Abstract

Provided herein are methods, kits, and articles of manufacture useful for treating a hepatitis B virus (HBV) infection. In some embodiments the methods comprise administering to a subject an antisense oligonucleotide specific for an HBV target and an HBV vaccine and / or an immunomodulator. In some embodiments, the methods comprise administering an antisense oligonucleotide specific for an HBV target and a siRNA specific for an HBV target. In some embodiments the methods comprise administering to a subject an antisense oligonucleotide specific for an HBV target, a siRNA specific for an HBV target, and an HBV vaccine and / or an immunomodulator. The methods and kits described herein may be useful for treating chronic HBV infection.

DOSING REGIMENS FOR HBV TREATMENTRELATED APPLICATIONS

[0003] Hepatitis B is a viral disease transmitted parenterally by contaminated material such as blood and blood products, contaminated needles, sexually and vertically from infected or carrier mothers to their offspring. The World Health Organization (WHO) estimates that 254 million people were living with chronic hepatitis B infection in 2022, with 1.2 million new infections each year. In 2022, hepatitis B resulted in an estimated 1.1 million deaths, mostly from cirrhosis and hepatocellular carcinoma (primary liver cancer) (world wide web: who. int / news-room / fact-sheets / detail / hepatitis-b) . Thus new anti-HBV therapies, including therapies capable of treating chronic HBV, are needed to reduce the risk of HBV-related conditions including liver fibrosis, cirrhosis, and hepatocellular carcinoma (HCC) , as well as to reduce the risk of vertical and horizontal transmission, improve quality of life, and reduce healthcare costs.

[0004] Without being held to theory or mechanism, chronic HBV infection involves a complex interplay between viral persistence and host immune responses. During chronic HBV infection, HBV antigens, including the HBV surface antigen (HBsAg) , are produced in large quantities (Kim, J. H., et al. Sci Rep 10, 1835 (2020) ) . Because they are produced in such large quantities, HBV antigens are believed to act as decoys to the immune system, leading to the exhaustion of B cells and T cells, and diverting the immune system away from effective antiviral responses that could otherwise neutralize the virus (Rydell, Gustaf E et al. Virology vol. 509 (2017) : 67-70) . In this way, chronic exposure to high levels of HBV antigens is believed to contribute to the exhaustion of HBV-specific immune responses, which makes treating chronic HBV infection particularly challenging. The efficacy of existing therapies can be transient and insufficient to treat chronic HBV. Accordingly, there exists a need for methods and dosing regimens useful for addressing the problem of transient efficacy and treating chronic HBV infections. Provided herein are dosing regimens and methods that address this need.SUMMARY

[0005] In one aspect, provided herein is a method of treating a hepatitis B virus (HBV) infection in a subject in need thereof comprising administering to the subject: an antisense oligonucleotide specific for an HBV target; and an HBV vaccine and / or an immunomodulator. In some embodiments, the HBV infection is a chronic HBV infection.
