOLINE DERIVATIVES AND USES THEREOFFIELD OF THE DISCLOSURE

[0001] The present disclosure generally relates to substituted tetrahydroisoquinoline derivatives which are positive allosteric modulators of human D1 dopamine receptor, pharmaceutical compositions comprising these compounds as well as uses of these compounds or the pharmaceutical compositions in the treatment of diseases or medical conditions via modulation of human D1 dopamine receptor. BACKGROUND OF THE DISCLOSURE

[0002] The dopamine D1 receptor plays an important role in motor activity, reward, and cognition. D1 receptors are expressed in defined neuronal classes and specific brain regions to mediate these functions. Due to their role in diverse physiological and pathobiological processes, D1 receptors have been implicated in a variety of disorders including cognitive and negative symptoms in schizophrenia, cognitive impairment related to classical antipsychotic therapy, impulsivity, attention-deficit / hyperactivity disorder (ADHD) , Parkinson's disease and other movement disorders (such as, Huntington's disease, dystonia, etc. ) , dementia with Lewy bodies, Alzheimer's disease, age-related cognitive decline, mild cognitive impairment (MCI) , drug addiction, sleep disorders, apathy. It has proven difficult to develop orally-bioavailable small molecules selectively targeting D1 receptors.

[0003] Efforts to develop D1 agonists have been mixed due to poor drug-like properties, tachyphylaxis, and inverted U-shaped dose-response curves. Recently, positive allosteric modulators (PAMs) for the dopamine D1 receptor were discovered and initial pharmacological profiling has suggested that several of the above issues could be addressed with this mechanism. (Svensson et al., Positive allosteric modulators of the dopamine D1 receptor: A new mechanism for the treatment of neuropsychiatric disorders, Adv Pharmacol. 2019: 86: 273-305. )

[0004] Accordingly, there is a need in the art to develop novel D1 positive allosteric modulators (D1 PAMs) . SUMMARY OF THE DISCLOSURE

[0005] The present disclosure provides compounds which are D1 positive allosteric modulators, the pharmaceutical compositions comprising these compounds and the use of such compounds or pharmaceutical compositions for treatment of a disease or medical condition via activation of D1 receptors.

[0006] In one aspect, the present disclosure provides a compound having a formula (I) : or a pharmaceutically acceptable salt thereof, wherein R1 is selected from the group consisting of hydrogen, halogen, cyano, alkyl, alkoxyl, haloalkyl, cycloalkyl, and heterocyclyl; W is C (Rb) or N; Rb is selected from the group consisting of hydrogen, deuterium, halogen, amino, cyano, hydroxyl, alkyl, alkoxyl and haloalkyl, ; Q is C, O, or N; Ring A is aryl or heteroaryl; each RA is independently selected from the group consisting of halogen, cyano, alkyl, alkenyl, alkynyl, heteroalkyl, alkoxyl, haloalkyl, cycloalkyl and heterocyclyl, wherein the alkyl, alkenyl, alkynyl, heteroalkyl, alkoxyl, haloalkyl, cycloalkyl and heterocyclyl are independently optionally substituted with one or more Rc; n is 1, 2, 3 or 4; provided that: when Q is O, then R2 and R3 are absent; when Q is N, then R2 is absent, and R3 is selected from the group consisting of hydrogen, alkyl, heteroalkyl, haloalkyl, hydroxyalkyl, -C (O) -alkyl, -S (O) -alkyl, -S (O) 2-alkyl and cycloalkyl, wherein the alkyl, heteroalkyl, haloalkyl, hydroxyalkyl, -C (O) -alkyl, -S (O) -alkyl, -S (O) 2-alkyl and cycloalkyl are optionally substituted with one or more Rd; and when Q is C, then each of R2 and R3 is independently selected from the group consisting of hydrogen, deuterium, halogen, cyano, hydroxyl, alkyl, heteroalkyl, alkoxyl, alkylthio, haloalkyl, hydroxyalkyl, and cycloalkyl, wherein the alkyl, heteroalkyl, alkoxyl, alkylthio, haloalkyl, hydroxyalkyl, heteroalkyl and cycloalkyl are optionally substituted with one or more Rd; or R2 and R3 taken together with the carbon to which they are attached form a cycloalkyl or heterocyclyl, wherein the cycloalkyl and heterocyclyl are optionally substituted with one or more groups independently selected from the group consisting of deuterium, halogen, cyano, hydroxyl, alkyl, heteroalkyl, alkoxyl, alkylthio, haloalkyl and hydroxyalkyl; and Rc and Rd at each occurrence is independently selected from the group consisting of deuterium, halogen, cyano, hydroxyl, alkyl, alkoxyl, haloalkyl, cycloalkyl and heterocyclyl.

[0010] In a further aspect, the present disclosure provides a method for modulating (such as, activating) D1 receptors in a subject in need thereof, comprising administering an effective amount of a compound of the present disclosure or a pharmaceutically acceptable salt thereof or the pharmaceutical composition of the present disclosure to a subject in need thereof.

[0319] Human dopamine D1 receptor expressing stable cell line was generated with HEK293 cells. The cells were cultured in complete growth media containing 90%DMEM, 10%fetal bovine serum (FBS) , 1 × Penicillin-Streptomycin (PS) , and 600 μg / mL G418. Then, the cells were cryopreserved in freezing media containing 90%FBS and 10%dimethyl sulfoxide (DMSO) .

[0320] Test compounds were solubilized in 100%DMSO. A 10-point intermediate dilution series using 3-fold dilutions were created in 100%DMSO by liquid handler, as 50 nL per well in 384-well assay plates. To the prepared assay plates with serially diluted compounds, 50 nL of 400 × EC20 concentration of dopamine were added to wells containing test compounds. Prior to compound testing, a concentration response curve was run for dopamine to determine the EC20 concentration.
