[0001] HAIR TREATMENT COMPOSITION

[0002] FIELD OF THE INVENTION

[0003] The present invention relates to a hair treatment composition designed to repair and reduce damage to hair during and after chemical treatments. Specifically, it relates to a composition comprising encapsulated Alpha Lipoic Acid (ALA) or its derivatives, to effectively mitigate internal hair damage.

[0004] BACKGROUND OF THE INVENTION

[0005] Hair care treatments, including chemical styling, coloring, and bleaching, have become commonplace for consumers seeking to achieve specific hair aesthetics or to manage their hair’s appearance. However, these treatments, while effective at altering the appearance of the hair, often lead to undesirable side effects, especially when used repeatedly. One of the most significant issues that arise from such treatments is the alteration of the hair's natural structure, particularly the loss of keratin protein within the hair fibers.

[0006] Keratin, the key structural protein in hair, plays a vital role in maintaining the strength, elasticity, and overall integrity of the hair. Chemical processes such as reduction, oxidation, and lightening, typically used in styling, coloring, and bleaching, can disrupt the natural arrangement of keratin molecules, causing the proteins to degrade or be leached from the hair. As a result, hair becomes weak, brittle, and prone to breakage, split ends, and frizz.

[0007] Chemical styling treatments typically involve the use of harsh reduction agents that break down the hair’s disulfide bonds to allow for reshaping, while coloring and bleaching treatments employ oxidative agents that lift the natural color from the hair fibers and deposit artificial color. These oxidative processes, while crucial for changing the color or texture of the hair, also generate free radicals and other reactive species that can cause significant damage to the protein structure. As these proteins are leached from the hair, the hair’s overall strength and elasticity diminish, leading to visible signs of damage, such as dullness, thinning, and breakage.Research has consistently shown that the protein content of chemically treated hair is significantly lower than that of untreated (virgin) hair. This depletion of protein is a primary cause of hair’s weakened condition following chemical treatments. When protein is removed from the hair fiber, the natural protective barrier of the hair is compromised, and the hair becomes more vulnerable to further damage from environmental factors, including UV exposure, pollution, and mechanical stress. Given the damage inflicted during these treatments, there has been a continuous demand for hair treatment compositions that can help prevent or reduce protein loss, repair damaged hair, and restore hair’s strength and resilience. While traditional treatments have focused on replenishing proteins or moisturizing hair, these solutions often fall short in addressing the specific needs of chemically treated hair, which requires targeted, time- sensitive interventions.

[0008] A major challenge in developing effective formulations for this purpose lies in the need for the ingredients which would not interfere with the desired effects of the chemical treatments themselves. For example, any composition added to the hair during or after chemical treatment must not interfere with the deposition of hair color or disrupt the styling process. Moreover, the composition should ideally act quickly and effectively, without compromising the integrity of the treatment. T. Doering, et al in International Journal of Cosmetic Science (IFSCC), 10 (2007) (4) 323-329 reported a-lipoic acid (ALA) having anti-hair damage effect in a permanent hair dye formulation. Alpha lipoic acid significantly protected human hair during oxidation coloring as demonstrated by (i) amino acid and protein analysis, (ii) surface polarity measurement, (iii) lipid analysis, (iv) differential scanning calorimetry, and (v) combing work measurement. The presence of disulfide bonds appeared to be very favorable and the intramolecular cross-link within a-lipoic acid provided a kinetically controlled protection for cysteine crosslinks during oxidation coloring.

[0009] ALA is a potent antioxidant known for its ability to neutralize free radicals, which are produced during oxidative chemical processes such as coloring and bleaching.Free radicals are highly reactive molecules that cause oxidative damage to the hair fibers, leading to the loss of protein and weakening of the hair structure. By incorporating ALA into the composition, the invention harnesses its antioxidant properties to mitigate the damage caused by these free radicals. ALA is effective in neutralizing these reactive species, which would otherwise degrade the keratin structure of the hair.

[0010] However, despite its remarkable antioxidant properties, ALA incorporation in hair care formulations gets limited due to their insolubility in water, or requires number of additional agents for stabilizing the formulations, which further increases the cost of productions of the formulations. ALA itself is sensitive to oxidation and light, which may reduce its effectiveness over time.

[0011] Thus, there is need to fill this gap by offering an innovative solution that protects the hair from damage, enhances repair, and ensures that the desired chemical treatment effects are preserved, improving both the health and appearance of the hair.

[0012] Csaba-Pal Racz et al in J Incl Phenom Macrocycl Chem (2013) 76:193-199 reported inclusion of a-lipoic acid (LA) in P-cyclodextrin (P-CD) increases the aqueous solubility and photostability, which can enhance its medicinal use in the oral administration. Different preparation methods involving 206 mg of LA and 1,294 mg of P-CD were employed to obtain an a-lipoic acid-P-cyclodextrin (LA-P-CD) inclusion complex. The crystalline structure of the compound belongs to the monoclinic system with four molecules in the unit cell.

[0013] Asli Celebioglu and Tamer Uyar in J. Agric. Food Chem (2019), 10.1021 / acs.jafc.9b05580 reported nanofibers from alpha-lipoic acid / cyclodextrin inclusion complex for enhancing water solubility and thermal stability of alpha-lipoic acid. Hydroxypropyl-beta-cyclodextrin (HP-P-CyD) and hydroxypropyl-gamma-cyclodextrin (HP-y-CyD) were chosen as host molecules for forming inclusion complexation with a-LA. Accordingly, a-LA was inclusion complexed with HP-P-CyD and HP-y-CyD by using very high concentrated aqueous solutionsof CyD (200%, w / v) having 1 / 1 and 2 / 1 molar ratio of a-LA / CyD. Except a-LA / HP-P-CyD (1 / 1) solution, other a-LA / CyD solutions were turbid indicating the presence of some non complexed a-LA whereas a-LA / HP-P-CyD (1 / 1) solution was very homogeneous signifying that a-LA was fully complexed with HP-P-CyD. Even so, electrospinning was performed for all of the a-LA / HP-P-CyD (1 / 1 and 2 / 1) and a-LA / HP-y-CyD (1 / 1 and 2 / 1) aqueous solutions, and defect-free beadless and uniform nanofibers were successfully obtained for all of the a-LA / CyD solutions. The better encapsulation efficiency of a-LA was obtained in a-LA / HP-P-CyD nanofibers. a-LA / CyD nanofibers showed significantly higher antioxidant performance along with enhanced thermal stability compared to pure a-LA. Such promising results supported that antioxidant a-LA / CyD nanofibers may have potential applications as orally fast-dissolving food supplements.

[0014] US20100247504A1 and US8512758B2 discloses use of thioctic acid (a-lipoic acid) with cyclodextrin as an antioxidant complex in a powder composition, for prevention of reaction induced by the contact with the other antioxidant substances present in the biological system. The cyclodextrin-included thioctic acid was prepared in US 20100247504 Al with 55 g of cyclodextrin powder, 20 g of starch syrup, and 20 g of thioctic acid. The antioxidant complex does not react with other anti-oxidants such as L-ascorbic acid, copper gluconate in a composition, and shows formation of homogeneous dispersion. The prior art document provides a powder composition comprising antioxidant that can be used as cosmetics for hair washing.

[0015] Cyclodextrins are cyclic oligosaccharides that are capable of forming inclusion complexes with ALA, at varied stoichiometric proportion. However, not all the proportions of a-lipoic acid-cyclodextrin inclusion complex are compatible with components of hair care formulations, particularly those containing surfactants and oxidizing agents and preserve the integrity and aesthetic outcomes of the hair styling treatments, and yet prevent the weakening of hairs during and after such treatments.Considering the solubility aspect of the a-lipoic acid-cyclodextrin inclusion complex there is required to develop stable hair care formulations comprising a-lipoic acid-cyclodextrin inclusion complex that can preserve the integrity and aesthetic outcomes of the hair styling treatments, and yet prevent the weakening of hairs during and after such treatments. The ability to protect and repair hair without altering the efficacy of the chemical treatment process is a critical factor for consumer satisfaction and long-term hair health.

[0016] OBJECTS OF THE INVENTION

[0017] Accordingly, the primary object of the present invention is to provide a hair treatment composition that effectively reduces protein loss and prevents damage to hair fibers during and after chemical treatments, such as styling, coloring, bleaching, and perming.

[0018] It is further an object of the invention to ensure that the hair treatment composition preserves the structural integrity and strength of the hair, minimizing brittleness, breakage, and split ends, while allowing chemical treatments such as color deposition, lightening, and styling to proceed without disruption.

[0019] It is also an object of the invention to provide a composition that can be applied as a standalone treatment or in combination with chemical treatments, such as during, before, or after the treatment process, to continuously protect and repair the hair, offering lasting benefits.

[0030] The use of “comprise”, “comprises”, “comprising”, “contain”, “contains”, “containing”, “include”, “includes”, and “including” are not intended to be limiting. It is to be understood that both the foregoing general description and this detailed description are exemplary and explanatory only and are not restrictive. The present invention relates to a hair treatment composition that is specifically designed to reduce protein loss and protect the hair from damage during and after chemical treatments such as styling, coloring, bleaching, and perming. These chemical treatments, while effective for altering the appearance of hair, often lead to the degradation of the hair's keratin proteins, causing it to become brittle, weak, and prone to breakage.

[0031] In an embodiment there is provided a hair treatment composition comprising an a-lipoic acid-cyclodextrin inclusion complex for providing targeted protection and repair of the keratin protein in the hair.

[0032] The present invention provides an a-lipoic acid-cyclodextrin inclusion complex containing Alpha Lipoic Acid (ALA) or its derivatives and cyclodextrin derivative, at a specific mole ratio of 1:4, in a hair treatment composition for enhancing hair strength, even after multiple chemical treatments.

[0033] The a-lipoic acid derivative of the hair treatment composition is selected from the group comprising alkyl, branched alkyl, hydroxy-substituted alkyl, or unsaturated fatty alcohol esters of a-lipoic acid, including methyl, ethyl, n-propyl, isopropyl, n-butyl, 2-ethylhexyl, n-hexadecyl, n-octadecyl, oleyl, linolyl, linolenyl, gadoleyl, and behenyl a-lipoate.

[0034] The cyclodextrin derivative of the hair treatment composition is selected from the group comprising 2,6-dimethyl-beta-cyclodextrin, hydroxyethyl-beta-cyclodextrin, hydroxyethyl-gamma-cyclodextrin, hydroxypropyl-beta-cyclodextrin, or hydroxy propyl-gamma-cyclodextrin.

[0035] In a preferred embodiment the cyclodextrin derivative is P-cyclodextrin (P-CD). The present hair composition comprising the a-lipoic acid-cyclodextrin inclusion complex provides a stable, effective, and targeted treatment.

[0036] The encapsulation of ALA in cyclodextrin enhances the stability of ALA by protecting it from environmental factors such as air and light, thus ensuring its effectiveness during chemical treatments. Additionally, encapsulating ALA withincyclodextrins helps in increasing its solubility and bioavailability, allowing for better incorporation into the hair fibers.

[0037] The use of cyclodextrin derivatives provides additional benefits by enabling a controlled release of ALA. Once applied to the hair, the encapsulated ALA is gradually released in a sustained manner, offering continuous protection against oxidative damage during and after chemical treatments. This controlled release ensures that the active ingredient is delivered to the areas of the hair most in need of repair and protection, such as those exposed to oxidative stress from chemical processes.

[0038] In an embodiment there is provided a hair treatment composition, comprising encapsulated a-lipoic acid-cyclodextrin inclusion complex, at an effective concentration of 0.1-2.0 wt% in the composition.

[0039] The present encapsulated a-lipoic acid-cyclodextrin inclusion complex provides tensile strength enhancement in hair fibers, when incorporated in a hair treatment composition at a concentration of 0.1-2.0 wt%.

[0040] The inventors of the present invention observed that concentrations below the range 0.1-2.0 wt% fails to sufficiently mitigate oxidative protein degradation, while concentrations above this range result in diminished mechanical reinforcement due to reduced bioavailability and aggregation effects.

[0041] The present ALA-cyclodextrin encapsulation complex encapsulated at a specific molar ratio of 1:4 and at an effective concentration in a hair treatment composition exhibits an unexpected and non-linear improvement in tensile strength.

[0042] The invention offers significant advantages over conventional hair treatment compositions, particularly those designed to address the issues of chemical treatment-induced damage.

[0043] Traditional treatments typically rely on surface-level moisturization or the replenishment of proteins to restore hair strength, but they often fail to provide sufficient protection during the chemical process itself. In contrast, the present hairtreatment composition comprising a-lipoic acid-cyclodextrin inclusion complex works proactively to protect the hair from the inside out by neutralizing oxidative damage as it occurs, thereby reducing protein loss. By preserving the integrity of the keratin proteins within the hair, the composition helps to maintain the hair’s elasticity, resilience, and overall strength, ultimately preventing the common issues of hair brittleness, breakage, and split ends.

[0044] The present invention provides a hair treatment composition which can be applied at various stages of the chemical treatment process. Before a chemical treatment, it can act as a protective pre-treatment, forming a barrier that helps to prevent damage from occurring. During a chemical treatment, such as while the hair is being colored or bleached, the composition can be integrated into the treatment mixture to provide continuous protection against oxidative stress. After the treatment, it can be used as a restorative leave-in treatment to repair and strengthen the hair that has been exposed to damaging chemicals. Whether used as a standalone treatment or in conjunction with other hair care products, the composition is formulated to be highly compatible with a wide range of chemical treatments, including hair coloring, bleaching, perming, straightening, and styling.

[0069] Example 1: Preparation of hair treatment composition containing encapsulated Alpha Lipoic Acid (ALA) or its derivatives in cyclodextrin derivative

[0070] The a-lipoic acid-cyclodextrin complex containing a-lipoic acid to cyclodextrin at 1:4 molar ratio was selected because it provides a stable inclusion complex suitable for incorporation into hair care formulations, particularly those containing surfactants and oxidizing agents. The a-lipoic acid-cyclodextrin inclusion complexes prepared at ratios other than 1:4 ratio exhibit stability issues. Accordingly, the present invention utilizes a pre-formed inclusion complex having a ratio selected to provide adequate stability. Table 1 shows the hair treatment composition details:

[0073] Example 2: Preparation of hair care formulation containing a-lipoic acid-cyclodextrin inclusion complex

[0074] Hair care formulations comprising the a-lipoic acid-cyclodextrin Alpha Lipoic Acid (ALA) inclusion complex at varied concentration were prepared.

[0075] The a-lipoic acid-cyclodextrin Alpha Lipoic Acid (ALA) inclusion complex were provided at a varied concentration ranges in control (0%), Fl (0.1%), F2(l%), F3(2%), F4(5%), and F5(10%) in a hair care formulation.Table 2 shows effect of different concentrations of the hair treatment composition on tensile strength of hair strands.

[0076] Table 2: Effect of wt% of complex on Tensile Strength

[0077]

[0078] The data demonstrates that tensile strength enhancement in hair fibres is critically dependent on a concentration window of 0.10-10 wt% of a-lipoic acid-cyclodextrin Alpha Lipoic Acid (ALA) inclusion complex. Concentrations below this range fail to sufficiently mitigate oxidative protein degradation, while concentrations above this range result in diminished mechanical reinforcement due to reduced bioavailability and aggregation effects. The inventive combination therefore exhibits an unexpected and non-linear improvement in tensile strength.
