[0001] The present application generally relates to a cell culture additive or supplement that may comprise glycine, a selenite, and optionally histidine and lysine. The present application also relates to the use of the cell culture additive or supplement in culturing a mammalian cell that over-expresses a recombinant protein (e.g., an antibody) , for reducing the level of acidic species of the recombinant protein.BACKGROUND OF THE INVENTION

[0002] Among the biologic drugs under development and on the market, most are monoclonal antibodies (mAbs) . Antibodies are commonly produced in CHO cells, due to high production level, easy culture and scale-up, and correct antibody folding with desired post-translational modifications. The post-translational modifications may play important roles in antibodies’ characteristics. For example, sialic acid modification may prolong the circulatory half-life of antibodies by masking the terminal galactose that would otherwise be recognized by the hepatic asialoglycoprotein receptors (Sean Chia, et al., (2023) , Enhancing pharmacokinetic and pharmacodynamic properties of recombinant therapeutic proteins by manipulation of sialic acid content, Biomedicine &Pharmacotherapy, 163: 114757) .

[0003] These post-translational modifications, along with protein degradations, may cause antibody heterogeneity, including charge heterogeneity. When analyzed by ion-exchange chromatography (IEX) or isoelectric focusing (IEF) -based methods, the antibodies may be generally divided into three groups, the main peak or constituents, the acidic species or variants that elute earlier than the main peak from CEX or later than the main peak from AEX or alternatively with lower isoelectric point (pI) than the major constituents or species in IEF, and basic species or variants that elute later than the main peak from CEX or earlier than the main peak from AEX or alternatively with higher isoelectric point (pI) than the major constituents or species in IEF. The charge variants may result in different efficacy and pharmacokinetics outcomes. Particularly, acidic charge variants may negatively affect the therapeutic efficacy of antibodies (Chung S, et al., (2019) Modulating cell culture oxidative stress reduces protein glycation and acidic charge variant formation. MAbs. 11 (1) : 205-216) . Asparagine (Asn) deamidation, sialylation, and glycation are the major contributors to the formation of acidic variants, and cysteine-related modifications such as free cysteine, trisulfide bonds, cysteination, and glutathioneylation also cause the formation of acid variants. For example, the Asn deamidation in the CDR region may reduce the antibody binding ability of the antibodies, and the free cysteines in the antibodies may induce antibody aggregation leading to an increase in the immunogenicity, or alter antigen-binding activity (Beck A, et al., (2022) Risk-Based Control Strategies of Recombinant Monoclonal Antibody Charge Variants. Antibodies (Basel) . 11 (4) : 73) . Therefore, controlling acidic charge variants is critical for a bioprocess, especially an industrial bioprocess, of antibodies.

[0004] However, controlling charge heterogeneity remains a major challenge for manufacturing of biological therapeutics. The strategies may include control of raw material, manufacturing process, and the downstream processing. The cell culture is thought to be the most impactful stage on charge heterogeneity, due to the long duration, complicated media, and enzymes within or outside of the host cells (Beck A, et al., (2022) supra) . The cell culture operating parameters, including pH and temperature may be optimized to control or lower the acidic charge species levels. Studies have shown low cell culture temperature and low pH are effective in reduction of acidic charge variants. However, a low culture temperature may decrease the enzyme activity in host cells and affect the cell growth and metabolism and thus the antibody production level, while pH downshift may negatively affect cell metabolism and protein production. Alternatively, cell culture medium may be optimized for acidic charge variant reduction, including adjusting concentration of glucose, trace metals (e.g., iron, copper) or antioxidants. Higher sugar level was reported to correlate to higher acidic charge species level, and Fe2+ can induce the formation of reactive oxygen species (ROS) and thus caused the increase of acidic variants (Sissolak B, et al., (2019) Impact of mammalian cell culture conditions on monoclonal antibody charge heterogeneity: an accessory monitoring tool for process development. J Ind Microbiol Biotechnol. 46 (8) : 1167-1178; Gangwar, et al., (2022) Achieving charge variant profile of innovator molecule during development of monoclonal antibody based biosimilars –Use of media components. Biochemical Engineering Journal. 182: 1-7) . Cell culture media supplementation of bioflavonoids such as rutin and gallic acid as antioxidants reduced the percent of acidic variants (Hossler, et al., (2015) Cell Culture Media Supplementation of Bioflavonoids for the Targeted Reduction of Acidic Species Charge Variants on Recombinant Therapeutic Proteins. Biotechnology progress. 31 (4) : 1039-52) . The cells cultured in a medium containing 1 g / L or more arginine (Arg) , histidine (His) , lysine (Lys) or their combination produced antibodies with lower percentage of acidic variants (US 9, 683, 033B2) .

[0006] The inventors of the disclosure, have designed a cell culture additive or supplement, that simply contains glycine, a selenite, and optionally histidine and lysine. When added to CHO fed-batch culture in the early exponential growth phase (e.g., Day 3 of cultivation) , the cell culture additive or supplement surprisingly reduced the level of acidic species or variants in the antibodies as produced by the CHO cells, without affecting the main species level or even increasing the main species level to some extent, no matter what culture media were used in the cell culture. The cell culture additive or supplement containing histidine and lysine enhanced cell viability in the late culture stage.

[0007] Glycine is a common component in the cell culture medium, but there is little report on its role in reducing acidic variants of the protein as produced by the host cells. Further, the glycine in the cell culture medium was said to increase ammonium production and thus usually used in a low amount (Zizhuo Xing et al., (2011) Optimizing amino acid composition of CHO cell culture media for a fusion protein production. Process Biochemistry. 46: 1423-1429) . However, the introduction of additional glycine to the cell culture in the early exponential growth phase (e.g., Day 3 of the fed-batch culture) in the present application, did not adversely affect host cell growth and metabolism, and surprisingly reduced the acidic variants of the antibody as produced by the cultured cells.

[0008] Antioxidants such as bioflavonoids, when supplemented in the feed medium and added to the cells for several times during the culture, can reduce acidic charge variants of the protein as produced by the host cells (Hossler, et al., (2015) supra; Chung S, et al., (2019) ) . In the present application, a selenite, as an antioxidant, was introduced to the cell culture only once on Day 3 of the fed-batch culture, but not during the whole culture, successfully reduced the acidic variants of the antibody produced by the cultured cells.

[0009] Histidine and lysine, when added respectively in the cell culture at relatively high amounts, can reduce acidic variants of the antibody as produced by the cultured cells. However, the high histidine or lysine levels in the cell culture medium may cause tremendous cell viability decrease in the late culture stage (US 9,683,033B2) . In the present application, the addition of histidine and lysine alone showed mild effect on acidic variant reduction, but maintained the cell viability at a relatively high level in the late culture stage. When used together with glycine and the selenite, histidine and lysine further decreased the acidic variant level.

[0010] Therefore, in a first aspect, the disclosure provides a cell culture additive composition that may comprise or consist of glycine and a selenite.

/ L and about 2.89×10-7–2.89×10-6mol / L, respectively, preferably about 3.33×10-3-6.66×10-3mol / L and about 7.2×10-7-1.45×10-6mol / L, respectively, on 1st, 2nd or 3rd day of the batch culture or the fed-batch culture.10.The method of claim 9, comprising adding to the cell culture medium the glycine and the selenite on the 3rd day of the batch culture or the fed-batch culture.11.The method of claim 10, comprising adding to the cell culture medium on 3rd day of the batch culture or the fed-batch culture the glycine and the selenite at final concentrations of about 6.66×10-3 mol / L and about 1.45×10-6 mol /
