Abstract

The present invention relates to a method for formulating semi-synthetic culture media using local tubers in mushroom culture. The invention provides low-cost semi-synthetic culture media for Schizophyllum commune mycelial growth using local tubers from potato (Solanum tuberosum), sweet potato (Ipomea batatas), tapioca (Manihot esculenta), and yam (Dioscorea alata). Each medium is prepared by boiling 200g sliced tuber in 1L distilled water for 20 minutes, filtering the decoction, supplementing with 20g dextrose and 15g agar, adjusting pH to 6, and autoclaving at 121°C / 15 psi for 20 minutes. Inoculated plates incubated at 24±1°C for 14 days show Sweet Potato Dextrose Agar (SPDA) yields maximum radial growth, followed by TDA and PDA, with YDA least. This sustainable method replaces expensive synthetic media for efficient mushroom spawn production.
