AI Technical Summary

Technical Problem

There is a need for identifying and utilizing effective antigens and T cell epitopes for diagnosing, treating, and developing vaccines for Alphavirus-related diseases, particularly Chikungunya virus, and for optimizing megapools to detect and characterize specific responses to the virus during infection and post-vaccination.

Method used

The development of compositions comprising specific peptides or proteins with defined amino acid sequences, fusion proteins, and polynucleotides encoding these sequences, which are used to stimulate T cell responses, and the creation of megapools and kits for detecting and characterizing immune responses to Chikungunya virus, including the use of adjuvants and immune modulators to enhance immunogenicity.

Benefits of technology

These compositions and methods enable effective detection and characterization of Chikungunya virus-specific immune responses, stimulation of T cell responses, and potential treatment or prevention of infections by eliciting protective immune reactions.

Abstract

The present invention includes compositions, including epitope megapools, and methods for detecting the presence of: an Alphavirus or an immune response relevant to an Alphavirus infection including T cells responsive to one or more Alphavirus peptides or proteins comprising, consisting of, or consisting essentially of: one or more amino acid sequences selected from any sequence set forth in Table 1 (SEQ ID NOS: 1 to 147), or a subsequence, portion, homologue, variant or derivative thereof; a fusion protein; a pool of 2 or more peptides; a polynucleotide that encodes one or more peptides or proteins. The invention further provides vaccines, diagnostics, therapies, and kits, comprising such proteins or peptides.
