Abstract
Description
A guaiacane-type sesquiterpene lactone pseudonatural product, its preparation method and uses Technical Field
[0001] This invention belongs to the field of pharmaceutical technology and relates to the preparation of guaiacol-type sesquiterpene lactone pseudo-natural products and their derivatives by chemical synthesis using (R)-Carvone (i.e., L-carvone) as the starting material, and the testing of the anti-inflammatory activity of these compounds on an established erythrocyte hemolysis model. Background Technology
[0002] The NLRP3 inflammasome, an organelle structure first discovered and reported in 2002 by the Biomedical Research Center at the University of Lausanne, Switzerland, consists of three parts: the NLRP3 scaffold, the adaptor protein ASC, and Caspase-1. Studies have shown that when the body responds to pathogen infection or danger-related signals, the NLRP3 inflammasome assembles and activates in the cytoplasm, leading to the activation and secretion of pro-inflammatory factors IL-1β and IL-18, thus playing an important role in innate immune-mediated inflammation. Furthermore, the NLRP3 inflammasome is closely related to the development of many major human diseases, including type 2 diabetes (T2D), cyropyrin-associated periodic syndrome (CAPS), gout, multiple sclerosis, Alzheimer's disease (AD), Parkinson's disease (PD), and inflammatory bowel disease (IBD). Therefore, NLRP3 is also an important target for the development of drugs to treat inflammation-related diseases; however, no drugs have been reported since its discovery. Taking IBD as an example, the main clinical drugs used to treat IBD are steroidal anti-inflammatory drugs such as glucocorticoids, nonsteroidal anti-inflammatory drugs (NSAIDs) such as ibuprofen, and immunosuppressants such as azathioprine. In addition, biological agents such as TNF-α monoclonal antibodies can also be used for treatment.
[0003] Sesquiterpene lactones exhibit significant anti-inflammatory potential by targeting and inhibiting various key pro-inflammatory molecules involved in different inflammatory pathways and preventing or reducing inflammatory damage to tissues. Therefore, sesquiterpene lactones are an important source of novel anti-inflammatory drugs. However, for known guaiacol-type sesquiterpene lactones, anti-inflammatory activity has only been verified in a few synthesized derivatives (see: CN117624098A, CN113968869A, etc.), and no novel parent or core structures of guaiacol-type sesquiterpene lactones that can meet the needs of large-scale drug molecule synthesis and screening have yet been discovered.
[0004] Peudo-natural products (PNPs) are a concept first proposed in 2018 by the Max Planck Institute for Molecular Physiology in Germany as part of a natural product chemistry optimization strategy. Currently, there are no research reports on the use of guaiacane-type sesquiterpene lactone pseudo-natural products and their derivatives as anti-inflammatory drugs, especially as anti-inflammatory drugs specifically targeting the NLRP3 inflammasome. [0083] Referring to Figures 1-1, 1-2, and 1-3, this invention uses (R)-Carvone (i.e., compound 1) and propynyl alcohol (i.e., compound 3a) and butynyl alcohol (i.e., compound 3b) as raw materials, and employs a designed series of synthetic routes. First, important intermediates (i.e., compounds 2, 6a, and 6b; where compounds 6a and 6b are obtained by nucleophilic substitution of propynyl alcohol and butynyl alcohol with protected hydroxyl groups in the presence of n-butyllithium, followed by electrophilic substitution with diisobutylaluminum hydride and iodoborane pinacol ester). Then, using these important intermediates (specifically, by performing tandem lactone reactions of compounds 2 and 6a, and compounds 2 and 6b, respectively, in the presence of boron trifluoride diethyl ether, followed by removal of the corresponding hydroxyl protecting groups from compounds 6a and 6b), novel 1,10-secreting guaiacol sesquiterpene lactones (see compound 8a, compound 6b) are prepared. Compound 8b was used to prepare novel guaiacol-type sesquiterpene lactones (specifically, by using compound 8a for Sven oxidation, reacting the product with vinyl magnesium bromide followed by catalytic cyclization using a Grubb second-generation catalyst, and by using compound 8b in the presence of Desmartin oxidant followed by Wittig reaction, and then catalytic cyclization using a Grubb second-generation catalyst). This led to the preparation of guaiacol-type sesquiterpene lactone pseudo-natural products (including: preparing compound I-3 from compound 20 and subsequently compound I-1, and preparing compound I-2 from compound 12) and the preparation of derivatives of these guaiacol-type sesquiterpene lactone pseudo-natural products (including: preparing compound II from compound I-1, preparing compound III from compound 12, and preparing compound V from compound I-3). The specific synthesis of compounds 12, 20, etc., is described below.
[0084] 1.1 Synthesis of Compound 2
[0085] According to the method reported in the literature (Reference: Elford TG, Hall D G. Total synthesis of (+)-chinensiolide B via tandem allylboration / lactonization. Journal of the American Chemical Society. 2010, 132(5): 1488-1489.), using (R)-Carvone (i.e., compound 1) as the raw material, cyclopentane-1-carboxaldehyde compounds (specifically compound 2) were obtained through 6-step transformation. 1 ¹H NMR (400MHz, CDCl₃) δ 9.52 (d, J = 3.8 Hz, 1H), 4.88–4.66 (m, 2H), 4.18 (t, J = 3.7 Hz, 1H), 3.60 (s, 1H), 3.30 (t, J = 10.9 Hz, 1H), 2.61 (m, 1H), 2.34–2.20 (m, 1H), 1.92–1.78 (m, 1H), 1.72 (d, J = 1.5 Hz, 3H), 0.98 (d, J = 6.8 Hz, 3H), 0.89 (d, J = 2.3 Hz, 9H), 0.05 (d, J = 4.8 Hz, 6H). [0086] 1.2 Synthesis of Compound 6a
[0087] (1) Add 110 mL of THF, 5 g of propynyl alcohol (i.e., compound 3a; 89.18 mmol, 1.0 equiv), and 15.49 g of imidazole (222.96 mmol, 2.5 equiv) to a 250 mL reaction flask containing a magnetic stirrer. Then, slowly add 28.32 mL of tert-butyldiphenylchlorosilane (TBDPSCl; 107.02 mmol, 1.2 equiv) dropwise to the reaction flask using a syringe over a period of approximately 25 minutes. Continue stirring at room temperature for 1.5 h. After confirming the complete reaction of the reactants by TLC, add 30 mL of saturated NH4Cl solution to terminate the reaction. Then, add 30 mL of water and extract with ethyl acetate (3 × 30 mL). Combine the organic phases and wash with 50 mL of saturated brine. Dry with anhydrous Na2SO4, filter, and concentrate the filtrate under reduced pressure to obtain the crude product. The crude product was purified by silica gel column chromatography with elution conditions of PE:EA = 50:1, and finally 21.96 g of colorless oil (i.e. compound 4a) was obtained. 1H NMR (400MHz, CDCl3) δ7.72 (d, J = 7.8 Hz, 4H), 7.48–7.35 (m, 6H), 4.33 (d, J = 2.4 Hz, 2H), 2.38 (t, J = 2.4 Hz, 1H), 1.08 (s, 9H).
[0088] (2) Under a nitrogen (N2) atmosphere and at -78°C, 254 mL of anhydrous THF and 15 g of compound 4a (50.94 mmol, 1.0 equiv.) were added to a 1 L round-bottom flask containing a magnetic stirrer. Then, 35.02 mL of a 1.6 M n-butyllithium hexane solution (containing 56.03 mmol, 1.1 equiv. of n-butyllithium) was slowly added dropwise to the flask over approximately 20 minutes. The temperature was then maintained, and stirring continued for 1 hour. Next, at -78°C, 11.93 m... Methyl chloroformate (152.82 mmol, 3 equiv.) was slowly added dropwise to the round-bottom flask using a syringe, and stirring was continued for 1.0 h. The flask was then slowly brought to room temperature. After confirming complete reaction of the reactants by TLC, 30 mL of saturated NaHCO3 solution was added to terminate the reaction. The mixture was then extracted with ethyl acetate (3 × 40 mL), and the organic phases were combined and washed with 80 mL of saturated brine. The mixture was dried over anhydrous Na2SO4, filtered, and the filtrate was concentrated under reduced pressure to obtain the crude product. The crude product was purified by silica gel column chromatography under elution conditions of PE:EA = 40:1, ultimately yielding 7.64 g of a colorless oil (compound 5a). 1 H NMR (400MHz, CDCl3) δ7.67 (d, J = 6.6 Hz, 4H), 7.46–7.37 (m, 6H), 3.83 (d, J = 7.1 Hz, 2H), 3.76 (s, 3H), 2.58 (t, J = 6.8 Hz, 2H), 1.06 (s, 9H).

Then, 21.67 mL of a 1 M diisobutylaluminum hydride (DIBAL-H) n-hexane solution (containing 32.51 mmol, 1.5 equiv. of DIBAL-H) was slowly added dropwise to the round-bottom flask over a period of approximately 15 min. After that, 7.64 g of compound 5a (21.67 mmol, 1.0 equiv.) was added to the round-bottom flask all at once, and stirring continued. After stirring for 5 hours, 8.06 mL of iodomethylpinacol boronic acid ester (43.35 mmol, 2.0 equiv.) was added to the round-bottom flask via syringe. The mixture was stirred for 30 minutes, then transferred to room temperature and stirred overnight while monitoring by TLC. After confirming the complete reaction of the starting material, 20 mL of 1M HCl solution was added to the round-bottom flask to terminate the reaction. Then, 40 mL of water was added, followed by extraction with ethyl acetate (3 × 40 mL). [0141] Small molecule compounds at different concentrations (0 μM, 2 μM, 4 μM, 8 μM, 16 μM) were co-incubated with erythrocytes at 37°C for 30 min. The small molecule compounds were I-1, I-2, I-3, II-9, III-12, V-13, or MCC950 (as a positive control). Subsequently, nigericin at a final concentration of 1 μM was added for 50 min (i.e., the N+0 μM, N+2 μM, N+4 μM, N+8 μM, and N+16 μM groups in Table 1). In the model group (i.e., the N+0 μM group in Table 1), nigericin induced pyroptosis, leading to cell death and hemoglobin efflux. After treatment, hemoglobin levels were quantified using a kit. The activity of the small molecule compounds was tested based on the direct proportionality between hemoglobin content and absorbance value (OD505).
[0142] Table 1. Experimental Groups and Treatments
[0143] Note: The total volume of each group in Table 1 is 200 μL. N represents Nigericin, and the final concentration of Nigericin in the system is 1 μM. X represents the volume of drug solution of I-1, I-2, I-3, II-9, III-12, V-13 or MCC950 (the solvent for preparing the drug solution is 0.9% physiological saline). NS represents 0.9% physiological saline.
[0144] 6.3 Specific Experimental Procedures
[0145] S1 adds 1 mL of physiological saline (0.9% NaCl) to a 1.5 mL EP tube.
[0146] S2 aspirates 100 μL of lower red blood cells (i.e., red blood cells separated from blood) and adds them to an EP tube containing 1.5 mL of physiological saline.
[0147] S3 was centrifuged at 3000 r / min for 5 min.
[0148] After centrifugation at S4, discard 1 mL of supernatant, add 1 mL of physiological saline, and centrifuge again at 3000 r / min for 5 min.
[0149] After centrifugation at S5, discard the supernatant; the remaining cells are the required red blood cells (this completes the washing of red blood cells).
[0150] S6 prepares suspensions of physiological saline and red blood cells according to the 7 regimens in Table 1.
[0151] S7. The small molecule compound to be tested was added to the corresponding group of red blood cell suspensions at concentrations of 0 μL, 32 μL, 0 μL, 4 μL, 8 μL, 16 μL, and 32 μL (so that the final concentrations of the small molecule compound in the Control, 16 μM, N+0 μM, N+2 μM, N+4 μM, N+8 μM, and N+16 μM groups are 0 μM, 16 μM, 0 μM, 2 μM, 4 μM, 8 μM, and 16 μM, respectively) and incubated at 37 °C for 30 min.
[0152] After S8 incubation, 4 μL of Nigericin was added to the N+0 μM, N+2 μM, N+4 μM, N+8 μM, and N+16 μM groups, and incubated at 37 °C for 50 min.
[0153] After S9 incubation is complete, centrifuge at 2000 r / min for 3 min.
[0154] S10 aspirates the supernatant and sets it aside for later use.
[0155] S11 uses a plasma free hemoglobin assay kit to detect hemoglobin in the supernatant (detection conditions: react at 37°C for 20 min after adding reagents).
[0156] After the S12 reaction was completed, the absorbance value at a wavelength of 505 nm was measured using an ELISA reader.
[0157] 6.4 Experimental Results
[0158] As shown in Figure 8, the experimental results indicate that, using the release of hemoglobin from erythrocytes stimulated with Nigericin after 30 min of co-incubation with MCC950 as a reference, compounds I-2, I-3, III-12, and V-13 all exhibited significant inhibitory effects on erythrocyte hemolysis induced by Nigericin stimulation at treatment concentrations of 2 μM and 4 μM, and their inhibitory effects were significantly better than those of the positive control drug MCC950. Compounds I-2 and III-12 showed the best inhibitory effects at a treatment concentration of 4 μM. These results suggest that guaiacol-type sesquiterpene lactones (or guaiacol-type sesquiterpene lactone pseudo-natural products), represented by compounds I-2 and I-3, and their derivatives, have a certain inhibitory effect on erythrocyte hemolysis and could serve as candidate drug molecules for the prevention and / or treatment of NLRP3 inflammasome-related diseases.
[0159] The results of the parallel experiments in the Control group and the N+0μM group in Figure 8 fluctuated (OD505 values were high and low), which may be due to the difference in red blood cells used when conducting experiments on different small molecule compounds.
[0160] In summary, based on the synthesis of guaiacol-type sesquiterpene lactone pseudo-natural products and their derivatives, this invention has discovered novel guaiacol-type sesquiterpene lactone compounds that have a significant inhibitory effect on erythrocyte hemolysis. These compounds have high application value in the development of anti-inflammatory drugs (especially anti-inflammatory drugs for the prevention and / or treatment of NLRP3-related inflammatory diseases such as inflammatory bowel disease) (e.g., large-scale synthesis and screening of drug molecules with anti-inflammatory activity, and their use as active ingredients in the preparation of anti-inflammatory drugs).
