AI Technical Summary

Technical Problem

The development and manufacturing of cell-based therapeutics face significant challenges due to the complexities of using living organisms as drugs, including the need for highly multiplexed cellular assays to characterize engineered cell populations.

Method used

The use of microfluidic systems with hydrogel chambers to perform large-scale multiplexed single cell assays, allowing for the characterization of cellular characteristics such as cytotoxicity, proliferative capacity, and vector copy number, and enabling the culture of individual cells into larger clonal populations.

Benefits of technology

This approach enables the efficient characterization and expansion of potent cells within a population, facilitating the development and quality assurance of cell-based therapeutics.

Abstract

The systems and methods described herein are directed to carrying out one or more highly multiplexed cellular assays. In some embodiments, one or more channels of a fluidic device are provided with photopolymerizable polymer precursors and cells randomly disposed on a surface, after which positions of cells are measured by a detector, hydrogel chambers are synthesized by photopolymerization to enclose individual cells, and channels are loaded with assay reagents. Assay signals indicative of desired cellular characteristics are generated for each of the enclosed cells.
