[0003] A PERSONAL CARE OR PHARMACEUTICAL COMPOSITION
[0004] Field of the Invention
[0005] The present invention relates to a pharmaceutical or personal care composition comprising 5 acetyl amino acid compound and vitamin C compound, wherein the molar ratio of the acetyl amino acid compound to the vitamin C compound is at least 7:1. It was unexpectedly found that such composition is capable of significantly upregulating the expression of COL17A1 (Collagen Type XVII Alpha 1 Chain) gene.
[0006] Background of the Invention
[0007] The skin is a primary barrier of the human body. It protects the organs in the body from external stimulations. It is subject to intrinsic aging and extrinsic aging. Skin aging is a complex process, and alterations in human skin due to aging have distinct characteristics as compared to other organs. Characteristics of intrinsic or chronological skin aging include dryness, visible free lines 15 and wrinkles, uneven skin pigmentation, loss of elasticity and skin sagging. Extrinsic factors including exposure to sunlight, pollutants and cigarette smoke can accelerate the skin aging process, especially on the face.
[0008] As humans age, their skin becomes thinner, more fragile, drier, shows abnormal pigmentation, 20 and heals wounds more slowly. The fragility of human skin during aging is partly due to the remodeling of the dermal-epidermal junction, particularly the hemidesmosomes component. Collagen XVII (COL17), a transmembrane protein in hemidesmosomes, regulates skin homeostasis, aging, and wound repair by mediating stem cell interactions with surrounding cells and the matrix. Therefore, Promoting the production of collagen XVII through upregulating the expression of COL17A1 (Collagen Type XVII Alpha 1 Chain) gene is an efficient way of providing anti-aging, and / or wound repair benefit.
[0009] Therefore, the present inventors have recognized that there is a need to develop solution to enhance the expression of COL17A1 gene. It was surprisingly found that by combining acetyl 30 amino acid compound and vitamin C compound, with a molar ratio of the acetyl amino acid compound to the vitamin C compound being at least 7:1, the expression of COL17A1 gene was significantly upregulated.P0001265W01 CPL

Gene expression of COL17A1 gene was analyzed by polymerase chain reaction (PCR) using SYBR® Green Premix Pro Taq HS qPCR Kit (Accurate Biotechnology, AG11701). The actin beta (ACTS) gene was selected as the housekeeping gene and all data on relative expression of the target genes was normalized to ACTB gene. The fold changes in expression were calculated relative to the blank control (medium having no active). All tests were conducted at least three times and Table 1 shows the fold change of the gene expression for COL17A1.
