Abstract
The invention discloses a preparation method of a mesenchymal stem cell exosome repair gel, comprising: mixing purified mesenchymal stem cell exosomes with a biocompatible gel matrix solution at low temperature, adding a composite lyophilization protectant composed of trehalose, mannitol, and polyethylene glycol, and then performing programmed cooling and freeze-drying to obtain the product. This method, through the synergistic optimization of the protectant formulation and the freeze-drying process, solves the problems in existing exosome wet-state gels such as poor stability, easy activity attenuation, and inconvenient storage and transportation. The obtained product is a porous sponge-like solid, rapidly reconstitutable and capable of restoring the original performance of the gel, with exosome biological activity well preserved, long-term storage achievable without cold chain, and the process scalable for industrial production, thereby providing a reliable solution for the clinical translation and industrialization of exosome repair gels.
