Description

METAL-CENTERED ANNULENES, AND THEIR PREPARATION AND APPLICATIONSTECHNICAL FIELDThis application relates to the fields of organic chemistry, coordination chemistry, organometallic chemistry, and aromatic chemistry, specifically relates to a complex in which a metal atom is located at the center of an annulene plane (metal-centered annulene) , and in particular, to a structure, a preparation method, and applications of an osmium

[0015] annulene complex. The complex includes five novel frameworks:

[55753] osmium

[0015] annulene,

[55735] osmium

[0015] annulene,

[55663] osmium

[0015] annulene,

[55636] osmium

[0015] annulene, and

[55555] osmium

[0015] annulene. In each of the complexes, the five digits in one pair of square brackets represent the number of atoms in each small ring in the complex, counted clockwise from the five-membered ring at the lower right corner; and the two digits in the other pair of square brackets represent the number of carbon atoms in the annulene.BACKGROUNDImines are of high commercial value. In the synthesis of many pharmaceuticals, fine chemicals, and bioactive substances, imine derivatives serve as important intermediates. Further, due to the presence of a C=N double bond, imines are widely used in various organic transformations such as reduction, addition, cyclization, and nitrogen heterocyclization reactions (ACS Catal. 2015, 5, 5851–5876) . Conventionally, imines are prepared through the condensation of amines with carbonyl compounds, especially unstable aldehydes. However, this preparation method typically requires the addition of a dehydrating agent and a Lewis acid catalyst, resulting in a large amount of waste residues (Org. Lett. 2015, 17, 2442-2445) . Therefore, from the perspective of green chemistry, the development of a novel method for the preparation of imines is crucial.Preparing imines by photocatalyzing the oxidative coupling of two amine molecules is a relatively green method because the method generally does not require additional additives other than a photocatalyst and an oxidant, thereby resulting in just a relatively small amount of waste residues. In particular, when oxygen in air serves as an oxidant, the reaction can be carried out in air without the need for additional oxidants, giving rise to no more byproducts than water, and making this method economical and environmentally friendly (ACS Catal. 2015, 5, 5851-5876) . The design and preparation of the catalysts is crucial for implementing such reactions. Therefore, the synthesis of mild and efficient catalysts is a major goal of scientists.Metalloannulene complexes, such as ferrocene, half-sandwich complexes, and diphenylchromium, are extremely important catalysts, and are widely used in various fields ranging from catalysis to materials science and biomedicine. Research on the coordination patterns of metalloannulene complexes has even revolutionized the bonding theory and significantly advanced the development of coordination chemistry. Nevertheless, in all currently reported metalloannulene complexes, the metal atom is located outside the annulene ring. Complexes in which the metal atom is located within the annulene plane have not yet been reported.SUMMARYIn view of the problems in the prior art, an objective of this application is to provide a novel osmium

[0015] annulene complex in which metal atoms are located at the center of the annulene plane, and their preparation methods and applications. As a type of metalloannulene complexes of a novel structure, the complexes are expected to embrace an application prospect as promising as the complexes such as ferrocene in which the metal atoms are located outside an annulene ring. Further, this application pioneers a feasible method for preparing such complexes. Most reactions involved in the method occur at room temperature, and the method is simple to operate. Considering the importance of oxidative coupling of amines in preparing imines, this application applies five novel types of osmium

[0015] annulene complexes in which the metal atoms are located at the center of an annulene plane to catalysis of the oxidative coupling, and achieves excellent results.To achieve the above objective, this application puts forward the following technical solutions:According to a first aspect, this application provides a novel complex in which metal atoms are located at a center of an annulene plane. The complex is

[55753] osmium

[0015] annulene 1aStep 1) Synthesis of a compound P2aA complex P1a is synthesized according to the existing publication (Nat. Commun. 2024, 14, 4378) . In an argon atmosphere, the complex P1a (200 mg, 0.16 mmol) and LiAlH4 (25 mg, 0.66 mmol) is successively added into a dichloromethane / diethyl ether (5 mL, volume ratio (V / V) =1 / 1) mixed solvent at room temperature. The mixture is stirred at room temperature for 5 minutes. Subsequently, the mixture is filtered in an argon atmosphere, and the solvent is removed from the filtrate under reduced pressure. The remaining solid is dissolved in methanol (5 mL) , and then a methanol (2 mL) solution ofNaBPh4 (140 mg, 0.41 mmol) is added into the solution. After 1 minute, the mixture is filtered. The solid precipitate is washed with methanol (3 mL×3) and then dissolved in dichloromethane (3 mL) . n-Hexane (30 mL) is added dropwise into the solution. The precipitate is collected by filtration, washed with n-hexane (5 mL×3) , and dried to derive a green solid compound P2a.Step 2) Synthesis of a compound P3aThe resulting P2a is dissolved in dichloromethane (5 mL) , and then p-toluenesulfonyl chloride (153 mg, 0.8 mmol) is added into the solution. After reacting at 35℃ for 12 hours, the solution is filtered through diatomaceous earth. The filtrate is vacuum-concentrated to approximately 2 mL, and then loaded into a 200-to 300-mesh silica gel column to undergo column chromatography separation. A green solid compound P3a (105 mg,at a yield rate of40%) is derived by using dichloromethane as an eluent. The structure of the compound P3a is confirmed by 1H, 31P {1H} , 13C {1H} nuclear magnetic resonance (NMR) and high-resolution mass spectrometry (HRMS) . The specific NMR and HRMS characterization data of the compound are as follows:1H NMR (600 MHz, CD2Cl2, ppm) δ: 7.62 (br, 1H, C5H) , 7.49 (d, J (HH) =4.1 Hz, 1H, C6H) , 7.24 (d, J (HH) =8.2 Hz, 1H, C8H) , 7.18 (br, 1H, C3H) , 6.56 (t, J (HH) =8.2 Hz, 1H, C9H) , 5.85 (d, J (HH) =8.2 Hz, 1H, C10H) , 2.39 (s, 3H, CH3) and 7.81-6.70 (69H, other aromatic protons) .31P {1H} NMR (243 MHz, CD2Cl2, ppm) δ: 6.19 (t, J (PP) =6.8 Hz, CPPh3) , -5.02 (d, J (PP) =6.8 Hz, OsPPh3) .13C {1H} NMR (151 MHz, CD2Cl2, plus 13C-DEPT 135, 1H-13C HSQC and 1H-13C HMBC, ppm) δ: 323.5 (td, J (PC) =15.7 Hz, J (PC) =10.9 Hz, C1) , 224.5 (t, J (PC) =10.9 Hz, C7) , 199.8 (t, J (PC) =12.3 Hz, C11) , 191.1 (d, J (PC) =29.2 Hz, C4) , 160.6 (t, J (PC) =4.9 Hz, C6) , 160.0 (s, C5) , 144.1 (d, J (PC) =17.5 Hz, C3) , 139.3 (s, C9) , 136.9 (s, C8) , 133.8 (br, C10) , 128.3 (td, J (PC) =89.8 Hz, J (PC) =6.0 Hz, C2) , 21.8 (s, CH3) and 143.6-121.4 (other aromatic carbon atoms) .HRMS (ESI) : (m / z) calcd for P3a [C72H58O2OsP3S] +1271.2980, found 1271.2957.Step 3) Synthesis of a compound P4aIn an argon atmosphere, P3a (200 mg, 0.12 mmol) and lithium trimethylsilylethynyl (50 mg, 0.48 mmol) are dissolved in tetrahydrofuran (10 mL) , and reacted at room temperature for 10 minutes. The solution is vacuum-dried, and the residue is loaded into a 200-to 300-mesh silica gel column to undergo column chromatography separation (using dichloromethane as an eluent) to derive a red solid compound P4a (115.6 mg,at a yield rate of60%) . The structure of the compound P4a is confirmed by 1H, 31P {1H} , 13C {1H} NMR and HRMS. The specific NMR and HRMS characterization data of the compound are as follows:1H NMR (600 MHz, CD2Cl2, ppm) δ: 7.46 (d, J (HH) =4.1 Hz, 1H, C5H) , 7.30 (d, J (HH) =4.1 Hz, 1H, C6H) , 7.22 (br, 1H, C8H) , 6.94 (br, 1H, C3H) , 5.57 (s, 1H, C10H) , 2.30 (s, 3H, C13H) , 1.69 (s, 3H, C12H) , and 7.71-6.77 (69H, other aromatic protons) .31P {1H} NMR (243 MHz, CD2Cl2, ppm) δ: 5.80 (t, J (PP) =7.9 Hz, CPPh3) , -3.65 (d, J (PP) =7.9 Hz, OsPPh3) .13C {1H} NMR (151 MHz, CD2Cl2, plus 13C-DEPT 135, 1H-13C HSQC and 1H-13C HMBC, ppm) δ: 323.3 (td, J (PC) =16.4, J (PC) =11.2 Hz, C1) , 226.2 (t, J (PC) =11.1 Hz, C7) , 200.3 (t, J (PC) =12.1 Hz, C11) , 188.1 (d, J (PC) =30.0 Hz, C4) , 164.6-163.6 (BPh4, αC) , 159.6 (s, C5) , 159.7 (s, C6) , 151.4 (t, J (PC) =3.1 Hz, C9) , 142.3 (m, C3) , 137.3 (s, C10) , 135.7 (s, C8) , 121.4 (d, J (PC) =90.6 Hz, C2) , 26.1 (s, C12) , 21.3 (s, C13) and 143.2-121.4 (other aromatic carbon atoms) .HRMS (ESI) : (m / z) calcd for P4a [C73H60O2OsP3S] +1285.3137, found 1285.3129.Step 4) Synthesis of
