Abstract
Description
[0001] PWHELOOIWO / 16. 04 . 2026 1 2026048716
[0002] Compositions containing Q-5 fatty acids, tocotrienol and fucoxanthin
[0003] The invention relates to compositions containing Q-5 fatty acids, tocotrienol and fucoxanthin.
[0004] Prior art includes formulations containing plant-based ingredients such as fucoxanthin, which offer metabolic benefits and reduce cellular inflammation (e.g., US Patent 11878007, Red Mountain Red Spa, LLC, January 23, 2024). Such formulations often contain a variety of ingredients with health-promoting effects. However, these prior art formulations are also associated with many problems. For example, one issue is that such formulations only treat health problems symptomatically, merely suppressing pain and inflammation. The health problems can usually only be controlled temporarily, and the symptoms often reappear after discontinuing the formulations. Therefore, continuous use of these formulations is often necessary.Such formulations can be associated with side effects and intolerances, such as increased susceptibility to infection, organ damage, or hormonal imbalances. These side effects can be particularly pronounced with long-term use of the formulation. Furthermore, existing formulations often do not offer a holistic approach to improving health, but rather focus primarily on individual aspects of health problems, which also does not contribute to a sustainable solution. Additional problems with current technology arise from the lack of efficacy of existing formulations. Improved formulations are therefore necessary. PWHELOOIWO / 16.04.2026 2 2026048716.
[0005] The object of the present invention is to overcome at least some, if not all, of the aforementioned disadvantages of the prior art.
[0006] The problem is solved by the compositions according to the independent claims. Advantageous embodiments and further developments are the subject of the dependent claims.
[0007] A first aspect of the invention relates to a composition with ingredients from at least two, preferably all, of the following groups: Q-5 fatty acids, tocotrienols, fucoxanthin. Preferably, the composition is a dietary supplement. The composition may contain ingredients from all three groups, or only from two of the three groups.
[0008] For example, a composition according to the invention may comprise one or more Q-5 fatty acids (n) and one or more tocotrienols (e), but not fucoxanthin.
[0009] It is also conceivable to have a composition according to the invention which comprises one or more tocotrienols (e) and fucoxanthin without including one or more Q-5 fatty acids (n).
[0010] It is also conceivable to have a composition according to the invention which comprises one or more Q-5 fatty acids (n) and fucoxanthin without including one or more tocotrienols (e).
[0011] A composition according to the invention comprising one or more tocotrienols (e), fucoxanthin and one or more Q-5 fatty acids (n) is particularly preferred. PWHELOOIWO / 16.04.2026 3 2026048716
[0012] Q-5 fatty acids are unsaturated fatty acids that contain a double bond at the fifth position, starting from the Q carbon. Such unsaturated fatty acids are known for their health-promoting effects. Q-5 fatty acids with three conjugated double bonds are called conjugated linolenic acids. A prime example of these Q-5 conjugated linolenic acids is punicic acid (CAS No. 544-72-9), a fatty acid with 18 carbon atoms and three conjugated double bonds.
[0013] Tocotrienols are vitamers and belong to the vitamin E compounds. Known examples are α-tocotrienol (CAS No. 58864-81-6), β-tocotrienol (CAS No. 490-23-3), and Y. _ T° cα-tocotrienol (CAS No. 14101-61-2) and 5-tocotrienol (CAS No. 25612-59-3), wherein in the composition according to the invention the tocotrienols are present in the naturally occurring (R) configuration. In contrast to tocopherols, the side chains of the tocotrienols are partially unsaturated. Tocotrienols are typically available as commercial products in the form of a mixture of isomers, whereby the concentration of certain desired isomers can be specifically increased. Examples include: 67 mg / g α-tocotrienol, <1 mg / g β-tocotrienol, 82 mg / g Y-tocotrienol. _ T° c °trienol, 5 mg / g 5-tocotrienol.
[0014] Tocotrienols are known, among other things, for their antioxidant properties.
[0015] Fucoxanthin (CAS No. 3351-86-8) is a molecule from the carotenoid group, often found in brown algae or microalgae. Fucoxanthin is associated with fat reduction.
[0016] Initial results showed a surprisingly improved anti-inflammatory effect for all the proposed compositions. Initial measurements show a reduced hs-PWHELOOIWO / 16.04.2026 4 2026048716
[0017] CRP, TNF-alpha and interleukin-6 concentration in serum, which is a strong indicator of improved anti-inflammatory action.
[0018] The ingredients from these three groups are known for a variety of effects. Q-5 fatty acids, especially puniic acid, are known for their positive effects on skin health and insulin sensitivity. Tocotrienol is known, among other things, for its anti-cancer properties and positive influence on cholesterol levels. Fucoxanthin, in turn, is associated with increased fat burning and cardiovascular effects. All three ingredients are attributed with antioxidant properties.
[0019] Initial experiments surprisingly revealed that the effect of a composition comprising ingredients from at least two of the three groups according to the invention exceeds the sum of the effects of the individual ingredients. While not bound by this explanation, it is assumed, based on previous experiments, that these synergistic interactions of the three ingredients are at least partially mediated via the NF-κB / TNF-α signaling pathway. Activation of NE-κB is considered critical for the development of inflammation and occurs, among other things, via pro-inflammatory cytokines such as TNF-α. Conversely, the activation of NE-κB in turn causes the expression of TNF-α, which can create a positive feedback loop that amplifies and maintains the inflammatory response in various ways. Consequently, interrupting this loop leads to the inhibition of inflammation.Fucoxanthin, Q-5 fatty acids, and tocotrienols achieve such an anti-inflammatory effect through interaction at various points in the NF-κB / TNF-α signaling pathway. It was shown by Hontecillas et al. (DOI: 10.1080 / 07315724.2009.10719770) that PWHELOOIWO / 16.04.2026 5 2026048716.
[0020] Punicic acid, in particular, which is a Q5 fatty acid, appears to exert an inhibitory effect on both NF-κB activation and TNF-α expression. Fucoxanthin also inhibits the NF-κB / TNF-α signaling pathway. Based on a study by Kim et al. (DOI: 10.1016 / j.ejphar.2010.09.032), it is assumed that fucoxanthin, among other things, inhibits the degradation of the IBa protein (which is an NF-κB inhibitor), leading to reduced NF-κB activity. Tocotrienol, especially 5-tocotrienol, has an effect according to Yang et al. (DOI: 10.1016 / j.jnut-bio.2018.10.013) also inhibits the NF-κB / TNF-α signaling pathway. Presumably, 5-tocotrienol increases the expression of NF-κB inhibitors (e.g., A20 and CYLD). Furthermore, 5-tocotrienol could also inhibit NF-κB activation.Based on this prior art information, it was not expected that a combination of at least two ingredients selected from the three groups would lead to a stronger effect than the sum of the effects of the individual ingredients. However, initial experiments surprisingly show a synergistic effect. This observed synergistic effect may also arise along other signaling pathways. In this context, the formation of specialized pro-resolving lipid mediators (SPMs), PPAR receptors (α and γ), the expression of the SIRT1 gene, and the MAPK cascade should be mentioned in particular. The multitude and mutual influence of the factors within these signaling pathways currently complicate a precise description of the effects underlying the observed synergistic effects.
[0021] In a preferred embodiment of the composition according to the invention, this contains punicic acid.
[0022] In a further preferred embodiment of all the foregoing aspects of the composition according to the invention, this contains 5-tocotrienol. PWHELOOIWO / 16.04.2026 6 2026048716
[0023] The composition can contain all tocotrienols in addition to 5-tocotrienol. The tocotrienols can be present individually or in any combination. The ratios of the tocotrienols to one another can vary and be adapted to the application of the composition according to the invention. For example, if the composition is used as a dietary supplement, a composition without 5-tocotrienol is also conceivable. In this case, the composition would contain α-tocotrienol and / or β-tocotrienol and / or Y. _ Tocotrienol. The composition preferably includes Y_ Tocotrienol and especially 5-tocotrienol.
[0024] 5-Tocotrienol has proven to be particularly effective for inhibiting inflammation in compositions according to the invention.
[0025] In another preferred embodiment of all the foregoing aspects of the composition according to the invention, this composition contains pomegranate seed oil.
[0026] A major component of pomegranate seed oil (INCI name: Punica Granatum Seed Oil; CAS No. 84961-57-9; EINECS No. 284-646-0) is punicic acid. Pomegranate seed oil also contains a high proportion of unsaturated fatty acids, with punicic acid typically making up over 70% by weight of these fatty acids. In addition to Q5 fatty acids, pomegranate seed oil contains other health-promoting ingredients, such as other fatty acids and polyphenols. Furthermore, pomegranate seed oil can also contain tocopherols and tocotrienols. However, these tocopherols are present in pomegranate seed oil in low concentrations, usually less than 1% by weight. Of the total weight of tocopherols in pomegranate seed oil, tocotrienols make up between 1.7% and 4.8%. 2026 7 2026048716
[0027] wt. %, with 5-tocotrienol making up less than 1 wt. % of the total tocol content in pomegranate seed oil .
[0028] In a further preferred embodiment of all the preceding aspects of the composition according to the invention, this composition contains fucoxanthin and tocotrienol, preferably 5-tocotrienol, wherein the weight ratio [fucoxanthin] : [tocotrienol] or [fucoxanthin] : [5-tocotrienol] is in the range of 1 : 800 - 3 : 5, preferably 1 : 400 - 1 : 2, particularly preferably 1 : 300 - 2 : 5.
[0029] It is assumed that the composition according to the invention achieves its advantages over the prior art, among other things, by the fact that the ingredients are present in improved proportions. Poor proportions of the ingredients can adversely affect efficacy. In addition, prior art compositions often have the problem that ingredients are combined that have an inhibitory effect, leading to further inefficiencies.
[0030] The composition in this formulation is particularly effective because the described weight ratios of [fucoxanthin] : [tocotrienol] particularly emphasize the synergistic interaction of the two ingredients.
[0031] In a further preferred embodiment of all the foregoing aspects of the composition according to the invention, this Q-5 fatty acid contains punic acid and tocotrienol, preferably 5-tocotrienol, wherein the weight ratio [Q-5 fatty acid] : [tocotrienol] or [punic acid] : [5-tocotrienol] is in the range of 1 : 40 - 40 : 1, preferably 1 : 10 - 14 : 1, particularly preferably 1 : 6 - 13 : 1. PWHELOOIWO / 16. 04 . 2026 8 2026048716
[0032] The tocotrienol content in the composition according to the invention is thus significantly higher than the natural tocotrienol content in pomegranate seed oil. It can be assumed that no synergistic effect occurs with a [Q-5 fatty acid] : [tocotrienol] ratio such as that found in pomegranate seed oil. The composition according to the invention in this embodiment is particularly effective because the described weight ratios of [Q-5 fatty acid] : [tocotrienol] especially emphasize the synergistic interaction of the two ingredients.
[0033] In a preferred embodiment of all the foregoing aspects, the composition according to the invention further comprises at least one Q-3 fatty acid, preferably selected from the group consisting of eicosapentaenoic acid (CAS No. 10417-94-4) and docosahexaenoic acid (CAS No. 6217-54-5). These Q-3 fatty acids are found, among other things, in fish oils.
[0034] The composition according to the invention can contain eicosapentaenoic acid but not docosahexaenoic acid; or it can contain docosahexaenoic acid but not eicosapentaenoic acid; or it can comprise docosahexaenoic acid and eicosapentaenoic acid.
[0035] Surprisingly, the Q-3 fatty acids, especially docosahexaenoic acid and eicosapentaenoic acid, also act synergistically with the other constituents, selected from the groups of Q-5 fatty acids, tocotrienols, and fucoxanthin. While not bound by this explanation, it is assumed based on previous experiments that the Q-3 fatty acids also achieve their synergistic effect at least partially via the NF-κB / TNF-α signaling pathway. (See Daak et al. [DOI: 10.1016 / j.bcmd.2015]).
[0036] As shown in 03.014), Q-3 fatty acids appear to inhibit the expression of NF-KB. PWHELO OIWO / 16.04.2026 9 202604 8716
[0037] In a preferred embodiment, the composition according to the invention contains tocotrienol, preferably 5-tocotrienol, and eicosapentaenoic acid, wherein the weight ratio [eicosapentaenoic acid] : [tocotrienol] or [eicosapentaenoic acid] : [5-tocotrienol] is in the range of 1 : 8 - 200 : 1, preferably 3 : 8 - 140 : 1, particularly preferably 2 : 3 - 80 : 1.
[0038] In a further preferred embodiment of all the foregoing aspects, the composition according to the invention comprises tocotrienol, preferably 5-tocotrienol, and docosahexaenoic acid, wherein the weight ratio [docosahexaenoic acid] : [tocotrienol] or [docosahexaenoic acid] : [5-tocotrienol] is in the range of 1 : 8 - 100 : 1, preferably 5 : 8 - 60 : 1, particularly preferably 5 : 3 - 40 : 1.
[0039] Particularly pronounced synergistic effects were observed at these weight ratios of tocotrienol, preferably 5-tocotrienol, to the Q-3 fatty acids eicosapenoic acid and eicosapenoic acid, respectively.
[0040] Docosahexaenoic acid observed.
[0041] In another preferred embodiment, the composition contains per dosage unit for oral administration: tocotrienols, preferably 5-tocotrienol: 50–800 mg, preferably 50–400 mg, particularly preferably 50–300 mg; fucoxanthin: 1–30 mg, preferably 1–25 mg, particularly preferably 1–20 mg; punicic acid: 20–2000 mg, preferably 40–700 mg, particularly preferably 50–650 mg.
[0042] In another preferred formulation, the composition contains per oral dosage unit: tocotrienols, preferably 5-tocotrienol: 50–800 mg, preferably 50–400 mg, particularly preferably 50–300 mg; fucoxanthin: 1–30 mg, preferably 1–25 mg, particularly preferably 1–20 mg; punicic acid: PWHELOOIWO / 16.04.2026 10 2026048716
[0043] 20 - 2000 mg, preferably 40 - 700 mg, particularly preferably 50 - 650 mg; Eicosapentaenoic acid: 100 - 10000 mg, preferably 150 - 7000 mg, particularly preferably 200 - 4000 mg; Docosahexaenoic acid: 100 - 5000 mg, preferably 250 - 3000 mg, particularly preferably 500 - 2000 mg.
[0044] The compositions according to the invention can be packaged in various dosage forms. Examples include hard capsules, soft capsules, tablets, powders, liquids, effervescent tablets, lozenges, and chewable tablets.
[0045] The compositions according to the invention can be taken daily in quantities of 3 to 5 capsules for the prevention of inflammation. In cases of acute inflammation, the quantity can be increased to up to 9 to 15 capsules daily. The capsules are advantageously taken during or after meals.
[0046] Depending on the type of treatment performed by the composition according to the invention, the dosage forms, ingredients, and proportions can be selected, and further ingredients can be added to the composition. Within the scope of the invention, the treatment of a condition also includes the prevention of that condition.
[0047] Another aspect of the invention relates to the composition according to the invention, which is used as a medicine.
[0048] Another aspect of the invention relates to the composition according to the invention, wherein this is used to manufacture an agent for the treatment of inflammation, in particular silent inflammation and / or chronic inflammation. PWHELOOIWO / 16.04.2026 11 2026048716
[0049] Silent inflammation is present when the concentration of C-reactive protein (CRP) in the blood is 3 - 10 mg / L.
[0050] Another aspect of the invention relates to the composition according to the invention, which is used to treat inflammations, in particular silent inflammations and / or chronic inflammations.
[0051] In addition to its anti-inflammatory properties, the composition according to the invention can also have other positive effects on health. These effects include, among others, metabolic regulation, oxidative protection mechanisms, the detrimental effects on senescent cells, and neuroprotection.
[0052] The invention will now be described using specific embodiments. These embodiments are not intended to restrict the core of the invention, but rather to facilitate its understanding. The first eight examples are summarized in Table 1.

[0061] Initial trials showed a slight decrease in serum TNF-alpha.
[0062] Example 2 shows a composition consisting solely of tocotrienols. Initial trials also observed a slight decrease in serum TNF-alpha.
[0063] Example 3 shows a composition that consists solely of fucoxanthin. Initial trials failed to demonstrate a significant reduction in inflammation markers.
[0064] Example 4 shows a composition comprising Q-5 fatty acids (in some trials only punicic acid), tocotrienols, and fucoxanthin. Surprisingly, initial trials demonstrated that the combination of these three ingredients led to a reduction in serum hs-CRP, TNF-alpha, and interleukin-6 that exceeded the sum of the effects of the individual ingredients.
[0065] Example 5 shows a composition comprising Q-5 fatty acids (in some trials only punicic acid) and tocotrienols, without fucoxanthin. Initial results for this example show a particularly strong reduction in serum TNF-alpha. Here, too, the extent of the reduction in serum TNF-alpha exceeds the sum of the effects of the individual ingredients.
[0066] Example 6 shows a composition comprising Q-5 fatty acids (in some trials only punicic acid) and fucoxanthin, without tocotrienols. Initial results for this example show a reduction in serum hs-CRP, TNF-alpha, and interleukin-6. This effect, too, would not have been expected based on the individual effects of the ingredients. PWHELOOIWO / 16.04.2026 13 2026048716
[0067] Example 7 shows a composition comprising tocotrienols and fucoxanthin, without Q-5 fatty acids. Initial results for this example show a reduction in serum hs-CRP, TNF-alpha, and interleukin-6 that exceeds the reduction achieved by the individual ingredients.
[0068] In example 8, which contains Q-3 fatty acids compared to example 4, the effect already observed in example 4 is improved once again.
[0069] In Example 9, a composition according to the invention containing olive oil, pomegranate seed oil, γ-tocotrienol, fucoxanthin, and an algal oil containing docosahexaenoic acid and eicosapentaenoic acid was provided and compared with a potent anti-inflammatory drug (prednisolone). The inhibition of TNF-α release in human whole blood was measured. Prednisolone served as a pharmacological positive control to evaluate the anti-inflammatory efficacy of the composition according to the invention compared to a known, potent anti-inflammatory drug. The inhibition of TNF-α release was determined by stimulating human whole blood with lipopolysaccharide (LPS) to achieve maximum TNF-α release. Subsequently, the compound or the prednisolone was incubated and the remaining TNF-α concentration was quantified after 24 hours using enzyme-linked immunosorbent assay (ELISA).The percentage TNF-α inhibition was calculated using the formula: Inhibition (%) = (1 - [TNF-α + substance] / [TNF-α + LPS alone]) x 100. The results are shown in Table 2. "Baseline" corresponds to the TNF-α concentration in LPS-stimulated whole blood without the addition of the compound or prednisolone. [0078] Table 3: Example 10.
[0079] The cytoprotective effect of the formulations was investigated in immortal primary creatine cells damaged by olive oil. The investigation was performed on cells with and without an inflammatory response. The cytoprotective effect was measured by cell viability. A viability assay (alamarBlue™ assay) was used to determine viability. In this assay, the non-fluorescent indicator dye resazurin was added to the culture medium. Metabolically active cells reduce resorufin to fluorescent resorufin. The resulting fluorescence, measured at 590 nm, is proportional to the number of viable cells. Pam2CSK4 (cat. No. tlrl-pm2s-l, invivo gene), a commercial Toll-like receptor 2 / 6 agonist, was used to induce an inflammatory response in the cells.Olive oil was used as the vehicle for the ingredients, which has a cytotoxic effect (see sample 12 in Figure 1) and thus has a negative effect on vitality.
[0080] The formulations (60 pL) were applied topically to the apical, air-exposed surface of the 3D NF-κB keratinocyte culture. Pam2CSK4 was simultaneously added systemically via the medium. The models were incubated in an incubator at 37 °C and 5% saturated COp in a humidified atmosphere. Cell viability was determined after 24 hours of experiment time (from the addition of Pam2CSK4) using the alamarBlue™ assay. PWHELOOIWO / 16.04.2026 16 2026048716
[0081] Figure 1 shows the mean values of n = 3 measurements and the standard deviation relative to the untreated vital control, to which no olive oil was added (100% vitality). The results are categorized as "with induced inflammation" (i.i.) and "without induced inflammation" (n.i.i.).
[0082] Figure 1 shows that olive oil has a negative effect on cell vitality. It is assumed that the oil damages cell membranes and hinders cell metabolism. The low vitality of formulation 12 is used as a baseline for assessing the protective effect of the formulations. For most formulations, the induction of inflammation leads to a further decrease in vitality, as the inflammation, in combination with the olive oil, represents a double burden. Formulation 1, which combines all ingredients, is the only formulation that maintains a stable vitality of over 80% despite the double burden caused by inflammation induction. For the other formulations with high vitality (4, 7, 9, and 11), vitality decreases upon the induction of inflammation. In contrast to formulations 2 to 11, which did not show fully maintained viability under inflammation induction, formulation 1 surprisingly showed consistently high viability even in cells with induced inflammation.
[0083] While formulation 4 (consisting of tocotrienol and olive oil) exhibits high viability, this formulation is also notable for its high standard deviation. The cytoprotective effect of tocotrienol appears to be subject to considerable fluctuations. It was surprisingly found that the cytoprotective effect of tocotrienol was reduced by its combination with PWHELOOIWO / 16.04.2026 17 2026048716
[0084] One or more of the other ingredients, in particular fucoxanthin, PSO, eicosapentaenoic acid, and docosahexaenoic acid, could be stabilized. This is evident from formulations 1, 7, 9, and 11 in Figure 1, whose standard deviations are significantly smaller.
