AI Technical Summary

Technical Problem

Existing treatments for oral mucositis, a common adverse event from antibody-drug conjugates like datopotamab deruxtecan, are inadequate in preventing or reducing mouth sores, and there is a need for more effective management strategies.

Method used

A mouthwash containing a 'naked' anti-TROP2 antibody or its antigen-binding fragment, potentially combined with a pharmaceutically acceptable excipient, is used to penetrate the oral mucosa and occupy TROP2 binding sites, reducing the binding of antibody-drug conjugates and mitigating oral mucositis symptoms.

Benefits of technology

The mouthwash effectively reduces the severity of oral mucositis by minimizing the binding of antibody-drug conjugates to healthy tissues, thereby alleviating symptoms and potentially preventing their onset.

Abstract

The invention provides the use of a "naked" antibody targeting TROP2 or a TROP2 antigen-binding fragment thereof in a mouth wash (oral rinse) to mitigate the on-target toxicity of TROP2 antibody-drug conjugates, which is oral mucositis (mouth sores). The oral delivery of a "naked" anti-TROP2 antibody in an appropriate vehicle penetrates the mucosa. The "naked" antibody occupies a sufficient proportion of the TROP2 binding sites such that the antibody-drug conjugate does not bind or has reduced binding with a concomitant reduction in symptoms.
