Abstract

The present invention discloses a dsRNA of an Oncorhynchus mykiss fertility-regulating gene and its application, which belongs to the field of biotechnology. The present invention provides an Oncorhynchus mykiss fertility-regulating gene ccne1, an interference gene fragment and its dsRNA. Firstly, the gene sequence of ccne1 is obtained by cloning with the total RNA of gonadal tissues of juvenile Oncorhynchus mykiss as the template, and its nucleotide sequence is SEQ ID NO.1 and amino acid sequence is SEQ ID NO.2. Primers for the interference gene fragment are designed with the ccne1 gene sequence as the template to obtain the interference gene fragment with the nucleotide sequence shown in SEQ ID NO.3, and the dsRNA of Oncorhynchus mykiss ccne1 gene is synthesized through in vitro transcription. The dsRNA of Oncorhynchus mykiss ccne1 gene of the present invention may efficiently silence the ccne1 gene and effectively reduce the expression of other meiosis-related genes in Oncorhynchus mykiss, providing research ideas for the analysis of Oncorhynchus mykiss fertility regulation pathways.
