AI Technical Summary

Technical Problem

Current methods for generating induced pluripotent stem cells (iPS cells) using viral delivery of reprogramming factors are inefficient and can lead to genomic integration, resulting in unpredictable outcomes and potential cancer risks.

Method used

Using purified single-stranded mRNA molecules containing modified nucleosides such as pseudouridine and 5-methylcytidine, free of RNA contaminants, to reprogram eukaryotic cells, including human cells, without genomic integration.

Benefits of technology

Achieves highly efficient and rapid generation of iPS cells with maintained genomic integrity, reducing the risk of cancer and immune responses.

Abstract

To provide compositions and methods for reprogramming somatic cells using purified RNA preparations comprising single-strand mRNA encoding an iPS cell inducing factor.SOLUTION: Provided is a method for inducing mammalian cells existing in vitro to produce a recombinant protein, the method including contacting the mammalian cells with a preparation of mRNA synthesized in vitro, containing an open reading frame encoding a recombinant protein, the mRNA being obtained by in vitro transcription and containing Ψ(5-β-D-ribofuranosyluracil), or Ψ(5-β-D-ribofuranosyluracil) containing a modified nucleobase, the mRNA preparation being purified using a purification step of removing RNA contaminant molecules that are immunogenic and toxic to the cells by inducing an innate immune response.SELECTED DRAWING: Figure 1
