Description

Novel Hydroxyacylnornicotine Derivatives and Uses thereof

[0001] The present application relates to a novel hydroxyacylnornicotine derivative and the medicinal use of said derivative for the treatment of bone diseases.

[0002] Bone is maintained through a balance between bone formation by osteoblasts and bone resorption by osteoclasts. Maintaining this balance between osteoblasts and osteoclasts is an essential element for maintaining bone homeostasis. The disruption of this balance due to aging occurs when the bone-destroying capacity of osteoclasts exceeds the bone-forming capacity of osteoblasts, thereby breaking down homeostasis. This implies that inhibiting bone resorption by osteoclasts is crucial for the prevention and treatment of age-related bone diseases. Excessive osteoclast activity that outweighs osteoblast activity causes various bone diseases and leads to a decrease in bone mass and structural deterioration of the skeleton.

[0003] Osteoporosis is a skeletal disease characterized by a weakened bone strength due to a decrease in bone mass and changes in microstructure, resulting in a condition prone to fractures. As a skeletal disease caused by various factors, osteoporosis leads to a reduction in bone mass and causes microstructural deformation. However, when estrogen secretion decreases due to menopause, menopausal osteoporosis occurs due to an imbalance between osteoclasts and osteoblasts. Therefore, inhibiting the differentiation of osteoclasts can be an important alternative for treating osteoporosis. Osteoclasts are generated from macrophages derived from hematopoietic stem cells, undergoing various differentiation stages mediated by M-CSF (macrophage colony-stimulating factor) and RANKL (receptor activator of nuclear factor-κB ligand). When RANKL binds to RANK (receptor activator of nuclear factor-κB), an osteoclast receptor present on the surface of osteoclast precursor cells, it induces the activation of c-Fos. Activated c-Fos induces the expression of another transcription factor, NFATc1 (nuclear factor of activated T cell c1). Subsequently, NFATc1 regulates bone resorption, activation, survival, and differentiation by activating the differentiation and function of osteoclasts through the promotion of the expression of transcription factors / osteoclast markers essential for osteoclast differentiation, such as TRAP (tartrate-resistant acid phosphatase).

[0004] Bisphosphonates are currently the most widely used treatments for osteoporosis. However, as numerous side effects have been observed in patients treated with bisphosphonates, concerns regarding this treatment have been raised; therefore, there is a need to develop new treatments that can demonstrate effective therapeutic efficacy with fewer side effects.

[0005] As a result of diligent efforts to develop a therapeutic agent derived from natural products, the inventors isolated a novel hydroxyacylnornicotine derivative from the plant *Nicotiana debenei*. Along with this, they confirmed that the isolated hydroxyacylnornicotine derivative not only exhibits low cytotoxicity but also effectively inhibits differentiation into osteoclasts, thereby being applicable as an active ingredient for the treatment of bone diseases, and based on this, they completed the present invention.

[0006] One aspect is to provide a hydroxyacylnornicotine derivative having an inhibitory effect on differentiation into osteoclasts.

[0007] Another aspect is to provide a pharmaceutical composition for the prevention or treatment of bone diseases comprising the above-mentioned hydroxyacylnornicotine derivative as an active ingredient.

[0076]

[0077] Another aspect provides a pharmaceutical composition for the prevention or treatment of bone disease comprising, as an active ingredient, a compound represented by the above chemical formula I, a solvate, a stereoisomer, or a pharmaceutically acceptable salt thereof.

[0078] In the above pharmaceutical composition, any terms or elements mentioned that are identical to those already mentioned are as described above.

[0079] In this specification, the term "prevention" refers to any act of suppressing or delaying the onset of a disease through the administration of the above composition.

[0080] In this specification, the term “treatment” means any form of treatment that provides effects to an individual suffering from a disease or at risk of developing a disease, including improvement of the individual’s condition (e.g., one or more symptoms), delay of disease progression, delay of symptom onset, or slowing of symptom progression. Accordingly, “treatment” and “prevention” are not intended to mean the cure or complete elimination of symptoms.

[0081] The "bone disease" targeted for prevention or treatment by the above pharmaceutical composition may be a collective term for diseases caused by an imbalance in the bone metabolic process between osteoblasts and osteoclasts. The bone disease may be, for example, a bone disease associated with excessive bone resorption by osteoclasts or excessive differentiation into osteoclasts, or a bone disease in which therapeutic efficacy can be exerted through the inhibition of osteoclast activity or differentiation into osteoclasts. Specifically, the bone disease may be selected from the group consisting of osteoporosis, osteoporotic fracture, osteopenia, osteolytic metastasis, senile kyphosis, Paget's disease, bone defect, nonunion fracture, osteomalacia, osteomalacia fracture, bone formation disorder, osteoarthritis, fibrous osteitis, aplastic bone disease, osteogenesis imperfecta, bone atrophy, periodontal disease, rheumatoid arthritis, metabolic bone disease, and bone damage caused by bone metastasis of cancer cells, but is not limited thereto.

[0082] Among the aforementioned bone diseases, "osteoporosis" refers to a pathological condition in which bone mass decreases and bone strength weakens due to qualitative changes, making fractures highly likely. In the case of osteoporosis, the disease develops and progresses as the balance between the activities of osteoblasts and osteoclasts is disrupted, leading to excessive bone destruction by osteoclasts. A major molecule in osteoporosis is RANKL (receptor-activator of nuclear factor kappa-B ligand), and it is known that RANKL binds to its receptor to activate various transcription factors, thereby promoting the differentiation of osteoclasts. Furthermore, it has been reported that when macrophages are treated with RANKL, they differentiate into TRAP (Tartrate-resistant acid phosphatase)-positive cells along with binding to RANK (Receptor-activator of nuclear factor kappa-B); when these TRAP-positive cells are stimulated with inflammatory factors such as RANKL and TNF-α, the cells fuse together and differentiate into multinucleated TRAP-positive cells. Under this technical background, the TRAP can be applied to evaluate the biological / pharmacological activity of an active compound according to one embodiment as one of the differentiation markers into osteoclasts.

[0083] According to one embodiment, the hydroxyacylnornicotine derivative isolated and purified in one embodiment not only did not exhibit cytotoxicity but was also able to inhibit differentiation into osteoclasts. Therefore, the hydroxyacylnornicotine derivative can be applied to regulate the imbalance in the bone metabolic process between osteoblasts and osteoclasts. Through this, the hydroxyacylnornicotine derivative can be utilized for medicinal purposes for the prevention or treatment of bone diseases, and specifically, can be included as an active ingredient in a pharmaceutical composition for the prevention or treatment of bone diseases.

[0084] In this specification, the term “included as an active ingredient” means that a compound, solvate, stereoisomer, or pharmaceutically acceptable salt thereof represented by Formula I is added to an extent that can produce the effects mentioned in this specification, and includes formulation in various forms by adding various ingredients as auxiliary ingredients for drug delivery and stabilization.

The pharmaceutical composition may be formulated according to conventional methods and may be prepared in various oral administration forms such as tablets, pills, powders, capsules, syrups, emulsions, microemulsions, or parenteral administration forms such as intramuscular, intravenous, or subcutaneous administration.

[0086] When the above pharmaceutical composition is prepared in the form of an oral formulation, examples of additives or carriers used include cellulose, calcium silicate, corn starch, lactose, sucrose, dextrose, calcium phosphate, stearic acid, magnesium stearate, calcium stearate, gelatin, talc, surfactants, suspending agents, emulsifiers, diluents, etc. When the pharmaceutical composition of the present invention is prepared in the form of an injectable formulation, examples of additives or carriers include water, saline solution, glucose aqueous solution, similar sugar aqueous solution, alcohol, glycol, ether (e.g., polyethylene glycol 400), oil, fatty acid, fatty acid ester, glyceride, surfactant, suspending agent, emulsifier, etc.

[0087] The dosage of the above pharmaceutical composition is an amount effective for the treatment or prevention of an individual or patient, and may be administered orally or parenterally as intended. When administered orally, it may be administered in an amount of 0.01 to 1000 mg per kg of body weight per day based on the active ingredient, more specifically 0.1 to 300 mg; when administered parenterally, it may be administered in an amount of 0.01 to 100 mg per kg of body weight per day based on the active ingredient, more specifically 0.1 to 50 mg. The dosage for a specific individual or patient should be determined in light of various relevant factors such as the patient's body weight, age, gender, health status, diet, time of administration, method of administration, and severity of the disease, and it should be understood that it may be appropriately adjusted by a professional. The above dosage is not intended to limit the scope of the present invention in any way. A physician or veterinarian with ordinary skills in the relevant technical field can easily determine and prescribe the required effective amount of the pharmaceutical composition. For example, a physician or veterinarian may start the dosage of the compound of the present invention used in a pharmaceutical composition at a level lower than that required to achieve the desired therapeutic effect, and gradually increase the dosage until the desired effect is achieved.

[0088] In one embodiment, the pharmaceutical composition includes within its scope a pharmaceutical composition comprising, as an active ingredient, at least one of the compounds according to one embodiment alone or in combination with a pharmaceutical carrier in a therapeutically effective amount. Optionally, the compound according to one embodiment may be administered alone, in combination with a compound according to another embodiment, or simultaneously, separately, or sequentially with one or more other therapeutic agents, e.g., a bone disease treatment or other pharmaceutically active substance.

[0089]

[0109]

[0110] Preparation Example 1. Isolation and purification of active ingredients derived from Nicotiana debenei extract

[0111] 1-1. Preparation of Ethyl Acetate Fraction Derived from Nicotiana Devnay Extract

[0112] The ground above-ground parts of Nicotiana debenay were extracted with a methanol solvent, and fractions were obtained for each solvent. Specifically, 900 g of the ground above-ground parts of Nicotiana debenay sample (W21) was added to 9 L of methanol, and a total of 18 L of methanol extract obtained by repeated extraction was concentrated under reduced pressure to obtain a concentrate. An equal amount of hexane was added to the concentrate and partitioned to separate it into a hexane layer and a water layer. Subsequently, ethyl acetate was added to the water layer and partitioned to separate it into an ethyl acetate layer and a water layer. Subsequently, the ethyl acetate layer exhibiting activity was concentrated and silica gel column chromatography was performed (elumination solvent, chloroform-methanol = 50:1, 20:1, 10:1, 5:1, 2:1, 1:1, v / v, eluted in 500 mL each), and as shown in Figure 1, the results were fractionated into 8 groups (EA-1 to EA-8) through thin layer chromatography (TLC) analysis.
