[0010] The present invention mainly relates to mixtures from extracts of stinging nettle and ginger root, and their uses in cosmetics and therapy, in particular cosmetic and / or therapeutic skincare.

[0011] Latest studies show that consumers’ perception of skin sensitivity including itching, tingling 5 sensation and skin redness has increased. Many examples exist of medicaments designed to relieve or sooth irritated skin. These ingredients, whilst on the whole successful, are sometimes considered by consumers as “over the top”. A need exists for novel cosmetic ingredients with soothing and skin redness reducing efficacy.

[0012] Thus, an object of the present invention is to provide cosmetically accepted ingredients that0 can be incorporated into topical cosmetic formulations and are capable to sooth skin and / or reduce skin redness when applied topically. In addition to their effectiveness they should be price competitive and, if possible, a “green” alternative, to current soothing agents.

[0013] The object is achieved by the present invention. According to a first aspect of the present invention, a mixture formed from extracts of stinging nettle and ginger root comprises5 zingiberene, 6-gingerol and caffeoylmalic acid, and a weight ratio of the stinging nettle extract to the ginger root extract ranges from 1 :10 to 10:1 , preferably from 1 :6 to 6:1 , more preferably 1 :4 to 4:1 . In case the weight ratio of the stinging nettle extract to the ginger root extract in a mixture of the invention is not known, the weight ratio is an approximation determined according to the following approach:

[0014] 1 . Determine the average content of 6-gingerol in the dry matter of a representative number of ginger extracts. 2. Determine the average content of caffeoylmalic acid in the dry matter of a representative number of stinging nettle extract. 3. Calculate a first ratio of the determined 6-gingerol and caffeoylmalic acid content. 4. Determine the 6-gingerol and caffeoylmalic acid content in the mixture in which the weight ratio is to be deteremined, and calculate a second ratio in the same way. 5. Determine the ratio of the first and second ratios to obtain an aproximation forthe weight ratio of the dry extracts of ginger and stinging nettle in the mixture.

[0016] The present invention is based on the inventors’ observation that an extract of ginger root as described herein in combination with an extract of stinging nettle is capable to inhibit cytokine production in the form of interleukin 6 (IL-6) and interleukin 8 (IL-8) in keratinocyte cells in a synergistic manner. Based on this observation the inventors hypothesize that the mixture of the invention exhibits a synergistic efficacy with respect to soothing skin and / or reducing skin redness.

[0017] Therefore, a preferred mixture contains the stinging nettle and ginger root extracts in an amount sufficient to inhibit IL-6 biosynthesis and / or IL-8 biosynthesis in keratinocytes by at least 25%, as determined using the assay described in the Example section.

[0018] The effects associated with the combined extracts can be exploited in cosmetic skincare and / or therapeutic skincare. Advantageously, the extracts of ginger root and stingle nettle are considered safe and well tolerated by the skin. They are thus not only effective, but due to their safety can be rapidly included in cosmetic and / or therapeutic skincare products that may be used even daily.

The dry matter was determined by lyophilization to be 4.9%.

[0051] 1 . Inhibition of interleukin biosynthesis

[0052] IL-8 / IL-6 ELISA: According to the test procedure, the IL-8 and IL-6 produced and secreted by cells are determined in an Enzyme linked immunosorbance assay (ELISA) from cell supernatants utilizing antibodies detecting the I L-8 / IL-6. The assay is performed on HaCaT cells by addition of test substances and their incubation for 6h (37°C). Afterwards, inflammation is triggered with IL1a for 24h or 2h, respectively, and IL-8 / IL-6 is measured. Therefore, the quantitative sandwich enzyme immunoassay technique is employed. A monoclonal antibody specific for human IL-8 / IL-6 is pre-coated onto a microplate. Standards and samples / supernatants are pipetted into the wells and any IL-8 present is bound by the immobilized antibody. After washing away any unbound substances, an enzyme-linked polyclonal antibody specific for human IL-8 / IL-6 is added to the wells. Following washing away any unbound antibody-enzyme reagent, a substrate solution is added. A color develops in proportion to the amount of IL-8 bound in the initial step. Results are calculated as inhibition [%]. Therefore, the IL-8 / IL-6 concentrations for unknown samples are interpolated from a standard curve. Finally, the concentration of the nonstimulated control is subtracted of each determined IL-8 / IL-6 concentration. Afterwards the inhibition is determined in relation to the stimulated control.

[0054] 2. Stabilization of 6-gingerol

[0055] Stability of raw materials and finished formulations upon storage and normal use is important. Normal use of finished formulations includes opening and closing the packaging over typically several months which leads to oxygen exposure or storage at different conditions such as e.g. elevated temperatures. Active ingredients delivering benefits need to be stable or have to be stabilized to deliver their activity also over longer times of use.

[0056] To evaluate the oxidative stability of ginger root extract, 6-gingerol was used as analyte. 6- gingerol is a known anti-inflammatory agent. Tests were performed in a simplified system consisting of water and dipropylene glycol (DPG), an organic solvent used in cosmetic and fragrance compositions.

[0057] The stability test was performed with an Oxipres device of Mikrolab. The device allows the determination of oxidative resistance and is a modification of the bomb method according to ASTM D941 , which is based on oxidation with oxygen. The test runs under elevated pressure and temperature, through which the degradation process is accelerated due to harder conditions such as temperature and pressure, allowing thus to simulate storage conditions for ingredients and investigate their behavior.

Claims

Claims:

1. A mixture formed from extracts of stinging nettle and ginger root, the mixture comprising zingiberene, 6-gingerol and caffeoylmalic acid, and a weight ratio of the stinging nettle extract to the ginger root extract ranging from 1 :10 to 10:1 , preferably 1 :6 to 6:1 , more preferably 1 :4 to 4:1 , on a dry weight basis.

2. The mixture of claim 1 being in the form of a powder, preferably a spray-dried powder.

3. The mixture of claim 1 or 2, further comprising one or more auxiliary agents selected from the group of maltodextrin, Arabic gum, and combinations thereof.

4. The mixture of claim 3, wherein the amount of maltodextrin ranges from 10 to 95% by weight, preferably 30 to 90%, more preferably 40 to 85%, on a dry weight basis relative to the total weight of the mixture.

5. The mixture of claim 3 or 4, wherein the amount of Arabic gum ranges from 1 to 25% by weight, preferably 3 to 20%, more preferably 5 to 15%, on a dry weight basis relative to the total weight of the mixture.

6. The mixture of any of the preceding claims, wherein the amount of the stinging nettle extract in the mixture ranges from 0.5 to 20% by weight, preferably 1 .0 to 10%, more preferably 2.0 to 5.0%, on a dry weight basis relative to the total weight of the mixture.

The mixture of any of the preceding claims, wherein the stinging nettle extract is characterized by a composition comprising, on a dry weight basis relative to the total weight of the ginger root extract, one or more of the following: neochlorogenic acid in an amount ranging from 0.2 to 4.0% by weight, preferably 0.3 to 2.0% by weight, more preferably 0.4 to 1 .5% by weight; cryptochlorogenic and chlorogenic acids in an amount ranging from 0.5 to 4.0% by weight, preferably 0.8 to 3.0% by weight, more preferably 1 .0 to 2.0% by weight; caffeoyl malic acid in an amount ranging from 1 .0 to 8.0% by weight, preferably 2.0 to 6.0% by weight, more preferably 3.0 to 5.0% by weight.

8. The mixture of any of the preceding claims, wherein the amount of the ginger root extract in the mixture ranges from 2 to 40% by weight, preferably 3 to 25%, more preferably 5 to 15%, on a dry weight basis relative to the total weight of the mixture.

The mixture of any of the preceding claims, wherein the ginger root extract is characterized by a composition comprising, on a dry weight basis relative to the total weight of the ginger root extract: zingiberene in an amount ranging from 10 to 50% by weight, preferably 15 to 45% by weight, more preferably 20 to 40% by weight, preferably with the proviso that the total amount of essential oils ranges from 50 to 99% by weight, preferably 70 to 97% by weight, more preferably 85 to 95% by weight.

10. The mixture of any of the preceding claims, wherein the ginger root extract is characterized by a composition comprising, on a dry weight basis relative to the total weight of the ginger root extract:6-gingerol in an amount ranging from 0.5 to 8.0% by weight, preferably 1 .0 to 5.5% by weight, more preferably 1 .5 to 4.0% by weight, preferably with the proviso that the total amount of gingerols and shogaols ranges from 0.5 to 10% by weight, preferably 1 .0 to 8.0% by weight, more preferably 2.0 to 5.0% by weight.11 . The mixture of any of the preceding claims, wherein the ginger root extract is a supercritical CO2 extract from dried ginger root and / or wherein the stinging nettle extract is a press juice extract from fresh plant material.

12. A method for producing a mixture formed from extracts of stinging nettle and ginger root as defined by any one of claims 1 to 11 , comprising: a) providing extracts of stinging nettle and ginger root and, optionally, one or more auxiliary agents selected from the group of maltodextrin, Arabic gum, and combinations thereof;b) mixing the extracts and, optionally, the one or more auxiliary agents, in appropriate relative proportions; and c) obtaining the mixture.

13. A method for soothing skin, reducing skin redness or inhibiting cytokine production in keratinocyte cells, comprising: providing a mixture as defined in any of claims 1 to 11 ; and topically applying the mixture on a region of skin, where the skin is to be soothed, wherein the skin redness is to be reduced, or where the cytokine production in keratinocyte cells is to be inhibited.

14. A mixture as defined in any of claims 1 to 11 for use in a method for treating a skin condition associated with cytokine overproduction in keratinocyte cells by topically applying the mixture on regions of skin showing signs of cytokine overproduction.

15. Use of a stinging nettle extract for stabilizing a ginger root extract.
