A METHOD FOR TREATING PSMA EXPRESSING CANCERThis application claims the benefit of priority to International Patent Application No. PCT / CN2025 / 079903, filed on February 28, 2025, the contents of which are incorporated herein by reference in their entirety.FIELDThe present application relates to a method of treating cancer. In particular, the present application relates to a method for treating PSMA expressing cancer by using the 225Ac radionuclide complexed compound provided herein.BACKGROUNDOriginally identified on the surface of prostate cancer cells, Prostate-Specific Membrane Antigen (PSMA) is a type II transmembrane glycoprotein encoded by the FOLH1 (folate hydrolase 1) gene located on chromosome 11. Despite its name suggesting organ specificity, PSMA is also expressed in various tissues throughout the body, including liver, kidney, and brain.Since its discovery, PSMA has garnered significant interest in the medical field, particularly in prostate cancer diagnostics and therapeutics. In diagnostics, PSMA-targeted imaging agents based on positron emission tomography (PET) radiotracers has demonstrated the accuracy of detecting prostate cancer metastases and recurrences. Therapeutically, PSMA serves as a target for precision medicine strategies, such as antibody-drug conjugates (ADCs) and PSMA-targeted radionuclide therapies, which deliver cytotoxic agents or radiation directly to PSMA-expressing cells. Considerable benefits have been demonstrated in patients with prostate cancer by using 177Lu-PSMA-617, a radio-conjugate which delivers beta-particle radiation to PSMA expressing cancers. However, eventually patients progress under β-emitting radio-conjugates and effective therapeutics with improved efficacy are still in demand.SUMMARYThe present application addresses these needs. In one aspect, provided herein is a method of treating PSMA expressing cancer in a subject in need thereof, comprising administrating an 225Ac radionuclide complexed compound to the subject at a unit dose of from about 0.5MBq to about 15MBq, wherein the compound comprises a structure of Formula (I) :andwherein:(1) T is a branching group which is at least trivalent,(2) Z is a PSMA targeting moiety specifically binding to PSMA expressed on a cell, and Z is conjugated to T via a linker LZ,(3) E is chelating group capable of complexing with the 225Ac radionuclide, and E is conjugated to T via a linker LE, and(4) A is an albumin binding group, and A is conjugated to T via a linker LA.In some embodiments of the method of the present application, the compound comprises a structure of:In some embodiments of the method of the present application, the PSMA expressing cancer is selected from the group consisting of prostate cancer, salivary gland cancer, bladder cancer, breast cancer, gastro-intestinal cancer, renal cancer, testicular-embryonal cancer, neurological tumors, and brain tumor. In some embodiments, the PSMA expressing cancer is prostate cancer. In some embodiments, the prostate cancer is metastatic castration resistant prostate cancer (mCRPC) . In some embodiments, the subject has received Androgen Deprivation Therapy prior to the administration of the 225Ac radionuclide complexed compound.In some embodiments, the method comprises administrating the 225Ac radionuclide complexed compound to the subject at a unit dose of no less than about 5 MBq. In some embodiments, the method comprises administrating the 225Ac radionuclide complexed compound to the subject at a unit dose of no less than about 6 MBq. In some embodiments, the method comprises administrating the 225Ac radionuclide complexed compound to the subject at a unit dose of about 5 MBq to about 10 MBq. In some embodiments, the method comprises administrating the 225Ac radionuclide complexed compound to the subject at a unit dose of about 6 MBq, about 7 MBq, about 8 MBq, or about 9 MBq. In some embodiments, the method comprises administrating the 225Ac radionuclide complexed compound to the subject at a unit dose of about 7MBq.In some embodiments of the method of the present application, the 225Ac radionuclide complexed compound is administrated to the subject intravenously.In some embodiments of the method of the present application, the 225Ac radionuclide complexed compound is administrated to the subject at an interval of at least 4 weeks. In some embodiments, the 225Ac radionuclide complexed compound is administrated to the subject at an interval of at least 6 weeks.In some embodiments, the 225Ac radionuclide complexed compound is administrated to the subject for at least 2 cycles. In some embodiments, the 225Ac radionuclide complexed compound is administrated to the subject for at least 4 cycles. In some embodiments, the 225Ac radionuclide complexed compound is administrated to the subject for at least 6 cycles.In some embodiments of the method of the present application, the 225Ac radionuclide complexed compound is administrated to the subject at an interval of 6 weeks for 6 cycles.In some embodiments of the method of the present application, the subject achieves PSA50 after the administration of the 225Ac r / nmol. After administration, the animals were sacrificed at different timepoints (0.5, 1, 4, 24, 48, 72, and 96 hours, n = 6 / each timepoint) . Then the organs were excised, and radioactivity of the organs was measured in a gamma-counter. Results of the biodistribution of 111In-Cpd I-21 in major organs, expressed as injected dose per gram of tissue (%ID / g) , are shown in Figure 1A.The biodistribution data expressed as percent of the injected dose per gram of tissue (%ID / g) were extrapolated to humans using the %-kg / g (relative organ-to-body masses) method. Results of this extrapolation give an estimate of the percent injected dose per organ in adult males. The time integrals of activity or residence times, also known as the area under the curve (AUC) , were computed for each set of source organs. Then, these AUCs were entered into the OLINDA 2.0 software to calculate the absorbed doses, using the adult male model through adult male dosimetry phantom. All organ radiation doses were calculated using the default organ masses.The human (adult male) radiation doses estimate for [225Ac] Ac-Cpd I-21 based on preclinical [111In] In-CPD I-21 biodistribution data in normal mice are as shown in Table 2.Table 2 Predicted [225Ac] Ac-Cpd I-21 Human Organ Absorbed Radiation Doses Based on [111In] In-Cpd I-21 Biodistribution Study inKunming MiceBased on the extrapolation results listed in Table 2, the organs receiving the highest radiation doses were osteogenic cells (0.904 Gy / MBq) , red marrow (0.241 Gy / MBq) , kidney (0.217 Gy / MBq) , and salivary glands (0.154 Gy / MBq) . Assuming absorbed dose thresholds of 23 Gy for kidneys, 2 Gy for bone marrow, and 35 Gy for liver, bone marrow would be the critical organ for 225Ac-Cpd I-21, with the corresponding maximum administered radioactivity dose of 8.33 MBq. At this administered dose, the absorbed radiation dose to kidney and salivary glands would be 1.8 Gy and 1.28 Gy, respectively (well below the thresholds) , while osteogenic cells would receive 7.53 Gy, which is in the range of osteogenic cell absorbed radiation dose estimated in clinical experience with other RLT compounds.1.2 Estimation of Human Organ Absorbed Radiation Dose for [225Ac] Ac-Cpd I-21 from [111In] In-Cpd I-21 Biodistribution Data inNMRI Nude MiceDosimetry is a methodology to determine the absorbed dose (absorbed energy of ionizing radiation per unit mass) to tumors and organs-at-risk. It allows the optimization of the injected amount of the radiopharmaceutical which enables treatment planning prior to new cycles of therapy. However, quantitative images, which are required for dosimetry, are difficult to obtain for some therapeutic radionuclides.225Ac could potentially be imaged due to its gamma emitting daughters (francium-221 (221Fr or Fr-221) and bismuth-213 (213Bi or Bi-213) ) , but as the injected activities being very low, it is challenging to obtain good counting statistics in a clinically acceptable scan duration. To overcome this issue, imaging nuclides such as indium-111 (111In or In-111) , copper-64 (64Cu or Cu-64) or Gallium-68 (68Ga or Ga-68) can be used as surrogates to predict therapeutic absorbed doses or to perform retrospective dosimetry.Biodistribution study was conducted inNMRI nude mice using [111In] In-Cpd I-21. Briefly, [111In] In-Cpd I-21 was administrated to each animal at a dose of 3 MBq / animal, with a specific activity of 20 MBq / nmol, via IV injection. After administration, animals were sacrificed at 1, 4, 24, 48, 72, and 168 hours, and then the organs were excised and counted in a gamma-counter. Results of the biodistribution of 111In-Cpd I-21 in major organs, expressed as injected dose per gram of tissue (%ID / g) , are shown in Figure 1B and Figure 1C.The biodistribution data were extrapolated to human expected uptake per gram of tissue by multiplying by the animal body weight and dividing by the human MIRD89 phantom weight; 60 kg for females and 73 kg for males. Then AUC was calculated to determine the total number of disintegrations per gram tissue. Activity still present at the last timepoint was assumed to fully decay in the tissue. The calculated total number of decays per gram tissue was converted to residence time per gram (MBq·h / MBq / g) . Values were then multiplied with the organ mass in the respective male or female MIRD89 phantom, to get the human residence times. The values for human residence times were used as input in OLINDA / / nmol. After the administration, animals were scarified at 1, 4, 24, 48, 72, and 168 hours, and then the organs were excised and counted in a gamma-counter at secular equilibrium. Results of the biodistribution of [225Ac] Ac-Cpd I-21 in major organs, expressed as injected dose per gram of tissue (%ID / g) , are shown in Figure 1D, Figure 1E and Figure 1F.The biodistribution data were extrapolated to human expected uptake per gram of tissue by multiplying by the animal body weight and dividing by the human MIRD89 phantom weight; 60 kg for females and 73 kg for males. Then AUC was calculated to determine the total number of disintegrations per gram tissue. Activity still present at the last timepoint was assumed to fully decay in the tissue. The calculated total number of decays per gram tissue was converted to residence time per gram (MBq·h EXM. The results are shown in Table 4.Table 4 Predicted [225Ac] Ac-Cpd I-21 Human Organ Absorbed Radiation Doses Based on Preclinical [225Ac] Ac-Cpd I-21 Biodistribution Study inNMRI Nude MiceBased on the organ absorbed doses indicated in Table 4, the dose limiting organ is estimated to be the red marrow, as an injection of 1 MBq 225Ac-Cpd I-21 gives an equivalent organ dose of 122 mGy to the red marrow. The dose limit of 2 Gy is expected to be reached when injecting 16.4 MBq of 225Ac-Cpd I-21.1.4 Estimation of Human Organ Absorbed Radiation Dose from the Study on Redistribution of Actinium-225 daughter isotopes Bismuth-213 and Francium-221 after Administration of 225Ac-Cpd I-21 inNMRI Nude MiceRedistribution of Actinium-225 daughter isotopes, Bismuth-213 and Francium-221, after administration of [225Ac] Ac-Cpd I-21 inNMRI nude mice was analyzed. Briefly, [225Ac] Ac-Cpd I-21 was administrated at a radioactive dose of 37 kBq to each animal via IV injection, with a specific radioactivity of 190.3 kBq / nmol. After the administration, the mice were scarified at 1, 4, 24, 48, and 120 hours, and then the organs were excised and counted in a gamma-counter.Blood, liver, and kidneys were continuously counted at non-equilibrium every 15-20 minutes for 3 hours, and the duration of each counting was 120 seconds (1-and 4-hour timepoints) or 180 seconds (24, 48, and 120 hours) . At least 6 hours after tissue harvest, all the collected tissue (including blood, liver, and kidneys) were counted 180 seconds in a gamma counter (equilibrium counting) . Data were analyzed in multiple channels including: channel 1: 15-630 keV (225Ac at secular equilibrium) , channel 2: 200-240 keV (221Fr) , channel 3: 400-480 keV (213Bi) .A biexponential expression was fitted to the decay curves obtained to determine the equilibrium 225Ac and free daughters 213Bi and 221Fr at the time of sacrifice. Results of the biodistribution of [225Ac] Ac-Cpd I-21 in major organs, expressed as injected dose per gram of tissue (%ID / g) , are shown in Figure 1G. Biodistribution of 225Ac, 213Bi, and 221Fr in the blood, liver, and kidneys were compared to demonstrate the relocation of daughter decays, as shown in Figure 1I.The 225Ac daughter redistribution data were further used for dosimetry extrapolations. Particularly, the daughter radionuclides dosimetry was calculated using the “decay-series” in OLINDA with relative ratios of the source organs used for each of the different daughters of 225Ac. The input was based on the re-distribution data for the liver, kidney, and blood as described above.The dosimetry result considering the redistribution effect of all daughters are listed in Table 5. Additionally, dosimetry calculation was also performed assuming all 225Ac daughter isotopes decayed in the same physical / anatomical location as the parent 225Ac nuclide (without considering redistribution effect of daughter isotopes) , as shown in Table 5.Table 5 Predicted [225Ac] Ac-Cpd I-21 Human Organ Absorbed Radiation Doses Based on the Redistribution Data Generated for daughter isotopes 221Fr and 213BiTable 6 Predicted [225Ac] Ac-Cpd I-21 Human Organ Absorbed Radiation Doses without considering the redistribution of the daughter isotopesA summary of all the dosimetry predictions above is summarized in Table 7 below.Table 7 Summary of Predicted Clinical [225Ac] Ac-Cpd I-21 Human DosimetryAs can be seen in Table 7, the biodistribution data presented some variability, as expected, due to the differences in animal models, isotopes, doses, and specific radioactivity. However, the time-activity profiles in blood and in the organs had a similar behavior in the different studies. The redistribution effect of daughter isotopes had limited impact on absorbed dose of liver and total body.Based on above data, the dosimetry extrapolations into human showed that the dose limiting organ might potentially be the bone marrow, with a predicted absorbed radiation dose per MBq in the range of 0.122-0.241 Gy / MBq. Therefore, the estimated maximum cumulative administered dose that would not exceed the red marrow threshold of 2 Gy is in the range 8.3-16.4 MBq.Based on above findings, an 8-fold safety margin to the lowest estimated dose is determined as the starting dose, and therefore the first-in-human dose is planned to be 1 MBq, as shown in Figure 2.Example 2 Dosimetry Evaluation with 111In labeled compound in human.In the clinical study, selected patients received [111In] In-Cpd I-21 at 185±20 MBq at least 8 days prior to the first injection of the corresponding [225Ac] Ac-Cpd I-21 for dosimetry evaluation, through imaging. Each of the selected patients received SPECT / CT scans from vertex to mid-thigh at approximately 4 ± 0.5 h, 24 ± 3 h, 72 ± 24 h and 168 ± 24 h after i. v. injection of 1 [111In] In-Cpd I-21. As exemplarily shown in Figure 3A and 3B, [111In] In-Cpd I-21 displayed high and prolonged tumor uptake and low salivary gland uptake, suggesting the potential for a differentiated safety profile, including low rate of xerostomia.Example 3 Treatment of PSMA expressing cancer by using the 225Ac complexed Cpd I-21.The study is a first-in-human, open-label, multicenter Phase I study of [225Ac] Ac-Cpd I-21 in patients with mCRPC. Inclusion and Exclusion Criteria of the patients are summarized in Table 8 below.Table 8 Inclusion and Exclusion CriteriaPart 1 Dose EscalationPart 1 was to establish the safety profile of [225Ac] Ac-Cpd I-21 and its treatment response which was evaluated by measuring PSA levels in blood. The initial part of the dose escalation was consisted of cohorts of 1-3 patients, followed by expansion cohorts of 3-6 patients each, as illustrated in Table 9 and Figure 2.The final decision to dose-escalate was made by the Data Review Committee (DRC) comprising the Investigators, the Medical Monitor, the Study Statistician and medical representatives of the Sponsor, based on the recommendation of the BLRM and review of all available safety data (including AEs and laboratory abnormalities that are not DLTs) , PK data, dosimetry data and efficacy information available at the time of the decision.Table 9 Provisional dose levels for dose escalationa Dose level -1 represents a dose that may be evaluated if dose level 1 is poorly tolerated. No dose de-escalation below this level is planned for this study. If dose level -1 is poorly tolerated, the study will be terminated.b It is possible for additional dose levels to be added during the course of the study.As of 02 January 2026:(1) Cohort 1: one patient was enrolled onto Cohort 1 at dose level 1 MBq and no DLT was observed during the DLT observation period. The patient experienced mostly AEs grades 1 and 2 after receiving treatment, and a SAE of pain which led to hospitalization. The patient later experienced disease progression during cycle 2 and was discontinued from treatment.(2) Cohort 2: one patient was enrolled onto Cohort 2 at dose level 2 MBq and no DLT was observed during the DLT observation period. The patient is ongoing in cycle 3.(3) Cohort 3: three patients were enrolled onto Cohort 3 at dose level 3 MBq and no DLT was observed during the DLT observation period. The patients experienced mostly AEs grades 1 and 2 after receiving treatment. All 3 patients are ongoing in cycle 1.(4) Cohort 4: four patients were enrolled in Cohort 4, 3 had received 3 doses and were ongoing and 1 received 2 doses and discontinued treatment due to a Grade 2 AE of dry mouth.(5) Cohort 5: six patients were enrolled in Cohort 5 and had received 1 dose. All were still ongoing with treatment.Figure 4 illustrates the decrease in PSA (%) after treatment with [225Ac] Ac-Cpd I-21. Figure 5A-5C and Figure 6A-6C illustrate the corresponding hematology and blood chemistry profile of the patients.Based on the results, [225Ac] Ac-Cpd I-21has been well tolerated up to 5 MBq per cycle. Out of 6 patients enrolled in Cohort 5, 2 showed a PSA50 response after the first dose. No dose-limiting toxicities or significant safety signals were identified. These findings support further dose escalation to 6MBq or above, given the favorable safety profile and possibility to improve anti-tumor activity at higher doses.Part 2 Dose ExpansionAfter dose escalation, enrollment into Part 1 will be considered complete and enrollment into Part 2 will begin. Further approximately 18 patients are planned to be enrolled in Part 2 to gain more safety and additional efficacy data of the patients. Patients may continue treatment after radiological or biochemical progression with approval of the Medical Monitor and, if in the Investigator’s opinion the patient is getting clinical benefit from the prolonged study therapy.Patients that benefit from treatment after 6 cycles of [225Ac] Ac-Cpd I-21 may receive additional cycles after discussing benefit and potential risks with their treating physician and the sponsor every two cycles.While the present application has been described with reference to examples, it is to be understood that the scope of the claims should not be limited by the embodiments set forth in the examples but should be given the broadest interpretation consistent with the description as a whole.All patents, patent applications and publications cited herein are hereby incorporated by reference in their entirety. The disclosures of these publications in their entireties are hereby incorporated by reference into this application in order to more fully describe the state of the art as known to those skilled therein as of the date of the application described and claimed herein.

A method of treating PSMA expressing cancer in a subject in need thereof, comprising administrating an 225Ac radionuclide complexed compound to the subject at a unit dose of from about 0.5MBq to about 15MBq, wherein the compound comprises a structure of Formula (I) :andwherein:(1) T is a branching group which is at least trivalent,(2) Z is a PSMA targeting moiety specifically binding to PSMA expressed on a cell, and Z is conjugated to T via a linker LZ,(3) E is chelating group capable of complexing with the 225Ac radionuclide, and E is conjugated to T via a linker LE, and(4) A is an albumin binding group, and A is conjugated to T via a linker LA.The method of claim 1, wherein the compound comprises a structure of:The method of claim 1 or 2, wherein the PSMA expressing cancer is selected from the group consisting of prostate cancer, salivary gland cancer, bladder cancer, breast cancer, gastro-intestinal cancer, renal cancer, testicular-embryonal cancer, neurological tumors, and brain tumor.The method of claim 3, wherein the PSMA expressing cancer is prostate cancer.The method of claim 4, wherein the prostate cancer is metastatic castration resistant prostate cancer (mCRPC) .The method of claim 5, wherein the subject has received Androgen Deprivation Therapy prior to the administration of the 225Ac radionuclide complexed compound.The method of any one of claims 1-6, wherein the method comprises administrating the 225Ac radionuclide complexed compound to the subject at a unit dose of no less than about 5 MBq.The method of any one of claims 1-6, wherein the method comprises administrating the 225Ac radionuclide complexed compound to the subject at a unit dose of no less than about 6 MBq.The method of any one of claims 1-8, wherein the method comprises administrating the 225Ac radionuclide complexed compound to the subject at a unit dose of about 5 MBq to about 10 MBq.The method of claim 9, wherein the method comprises administrating the 225Ac radionuclide complexed compound to the subject at a unit dose of about 6 MBq, about 7 MBq, about 8 MBq, or about 9 MBq.The method of claim 10, wherein the method comprises administrating the 225Ac radionuclide complexed compound to the subject at a unit dose of about 7MBq.The method of any one of claims 1-11, wherein the 225Ac radionuclide complexed compound is administrated to the subject intravenously.The method of any one of claims 1-12, wherein the 225Ac radionuclide complexed compound is administrated to the subject at an interval of at least 4 weeks.The method of claim 13, wherein the 225Ac radionuclide complexed compound is administrated to the subject at an interval of at least 6 weeks.The method of any one of claims 1-14, wherein the 225Ac radionuclide complexed compound is administrated to the subject for at least 2 cycles.The method of claim 15, wherein the 225Ac radionuclide complexed compound is administrated to the subject for at least 4 cycles.The method of claim 15, wherein the 225Ac radionuclide complexed compound is administrated to the subject for at least 6 cycles.The method of any one of claims 13-17, wherein the 225Ac radionuclide complexed compound is administrated to the subject at an interval of 6 weeks for 6 cycles.The method of any one of claims 1-18, wherein the subject achieves PSA50 after the administration of the 225Ac radionuclide complexed compound.The method of claim 19, wherein the subject achieves PSA50 by no more than 2 cycles of the administration of the 225Ac radionuclide complexed compound.The method of claim 19, wherein the subject achieves PSA50 by 1 cycle of the administration of the 225Ac radionuclide complexed compound.The method of any one of claims 1-21, wherein the method comprises administrating the 225Ac radionuclide complexed compound comprising a structure of formula (II) to the subject at a unit dose of 7Mbq at an interval of 6 weeks for 6 cycles.The method of any one of claims 1-21, wherein the method comprises administrating the 225Ac radionuclide complexed compound comprising a structure of formula (II) to the subject at a unit dose of 8Mbq at an interval of 6 weeks for 6 cycles.An 225Ac radionuclide complexed compound for use in treating PSMA expressing cancer in a subject in need thereof, comprising administrating the 225Ac radionuclide complexed compound to the subject at a unit dose of from about 0.5MBq to about 10MBq.A kit for treating PSMA expressing cancer in a subject in need thereof comprising:(1) an 225Ac radionuclide complexed compound, and(2) a package insert indicating administrating the 225Ac radionuclide complexed compound to the subject at a unit dose of from about 0.5MBq to about 10MBq.The kit of claim 25, wherein the kit further comprises an 111In radionuclide complexed compound for conducting dosimetry analysis before administrating the 225Ac radionuclide complexed compound.
