Immobilized hemeprotein scavengers for the treatment of carbon monoxide poisoning

Immobilizing recombinant hemeproteins on agarose resin in dialysis cartridges addresses the limitations of intravenous treatments by safely and effectively removing carbon monoxide from the blood, reducing toxicity risks and enhancing treatment efficacy.

Abstract

Compositions that include recombinant globin proteins, such as human recombinant neuroglobin or cytoglobin or recombinant regulator of carbon monoxide metabolism (RcoM) protein, that bind carbon monoxide with high affinity are described. The recombinant hemeproteins are immobilized on a solid support, such as an agarose resin. The recombinant hemeproteins can be cross-linked to the agarose resin using N-hydroxysuccinimide (NHS). The immobilized hemeproteins can be incorporated into dialysis cartridges and extracorporeal systems to filter CO from the blood of a subject with CO poisoning. Non-immobilized hemeproteins for use in a standard dialysis system are also described.
