DescriptionSORTING DEVICE AND CORRESPONDING SORTING METHOD Technical Field

[0001] The invention is in the field of the handling and sorting of sensitive biological entities, essentially multicellular entities, such as cell aggregates, cell colonies, organoids, spheroids, zebrafish eggs, zebrafish larvae, c. elegans, tissue sections, biopsy tissue, assembloids or others, herein generally named by "biological entities" in the context of the present disclosure.

[0003] Organoids are three-dimensional structures grown from stem cells that mimic the structure and function of organs. They are typically cultivated in laboratory settings under controlled conditions, allowing researchers to study aspects of organ development, disease progression, and drug responses in a more accurate and ethically acceptable manner compared to traditional animal models or cell cultures. Those biological entities represent a powerful tool in biomedical research with diverse applications spanning from basic science to clinical translation. The size of an organoid can range from very small structures resembling a few hundred micrometers in diameter to larger, more complex structures that can be several millimeters in size.

More particularly, this sorting device comprises an image acquiring system arranged to identify and locate biological entities of interest. Once a mapping of the sample contained in a source vessel is done, the source vessel is cautiously moved with high precision, along the xy plane, to bring a biological entity to be selectively picked up in a suction position, where it can be picked up.

[0007] In general, the handling of such biological entities with sizes larger than 1 mm (up to 5 mm) is difficult because of their tendency to sediment and their fragility to mechanical stress. One known problem is that in some circumstances, especially when working with cultures in smaller wells, the biological entities may lie close to the recipient walls, and it becomes mechanically impossible to reach them with the pipette tip.

[0008] These problematics are illustrated in Fig. 1 which is a photography of a source vessel 100 containing a sample including a plurality of organoids. One can clearly see from Fig. 1 that some of them may be too close to the wall of the source vessel to reach them with a standard pipette tip. The relevant diameters of the pipette tip are also represented at scale in Fig. 1, where 101 is the tip inner diameter, 102 is the outer diameter at the bottom of the tip and 103 is the diameter at the top of the tip.

[0009] The automated cell sorting devices of the prior art do not address this problem and, consequently, the need still exists to provide a solution to be able to selectively pick up organoids as other types of sensitive biological entities from a sample with reduced risks of damages caused to one or several of the biological entities included in this sample.Disclosure of the invention

[0010] An aim of the present invention is to propose a sorting device making it possible to selectively pick up biological entities from a sample with reduced risks of damages that could be caused to one or several of the biological entities included in this sample. Another aim of the present invention is to propose a method taking advantage of the above device for the selective picking up of at least one biological entity from a source vessel containing a sample including a plurality of biological entities, to be able to selectively dispense this picked up biological entity into a destination vessel, in a precise, reliable and non-damaging manner.

Indeed, the Applicant noticed that implementation of a stirring in the form of looped trajectories with respect to the frame of the sorting device, with adapted predefined shapes and speed parameters, provide relative movements of the plurality of biological entities between each other and with respect to the source vessel, which may solve the above-mentioned problem.

[0027] The sorting device 1 is an inspection and sorting device for sorting sensitive biological entities such as organoids, advantageously by selective picking up of at least one organoid from a source vessel 2, containing a sample including a plurality of organoids and located in a picking station 4, to be able to selectively dispense this organoid into a destination or target vessel 6 located in a delivery station 8. As already mentioned, in the context of the present disclosure, the biological entities of interest are more generally multicellular entities, such as cell aggregates, cell colonies, organoids, spheroids, zebrafish eggs, zebrafish larvae, c. elegans, tissue sections, biopsy tissue, assembloids or others. However, for the sake of simplicity, the remainder of the detailed description will be focusing on organoids in an illustrative and non-limiting manner.

Then, the additional camera 26 might follow the pipetting unit 14 to the delivery station 8, to be able to check that the delivery of a given organoid in a destination vessel 6 is properly carried out. Advantageously, one of the cameras 24, 26 or an additional part of the identification and localization system may further comprise a microscope system.

Claims

Claims1. Sorting device (1) comprising a picking station (4) and an identification and localization system, adapted to allow selective picking up of at least one biological entity from a source vessel (2) containing a sample including a plurality of biological entities, to be able to selectively dispense said at least one biological entity into a destination vessel (6), the sorting device (1) including a frame (10) on which a vessel support (30) for bearing at least one source vessel (2) is mounted in a movable manner, as well as a control unit and a driving organ adapted to control the movements of said vessel support (30) with respect to said frame (10), characterized in that said control unit and said driving organ are arranged so as to drive said vessel support (30) along successive looped trajectories with respect to said frame (10), inside a xy plane, in response to a corresponding manual request by a user or to a corresponding automatic request by said identification and localization system, andin that said control unit and said driving organ are arranged so as to provide said looped trajectories with predefined shapes and speed parameters which are adapted to stir the sample and provide relative movements of the plurality of biological entities between each other and with respect to the source vessel (2) so as to be able to substantially gather the plurality of biological entities inside a central region of the source vessel (2), in the xy plane.

2. Sorting device (1) according to claim 1, characterized in that said control unit and said driving organ are arranged so as to provide said vessel support (30) with an acceleration having a radial component of at least 0.1 m / s2.

Sorting device (1) according to claim 1 or 2, characterized in that said control unit and said driving organ are arranged so as to drive said vessel support (30) along successive looped trajectories with a rotation speed of 120 rpm or less, so as to be able to substantially gather the plurality of biological entities inside a central region of the source vessel (2), in the xy plane.

4. Sorting device (1) according to any of the preceding claims, characterized in that said looped trajectories are substantially circular or elliptical.

5. Sorting device (1) according to any of the preceding claims, characterized in that said looped trajectories have a mean characteristic diameter of 2 cm or smaller, preferably 1 cm or smaller.

6. Sorting device (1) according to any of the preceding claims, characterized in that said vessel support (30) is connected to said frame (10) by at least one elastic blade, preferably at least two elastic blades.

7. Sorting device (1) according to any of the preceding claims, characterized in that said driving organ comprises a crank arranged in contact with said vessel support (30) to drive the latter along said looped trajectories.

Sorting device (1 ) according to any of the preceding claims,characterized in that said vessel support (30) has at least one see-through window (28), andin that said identification and localization system comprises at least one image acquisition device mounted on said frame (10) and facing said see-through window (28) so as to be able to acquire an image of biological entities included in a sample contained in a transparent source vessel (2) while it is located on said vessel support (30).

Method for the selective picking up of at least one biological entity from a source vessel (2) containing a sample including a plurality of biological entities, to be able to selectively dispense said at least one biological entity into a destination vessel (6), characterized in that it comprises the steps consisting in:a) providing a sample including a plurality of biological entities in a source vessel (2) and put the latter down on a vessel support (30) of a sorting device (1) according to any of claims 1 to 8,b) assessing the distribution in said source vessel (2) of the plurality of biological entities included in said sample, in particular, if said at least one biological entity to be selectively picked up is close to a wall of said source vessel (2),c) implementing a stirring operation of said sample, on the basis of a manual request by a user or of an automatic request by said identification and localization system, during which said driving organ drives said vessel support (30) according to successive looped trajectories, inside said xy plane, with predefined shapes and speed parameters which are adapted to stir said sample and provide movements ofthe plurality of biological entities between each other and with respect to said source vessel (2) so as to be able to substantially gather the plurality of biological entities inside a central region of the source vessel (2), in the xy plane,d) acquiring at least one image of said sample to locate said at least one biological entity to be selectively picked up, ande) selectively picking up said at least one biological entity to be selectively picked up.

10. Method according to claim 9, characterized in that said vessel support (30) is driven by said driving organ with an acceleration having a radial component of at least 0.1 m / s2in step c).

Method according to claim 9 or 10, characterized in that, if it appears in step b) that said at least one biological entity to be selectively picked up is close to a wall of said source vessel (2), said vessel support (30) is driven, in step c), along successive looped trajectories with a rotation speed of 120 rpm or less, so as to be able to substantially gather the plurality of biological entities inside a central region of said source vessel (2), in said xy plane.

12. Method according to any of claims 9 to 11, characterized in that said looped trajectories are substantially circular or elliptical.
