Abstract

The present disclosure provides a method of treating cytokine release syndrome (CRS) associated with CAR T-cell therapy in a subject. The method comprises administering to the subject a pharmaceutical composition comprising an effective amount of CTO1681 for prophylactically inhibiting the CRS, and subsequently administering to the subject a population of CAR T-cells. The effective amount of CTO1681 is effective to reduce a severity of one or more symptoms of CRS. The pharmaceutical composition may inhibit levels of one or more cytokines upon administration of the CAR T-cells in the subject. The method may further comprise administering a second pharmaceutical composition comprising a corticosteroid, tocilizumab, or an IL-6 receptor blocker.
