Abstract
Description
An NMT inhibitor, its antibody-drug conjugate, and pharmaceutical uses
[0001] Cross-reference to related applications
[0002] This application claims priority to Chinese Patent Application No. CN202510501070.3, filed on April 18, 2025, entitled “An NMT Inhibitor and Its Antibody-Drug Conjugate and Pharmaceutical Use”, the contents of which are incorporated herein by reference in their entirety. Technical Field
[0003] This application relates to the field of pharmaceutical technology, specifically to an NMT inhibitor, its antibody-drug conjugate, and its pharmaceutical uses. Background Technology
[0004] Myristyltransferase (NMT) catalyzes the irreversible co-translational transfer of myristic acid from myristoyl-CoA to a protein substrate containing an N-terminal glycine. N-Myristylation is an important protein-lipid modification and co-translational modification process in eukaryotes (fungi and protists), playing a crucial role in cellular processes such as signal cascade transduction, vesicle transport, and protein localization. Higher organisms, such as humans, have two types of NMT (NMT1 and NMT2), widely expressed in various tissues, with potential substrates distributed across different cellular locations and possessing diverse biological functions. NMT is involved in the development and progression of various diseases, such as cancer, epilepsy, Alzheimer's disease, and viral, bacterial, and parasitic infections.
[0005] NMT inhibitors have been extensively studied for their application in treating microbial infections and proliferative disorders. In animal models, NMT inhibitors selectively kill cancer cells in vitro, induce regression of hematologic malignancies, and inhibit the growth of lung and breast cancer. In acute myeloid leukemia models, NMT inhibitors kill leukemia stem cell populations and promote the regeneration and growth of normal bone marrow cells. Myricx Bio has achieved complete tumor regression in multiple tumor models by using NMT inhibitors as the payload of its ADC (anti-tumor agonist) drugs.
[0006] Antibody-drug conjugates (ADCs) are therapeutic agents composed of an antibody that specifically targets a related antigen and a small molecule toxin conjugated via a linker. They combine the potent killing effect of small molecule chemotherapeutic agents with the tumor-targeting properties of antibody drugs. After entering the body, ADC molecules bind to antigens on the surface of target cells through antibody guidance, releasing toxin molecules via chemical or enzymatic reactions to destroy the target cells. Due to their targeting and intracellular drug release, ADCs can precisely bind to tumors and reduce the impact on normal cells, increasing local drug concentration, reducing toxic side effects on normal cells, and expanding the therapeutic window. ADCs have broad application prospects in cancer treatment and have achieved some encouraging results in clinical research.
[0007] Currently, there are no NMT inhibitor drugs on the market. Therefore, it is still necessary to develop new compounds with better efficacy, pharmacokinetic behavior, and reduced toxicity, or to use them as payloads for ADC molecules to improve efficacy and safety in vivo. Summary of the Invention
[0008] In order to achieve the purpose of this invention, this application provides an NMT inhibitor and its antibody-drug conjugate and pharmaceutical use. The NMT inhibitor and its antibody-drug conjugate described in this application have good anticancer activity.
[0009] In a first aspect, this application provides an NMT inhibitor, which is a compound of formula (I), or an isotopically labeled compound thereof, or an optical isomer, geometric isomer, tautomer, or mixture of isomers thereof, or a pharmaceutically acceptable salt thereof.

[0168] The term "linker group" refers to a chemical structural fragment or bond that is linked to an antibody at one end and to a toxin drug at the other end. It can be linked to other linker groups before being linked to an antibody (or antigen-binding fragment) or a toxin drug.
[0169] The term "antibody drug conjugate" (ADC) refers to an antibody or antigen-binding fragment linked to a biologically active toxic drug, such as an NMT inhibitor, via a stable linker unit.
[0170] In this application, the term "antibody" refers to immunoglobulin, which is a tetrapeptide chain structure composed of two identical heavy chains and two identical light chains linked by interchain disulfide bonds. The amino acid composition and sequence of the constant region of the immunoglobulin heavy chain differ, thus their antigenicity also differs. Based on this, immunoglobulins can be divided into five classes, or isotypes of immunoglobulins: IgM, IgD, IgG, IgA, and IgE, with their corresponding heavy chains being μ, δ, γ, α, and ε chains, respectively. Within the same class of Ig, based on differences in the amino acid composition of its hinge region and the number and position of disulfide bonds in the heavy chain, different subclasses can be further divided; for example, IgG can be divided into IgG1, IgG2, IgG3, and IgG4. The light chains are classified as κ or λ chains based on differences in the constant region. Each of the five classes of Ig can have either a κ or λ chain. The sequence of approximately 110 amino acids near the N-terminus of the antibody heavy and light chains varies considerably, forming the variable region (Fv region); the remaining amino acid sequence near the C-terminus is relatively stable, forming the constant region. The variable region comprises three hypervariable regions (HVRs) and four relatively conserved backbone regions (FRs). The three hypervariable regions determine the antibody's specificity and are also known as complementarity-determining regions (CDRs). Each light chain variable region (LCVR) and heavy chain variable region (HCVR) consists of three CDRs and four FRs, arranged in the following order from the amino terminus to the carboxyl terminus: FR1, CDR1, FR2, CDR2, FR3, CDR3, FR4. The three CDRs of the light chain refer to LCDR1, LCDR2, and LCDR3; the three CDRs of the heavy chain refer to HCDR1, HCDR2, and HCDR3. The CDR amino acid residues in the LCVR and HCVR regions of the antibody or antigen-binding fragment described in this application conform to the known Kabat numbering rules (LCDR1-3, HCDR2-3) in number and position, or conform to the Kabat and Chothia numbering rules (HCDR1).
[0171] The term "antigen-binding fragment" refers to one or more fragments of an antibody that maintain the ability to specifically bind to an antigen. It has been shown that fragments of full-length antibodies can be used for antigen-binding function. Examples of binding fragments included in "antigen-binding fragments" include: (i) Fab fragments, monovalent fragments consisting of VL, VH, CL, and CH domains; (ii) F(ab')2 fragments, bivalent fragments comprising two Fab fragments linked by disulfide bridges on the chain region; (iii) F fragments consisting of VH and CH domains; (iv) Fv fragments consisting of VH and VL domains of a single arm of the antibody; (v) single-domain or dAb fragments consisting of a VH domain; and (vi) separate complementarity-determining regions (CDRs) or (vii) combinations of two or more separate CDRs optionally linked by synthetic linkers. Furthermore, although the two domains VL and VH of the Fv fragment are encoded by separate genes, they can be linked by synthetic linkers using recombinant methods, thereby enabling the production of a single protein chain (called a single-chain Fv (scFv)) in which the VL and VH regions pair to form a monovalent molecule. Such single-chain antibodies are also included in the term "antigen-binding fragment." Such antibody fragments are obtained using conventional techniques known to those skilled in the art, and fragments are screened for functionality in the same manner as for intact antibodies. Antigen-binding moieties can be generated by recombinant DNA technology or by enzymatic or chemical cleavage of intact immunoglobulins. Antibodies can be different types of antibodies, such as IgG (e.g., IgG1, IgG2, IgG3, or IgG4 subtypes), IgA1, IgA2, IgD, IgE, or IgM antibodies.

[0194] In addition, this application may use protecting groups to protect certain functional groups from unwanted reactions. Protecting groups suitable for various functional groups and their protection or deprotection conditions are well known to those skilled in the art. For example, TW Greene and GMWuts' "Protecting Groups in Organic Preparations" (3rd edition, Wiley, New York, 1999 and cited references in the book) describes in detail a large number of protecting or deprotecting groups.
[0195] The separation and purification of compounds and intermediates employ appropriate methods and procedures depending on specific needs, such as filtration, extraction, distillation, crystallization, column chromatography, preparative thin-layer chromatography, preparative high-performance liquid chromatography, or a combination of the above methods. Specific methods can be found in the examples described in this application. Of course, other similar separation and purification techniques can also be used. Conventional methods (including physical constants and spectroscopic data) can be used for characterization.

Then, a solution of dichloroethane (21 mL) containing compound 7 (2.10 g, 10.2 mmol, 1.00 eq) and 2,4,6-trimethylpyridine (4.75 g, 39.2 mmol, 5.18 mL, 3.85 eq) was added dropwise. After the addition was complete, the temperature was raised to 25 °C and stirred. The temperature was then raised to 90 °C and stirred for 15 h. The reaction was monitored by LC-MS. The reaction mixture was cooled to 25 °C, purified water (21 mL) was added, and the temperature was raised to 100 °C and stirred for 2 h. The reaction solution was cooled to 25°C, and the reaction was quenched with purified water (20 mL). The solution was then diluted with dichloromethane (60 mL). The organic phase was separated, washed with saturated citric acid aqueous solution (60 mL × 3), and dried over anhydrous sodium sulfate.
