[0002] The application claims the priority to PCT Application No. PCT / CN2025 / 081856, filed on March 11, 2025. The content of the prior PCT applications is considered as a part of the present disclosure and are incorporated herein in its entirety.Technical Field

[0003] The present disclosure relates to field of biomedicine. In particular, the present disclosure relates to recombinant extracellular vesicles for bioactive protein delivery and uses thereof.Background

[0004] Skin aging and related cell senescence are caused by extrinsic factors (e.g., ultraviolet light exposure, smoking, pollution) and intrinsic factors (e.g., time, genetic factors, hormones) . UV radiation is the most potent extrinsic driver of age-related change in the skin, known as “photoaging” . Photoaging accounts for approximately 80%of facial aging. Photoaging of the skin is clinically manifest principally as actinic elastosis, wrinkles and dyspigmentation. UV radiation triggers the excessive creation of reactive oxygen species (ROS) in the epidermis and dermis layers of the skin, mitochondria is the main site of ROS production in cells. ROS exert negative effects on skin cells, including keratinocytes and fibroblasts, by inducing oxidative damage and promoting inflammatory responses. These effects lead to a reduction in cellular viability, accelerated collagen degradation, and upregulation of pro-inflammatory cytokines. Multiple characteristics of skin aging are associated with inflammation. During the polarization of M0 macrophages into pro-inflammatory M1 macrophages, the increase in ROS levels is often observed. Meanwhile, excessive ROS further promotes M1 polarization. Cytokines such as IL-6 and TNF-α are markers of M1 macrophages, and the upregulated levels can be found in the photoaging skin.

[0005] The use of small molecules therapeutically has been disappointing, which are ineffective to scavenging ROS inside cells. Antioxidant enzymes react thousands to millions of times more rapidly with those oxidants than small molecules do and provide the predominant antioxidant defense. As the most active antioxidant enzyme in both nature and the human body, catalase plays a crucial role in mitigating oxidative stress by efficiently scavenging ROS. Catalase's ability to decompose hydrogen peroxide into water and oxygen significantly reduces oxidative damage caused by UV radiation. This enzymatic activity not only protects keratinocytes and fibroblasts from ROS-induced cellular damage but also helps maintain skin integrity and elasticity. Catalase can serve as an effective strategy for ameliorating the visible signs of photoaging, such as wrinkles and dyspigmentation.

[0006] Extracellular vesicles (EVs) are lipid bilayer nanoparticles released from all cell types into extracellular space. EVs naturally transport essential cellular components (i.e., proteins or active enzyme, lipids, nucleic acids such as mRNA, micro-RNA) for intercellular communication. Therefore, EVs are promising in delivering an active ingredient, payload or therapeutic agent to or within a target cell. As natural secretomes from cells, EVs have numerous advantages as drug delivery system / vehicle compared to synthetic drug delivery vehicles (e.g. liposomes, lipid nanoparticles and virus vectors) . EVs are naturally derived from cells, making them inherently biocompatible and less likely to provoke an immune response compared to synthetic vehicles. This characteristic is crucial for reducing potential side effects in therapeutic applications; EVs possess intrinsic tissue-targeting abilities and an navigate through biological barriers; EVs exhibit great stability in biological fluids, which contributes to a prolonged circulation time in the extracellular matrix and bloodstream. EVs have been extensively validated in both scientific research and clinical applications with proven efficacy and safety in the field of medical aesthetics. Their potential therapeutic applications include anti-aging, anti-pigmentation, wound healing, hair regeneration, and others. However, active ingredient is very limited in natural stem cell derived EV.

[0007] Catalase has a short half-life due to rapid proteolytic degradation and immunogenicity when introduced into the body. Therefore, encapsulating catalase in EVs could enhance its stability and prolong its effective half-life, allowing it to better penetrate target tissues and cells, where it can scavenge reactive oxygen species effectively. Different methodologies have been developed for the loading of therapeutic agents into EVs including sonication, electroporation, and passive incubation. However, these conventional methods involve multiple manufacturing steps including catalase protein expression, EV production, and loading. They are plagued by drawbacks such as relatively low loading efficiency and high production costs.

[0008] There is unmet need to provide low-cost and efficient catalase and catalase delivery system to improve photoaging skin conditions.Summary of Invention

[0009] To solve the aforementioned technical problems, the present disclosure provides catalase and catalase delivery system by genetic engineering approaches, the producing cells are modified to achieve high-level expression of recombinant human catalase protein (rhCAT) and recombinant human extracellular vesicles (rhEV) , which streamlines the production and loading processes into a one-step approach, not only enhances loading efficiency but also significantly reduces overall production costs by minimizing the complexity of manufacturing workflows. Further, catalase is modified to target mitochondrial, which is more specific. The catalase and catalase delivery system in present disclosure presents a promising avenue for scavenging reactive oxygen species (ROS) , mitigating oxidative stress, and ameliorating the visible signs and / or biomarkers of photoaging of the skin.

[0010] In the first aspect, the present disclosure provides a catalase or its variant, which at least has one of the bioactivities of scavenging reactive oxygen species (ROS) , mitigating oxidative stress, and / or ameliorating the visible signs and / or biomarkers of photoaging of the skin.

[0263] 4.1 Animal models of photoaging

[0264] The female SD rats (6 weeks old, with an average weight of approximately 150 g) were purchased from Liaoning Changsheng Biotechnology Co. The rats were acclimated for one week in the SPF-grade animal facility at the College of Life Sciences, Jilin University, under controlled conditions of 25℃, with free access to water and food to adapt to the environment. The dorsal hair of SD rats was removed. All rats except the control group (n=4) were exposed to UV irradiation to simulate photoaged skin conditions. The total doses of UVA and UVB were set at 14.07 J / cm2 and 11.66 J / cm2 respectively, with the UV lamp positioned 30 cm away. Rats received UV irradiation once daily for 7 weeks. EV-treated groups (313EVCAT and naked EV) and hyaluronic acid (HA) -treated group were delivered via intradermal injection, and 0.5%retinoic acid (RA) was topically applied to the dorsal skin (RA-treated group) . During the experimental period, photographs were taken and the condition of the dorsal skin of each rat was documented regularly. At the end of the experiment, the rats were euthanized by cervical dislocation. Blood samples were collected from the orbital sinus of each rat, and the dorsal skin was excised and collected for subsequent analysis.

[0265] 4.2 Evaluation of effect of attenuating UV-induced dermal wrinkles

[0266] The photographs of the dorsal skin of the rats were imported into the image analysis software (Image Pro Plus) , and wrinkle detection was performed using the software. The collected wrinkle data were subjected to statistical analysis to assess the construction of the photoaging model and the effects of treatment on wrinkle quantity.

[0267] Following successful model establishment, treatments were administered on days 1, 5, 8, 15, 22, and 28. Rats were photographed and weighed at each time point (Figure 24) . At the end of the study, dorsal skin samples were collected for histological analysis and skin replica casting. Beginning on day 22, the high-dose (1×1010 particles / mL) 313EVCAT group exhibited a significantly greater reduction in wrinkle formation compared to the naked EV and hyaluronic acid (HA) groups, with further improvement noted by day 28. These findings indicate that intradermal administration of 313EVCAT effectively improves skin condition and reduces photoaging-induced wrinkle (Figure 26, Table 4) . Skin replicas obtained on day 28 confirmed the superior anti-wrinkle efficacy of 313EVCATs relative to the UV, naked EVs, HA, and retinoic acid (RA) control groups (Figure 25) . Remarkably, high-dose (1×1010 particles / mL) 313EVCAT-treatment over 28 days markedly reduced dermal thickening, with therapeutic outcomes surpassing those of the RA group (Figure 27, Table 5) , underscoring the potent anti-photoaging effect of 313EVCATs.

[0270] 4.3 Evaluation of effects of 313EVCAT on pathological features and collagen content

[0271] To evaluate the histopathological impact of 313EVCAT-treatment on photoaged skin, H&E staining was performed on dorsal skin samples from SD rats. The SD rats were euthanized by cervical dislocation, and the dorsal skin tissues were excised using surgical scissors. Excess fat tissues were removed, and the skin samples were placed in centrifuge tubes containing 4%paraformaldehyde solution for fixation for 24 h. The fixed skin tissues were then removed from the tubes and embedded to prepare paraffin sections. Hematoxylin and eosin (H&E) staining was performed, and the stained sections were examined under an optical microscope to observe the structure of each skin layer, as well as the presence of collagen and inflammatory cell infiltration within the skin. Paraffin sections were prepared and stained with Masson's trichrome staining. The morphology and distribution of collagen fibers, as well as the collagen content, were examined under an optical microscope. The collagen content in the tissues was quantitatively assessed using ImageJ software.

[0272] In the UV-irradiated control group, marked epidermal erosion, necrosis, and extensive inflammatory cell infiltration were observed, consistent with characteristic features of photoaging and photodamage. In contrast, 313EVCAT (1×1010 particles / mL) -treatment significantly attenuated these pathological changes, reducing epidermal injury and inflammation while restoring dermal collagen fiber structure and density (Figure 28) . To assess collagen deposition, skin samples collected on day 28 post-treatment were subjected to Masson’s trichrome staining, followed by quantitatively analysis for collagen content. Compared to HA and RA treatments, 313EVCATs administration led to a pronounced increase in collagen synthesis and remodeling, reflecting its superior capacity to promote tissue repair and matrix regeneration (Figure 28) . Immunohistochemical analysis was performed to assess the collagen content in skin sections. The 313EVCAT-treatment group exhibited significantly improved regeneration of Col 1a1 compared to the untreated group, with skin and collagen structure showing better therapeutic effects than those observed with hyaluronic acid (HA) and retinoic acid (RA) treatments (Figure 29, Table 6) . Moreover, RT-qPCR analysis (Bio-Rad T100) revealed a 4.38-fold increase in Col 1a1 mRNA expression in the high-dose (1×1010 particles / mL) 313EVCAT-treated group compared to controls, confirming enhanced collagen synthesis (Figure 30, Table 6) .

[0274] Tissue sections were immersed in the dihydroethidium (DHE) working solution, ensuring that the dye uniformly covered the sections. The sections were then incubated in the dark for a period of time to allow DHE to penetrate the cells and be oxidized by ROS. After incubation, the sections were washed with PBS or another appropriate buffer to remove unbound DHE dye.

[0275] In ROS detection, the sections were observed under a fluorescence microscope with appropriate excitation and emission wavelengths set to detect the fluorescence signal generated by oxidized DHE. The intensity and distribution of the fluorescence signal within the cells were observed and recorded, with the fluorescence intensity being proportional to the level of ROS. The fluorescence signal was quantitatively analyzed using ImageJ software to assess the intracellular ROS levels. The antioxidant properties of 313EVCATs were investigated by dihydroethidium (DHE) staining to quantify ROS levels in skin tissues (Figure 31) . Fluorescence imaging and quantitative analysis revealed a significant reduction in ROS accumulation in the 313EVCAT-treated groups, with the high-dose group (313EVCAT (1×1010 particles / mL) ) demonstrating more potent ROS scavenging activity than the low-dose group (313EVCAT (1×109 particles / mL) ) , indicating a dose-dependent antioxidant effect (Figure 32, Table 7) .

[0277] 4.4 Evaluation of effect of 313EVCAT on anti-inflammatory

[0278] To investigate the anti-inflammatory effects of 313EVCATs, immunofluorescence was used to examine the levels of CD206 (Anti-CD206 is purchased from PROTEINTECH, 18704-1-AP) and CD86 (Anti-CD86 is purchased from Thermo Fisher, RAB02398) in skin sections. In the 313EVCAT-treated group, the immunofluorescence sections showed a substantial expression of green fluorescence for CD206, while the expression of CD86 was significantly reduced compared to the untreated group. The green fluorescence of CD206 in the 313EVCAT-treated group was significantly increased compared to other treatment groups, while CD86 showed the opposite trend. This indicates that 313EVCAT-treatment has the effect of transforming pro-inflammatory M1 macrophages into anti-inflammatory M2 macrophages. The results indicated that the low-dose (1×109 particles / mL) 313EVCAT-treated group also exhibited significant therapeutic effects. 313EVCAT-treated group demonstrated superior therapeutic effects in reversing macrophage phenotypes compared to the commonly used drugs HA and RA (Figure 33) .

[0279] In addition, to investigate the therapeutic efficacy of 313EVCATs in vivo, their impact on anti-inflammatory responses was systematically analyzed in a UV-induced photoaging rat model. RT‐qPCR analysis (Bio-Rad T100) demonstrated that 313EVCAT-treatment significantly downregulated mRNA expression of proinflammatory cytokines IL‐6 (Figure 34) , and TNF‐α (Figure 35) compared to the control group. Notably, even low-dose (1×109 particles / mL) 313EVCAT-treatment led to substantial reductions in inflammatory gene expression. In the high-dose (1×1010 particles / mL) group, IL-6 mRNA levels were comparable to those in the NC group, suggesting effective suppression of UV-induced inflammation. Consistent with the mRNA findings, ELISA results showed a significant dose-dependent reduction in IL‐1β (Figure 36) , IL‐6 (Figure 37) , and TNF‐α (Figure 38) protein levels in serum of 313EVCAT-treated rats. IL-1β was measured by ELISA reagent (Elabscience, E-EL-R0012) , IL-6 was measured by ELISA reagent (Elabscience, E-EL-R0015) , TNF-α was measured by ELISA reagent (Elabscience, E-EL-R2856) .
