USES OF HuR MOLECULAR GLUE DEGRADERS FOR THE TREATMENT OF BRAF-MUTANT TUMOR OR CANCERFIELD OF THE INVENTION

[0001] Provided herein is the use of HuR molecular glue degraders (MGD) for the treatment of BRAF-mutant tumors or cancers, particularly BRAF V600 mutant tumors. Also provided herein is the use of HuR MGD in combination with BRAF inhibitors for the treatment of BRAF-mutant tumors or cancers. These tumors include, but are not limited to, colorectal cancer (CRC) , melanoma, glioblastoma, breast carcinoma, high-grade spindle cell sarcoma, non-small cell lung cancer, and synovial sarcoma.BACKGROUND OF THE INVENTION

[0002] BRAF protein, a serine / threonine kinase in the RAF family, regulates the MAPK / ERK signaling pathway, which governs cell proliferation and differentiation. Mutations in the BRAF gene lead to constitutive activation of the MAPK / ERK signaling pathway, promoting uncontrolled cell proliferation, survival, and metastasis. More than 100 distinct BRAF mutations have been identified in cancers and are categorized into Class I, Class II and Class III mutations based on their dimerization dependency and RAS activation requirements. BRAF mutations, particularly the V600E mutation, are well-established oncogenic drivers in a variety of cancers, including CRC, melanoma, glioblastoma, breast carcinoma, high-grade spindle cell sarcoma, lung cancer, synovial sarcoma, and mutations may lead to unregulated activation of BRAF, which leads to activation of MEK and ERK and thus uncontrolled cell growth. Despite the development of BRAF inhibitors (e.g., vemurafenib, dabrafenib, encorafenib) and combination therapies targeting the MAPK pathway, those therapies show limited efficacy, and resistance mechanisms often emerge, limiting their long-term efficacy.

[0003] Therefore, there is an urgent need for novel therapeutic strategies that can overcome resistance and provide durable suppression of BRAF-driven tumor growth. BRIEF DESCRIPTION OF THE DRAWING

[0009] The inventors of the instant invention surprisingly and unexpectedly found that molecular glue degraders targeting HuR show improved efficacy in BRAF-mutant tumors or cancers, which exhibit marked sensitivity to this therapy regardless of tissue origin. The improved efficacy can be attributed to the mediation of HuR degradation, which disrupts BRAF RNA splicing and reduces BRAF protein stability, followed by the sustained decrease of MAPK pathway signaling.

[0010] In one aspect, provided herein is a method of treating a BRAF-mutant tumor or cancer in a subject in need thereof, comprising administering to the subject a therapeutically effective amount of a molecular glue degrader compound targeting HuR (also referred to as "HuR MGD compound" ) or a pharmaceutically acceptable salt thereof.
