Abstract
A method for evaluating a validity period of qualitative nucleic acid reference samples for Potebniamyces pyri includes: acquiring Ct value data of a reference sample obtained by performing real-time fluorescent polymerase chain reaction (PCR) tests at different time points; constructing a fitted curve based on the Ct value data and corresponding time points; calculating a standard error of a slope of the fitted curve, and obtaining, in combination with the slope of the fitted curve, a t-value of the fitted curve; determining whether the t-value of the fitted curve is less than a critical value for a t-test; calculating a confidence probability of a preset validity period of the reference sample if the t-value of the fitted curve is less than the critical value for the t-test; and taking the preset validity period as a validity period of the reference sample if the confidence probability is greater than a threshold. According to the method of the present application, whether the stability of the reference sample meets requirements within the preset validity period is determined by analyzing the Ct value data and the preset validity period, and if the stability does not meet the requirements, data of the validity period is output. In this way, experimental personnel can effectively reduce a number of experimental tests on the reference sample, thereby decreasing the consumption of experimental resources and labor, and ensuring the stability of the reference sample.
