Abstract
The present disclosure belongs to the technical field of animal experimental model construction and relates to a method for constructing a porcine endometrial injury model. The construction method is non-invasive, and may simulate a pathological state of clinical intrauterine adhesions without destroying a full-thickness structure of porcine uterus, specific steps are as follows: Administering 1.5 mg / kg of Zoletil for sedation via intramuscular injection to experimental pigs. Subsequently, inducing anesthesia and providing analgesia, followed by anesthesia induction is successful and vital signs are stable, administering phloroglucinol via intramuscular injection, examining a cervix to ensure the cervix relaxed; inserting a NO. 6 curette gently through the cervix into a uterine body and scraping an endometrium mechanically until a surface becomes rough; then injecting 95 % ethanol and allowing it to remain for a specified duration, finally, removing any residual ethanol. The present disclosure does not require laparotomy, features a simple procedural pathway, and preserves the full-layer structure of the myometrium and serosal layer. The modeling effect closely resembles clinical intrauterine adhesions, with a success rate as high as 95%, excellent reproducibility, and stable modeling outcomes. This provides a standardized, scalable large animal model for research on the mechanisms of intrauterine adhesions and evaluation of clinical interventions.
